Balancing repair and tolerance of DNA damage caused by alkylating agents
Alkylating agents constitute a major class of frontline chemotherapeutic drugs that inflict cytotoxic DNA damage as their main mode of action, in addition to collateral mutagenic damage. Numerous cellular pathways, including direct DNA damage reversal, base excision repair (BER) and mismatch repair (MMR), respond to alkylation damage to defend against alkylation-induced cell death or mutation. However, maintaining a proper balance of activity both within and between these pathways is crucial for a favourable response of an organism to alkylating agents. Furthermore, the response of an individual to alkylating agents can vary considerably from tissue to tissue and from person to person, pointing to genetic and epigenetic mechanisms that modulate alkylating agent toxicity
Determination of Preferred pH for Root-knot Nematode Aggregation Using Pluronic F-127 Gel
Root-knot nematodes (Meloidogyne spp.) are obligate endoparasites of a wide range of plant species. The infective stage is attracted strongly to and enters host roots at the zone of elongation, but the compounds responsible for this attraction have not been identified. We developed a simple assay to investigate nematode response to chemical gradients that uses Pluronic F-127, a synthetic block copolymer that, as a 23% aqueous solution, forms a liquid at low temperature and a gel at room temperature. Test chemicals are put into a modified pipette tip, or ‘chemical dispenser,’ and dispensers are inserted into the gel in which nematodes have been dispersed. Meloidogyne hapla is attracted to pH gradients formed by acetic acid and several other Brønsted acids and aggregates between pH 4.5 and 5.4. While this pH range was attractive to all tested root-knot nematode strains and species, the level of aggregation depended on the species/strain assessed. For actively growing roots, the pH at the root surface is most acidic at the zone of elongation. This observation is consistent with the idea that low pH is an attractant for nematodes. Root-knot nematodes have been reported to be attracted to carbon dioxide, but our experiments suggest that the observed attraction may be due to acidification of solutions by dissolved CO2 rather than to CO2 itself. These results suggest that Pluronic F-127 gel will be broadly applicable for examining responses of a range of organisms to chemical gradients or to each other
Ultralong C100 Mycolic Acids Support the Assignment of Segniliparus as a New Bacterial Genus
Mycolic acid-producing bacteria isolated from the respiratory tract of human and non-human mammals were recently assigned as a distinct genus, Segniliparus, because they diverge from rhodococci and mycobacteria in genetic and chemical features. Using high accuracy mass spectrometry, we determined the chemical composition of 65 homologous mycolic acids in two Segniliparus species and separately analyzed the three subclasses to measure relative chain length, number and stereochemistry of unsaturations and cyclopropyl groups within each class. Whereas mycobacterial mycolate subclasses are distinguished from one another by R groups on the meromycolate chain, Segniliparus species synthesize solely non-oxygenated α-mycolates with high levels of cis unsaturation. Unexpectedly Segniliparus α-mycolates diverge into three subclasses based on large differences in carbon chain length with one bacterial culture producing mycolates that range from C58 to C100. Both the overall chain length (C100) and the chain length diversity (C42) are larger than previously seen for mycolic acid-producing organisms and provide direct chemical evidence for assignment of Segniliparus as a distinct genus. Yet, electron microscopy shows that the long and diverse mycolates pack into a typical appearing membrane. (How) do medical students regulate their emotions?
BACKGROUND: Medical training can be a challenging and emotionally intense period for medical students. However the emotions experienced by medical students in the face of challenging situations and the emotion regulation strategies they use remains relatively unexplored. The aim of the present study was to explore the emotions elicited by memorable incidents reported by medical students and the associated emotion regulation strategies. METHODS: Peer interviewing was used to collect medical students’ memorable incidents. Medical students at both preclinical and clinical stage of medical school were eligible to participate. In total 104 medical students provided memorable incidents. Only 54 narratives included references to emotions and emotion regulation and thus were further analyzed. RESULTS: The narratives of 47 clinical and 7 preclinical students were further analyzed for their references to emotions and emotion regulation strategies. Forty seven out of 54 incidents described a negative incident associated with negative emotions. The most frequently mentioned emotion was shock and surprise followed by feelings of embarrassment, sadness, anger and tension or anxiety. The most frequent reaction was inaction often associated with emotion regulation strategies such as distraction, focusing on a task, suppression of emotions and reappraisal. When students witnessed mistreatment or disrespect exhibited towards patients, the regulation strategy used involved focusing and comforting the patient. CONCLUSIONS: The present study sheds light on the strategies medical students use to deal with intense negative emotions. The vast majority reported inaction in the face of a challenging situation and the use of more subtle strategies to deal with the emotional impact of the incident. Ecological Guild Evolution and the Discovery of the World's Smallest Vertebrate
Living vertebrates vary drastically in body size, yet few taxa reach the extremely minute size of some frogs and teleost fish. Here we describe two new species of diminutive terrestrial frogs from the megadiverse hotspot island of New Guinea, one of which represents the smallest known vertebrate species, attaining an average body size of only 7.7 mm. Both new species are members of the recently described genus Paedophryne, the four species of which are all among the ten smallest known frog species, making Paedophryne the most diminutive genus of anurans. This discovery highlights intriguing ecological similarities among the numerous independent origins of diminutive anurans, suggesting that minute frogs are not mere oddities, but represent a previously unrecognized ecological guild
A Survey of Bayesian Statistical Approaches for Big Data
The modern era is characterised as an era of information or Big Data. This
has motivated a huge literature on new methods for extracting information and
insights from these data. A natural question is how these approaches differ
from those that were available prior to the advent of Big Data. We present a
review of published studies that present Bayesian statistical approaches
specifically for Big Data and discuss the reported and perceived benefits of
these approaches. We conclude by addressing the question of whether focusing
only on improving computational algorithms and infrastructure will be enough to
face the challenges of Big Data
Community Characteristics of Sympatric Freshwater Turtles from Savannah Waterbodies in Ghana
Despite increasing pressures on freshwater resources worldwide, and the threatened status of most freshwater turtles, there is still limited knowledge of habitat use and niche partitioning in Afrotropical freshwater turtle communities. In this study, we describe habitat associations, community diversity, and temporal patterns of occurrence of freshwater turtle species in the Dahomey Gap ecoregion of Ghana (West Africa). We gathered data from 13 sites in central Ghana and along the Sene Arm of Lake Volta in the Digya National Park (Bono East Region). We employed opportunistic short-term surveys (at seven sites) together with longer-term (six-months duration) standardized evaluations of turtle presence and numbers in different habitats (at six sites). Overall, a total of 210 turtle individuals of four species (Trionyx triunguis, Cyclanorbis senegalensis, Pelomedusa sp. and Pelusios castaneus) were recorded; precise capture sites and habitat type were recorded for 139 individuals, but the 71 individuals observed in marketplaces were not considered in our analyses. At a local scale, we observed three sympatric species in various study sites. In each of these sites, the dominant species was either C. senegalensis or Pelomedusa sp., with the latter species being more abundant in temporary waterbodies and C. senegalensis more numerous in permanent ones. A Multiple Correspondence Analysis suggested that, in permanent waterbodies all species were associated with similar physical habitat variables. In a Canonical Correspondence Analysis, we showed that the density of herbaceous emergent vegetation was more important for P. castaneus than for C. senegalensis. Comparisons of diversity metrics between our study sites and previous studies revealed that turtle community composition was similar across savannah sites
Supernova remnants: the X-ray perspective
Supernova remnants are beautiful astronomical objects that are also of high
scientific interest, because they provide insights into supernova explosion
mechanisms, and because they are the likely sources of Galactic cosmic rays.
X-ray observations are an important means to study these objects.And in
particular the advances made in X-ray imaging spectroscopy over the last two
decades has greatly increased our knowledge about supernova remnants. It has
made it possible to map the products of fresh nucleosynthesis, and resulted in
the identification of regions near shock fronts that emit X-ray synchrotron
radiation.
In this text all the relevant aspects of X-ray emission from supernova
remnants are reviewed and put into the context of supernova explosion
properties and the physics and evolution of supernova remnants. The first half
of this review has a more tutorial style and discusses the basics of supernova
remnant physics and thermal and non-thermal X-ray emission. The second half
offers a review of the recent advances.The topics addressed there are core
collapse and thermonuclear supernova remnants, SN 1987A, mature supernova
remnants, mixed-morphology remnants, including a discussion of the recent
finding of overionization in some of them, and finally X-ray synchrotron

Trilingual education in Hong Kong primary schools: an overview
Hong Kong is linguistically complex and diverse with three principal languages: Cantonese, English and Putonghua. A substantial debate on the language policies governing the three principal languages has continued for more than two decades among policy-makers and educators. The political transition in 1997 has greatly affected Hong Kong society, including language education. Since then, the HKSAR government has made a series of language policy reforms trying to create a reasonable balance among the three languages in Hong Kong. The policies of ‘biliteracy and trilingualism’ and ‘mother-tongue teaching’ are two of the most significant in terms of controversy and impact. They are now guiding the curriculum design in Hong Kong language education. The goal of the former policy is to train Hong Kong people to be truly biliterate (written English and Chinese) and trilingual (spoken English, Cantonese and Putonghua). However, Hong Kong primary schools presently do not have an agreed method for the implementation of trilingual education. After a comprehensive historical review of the development of language education in Hong Kong schools, this study aims to find out how the ‘biliterate’ and ‘trilingual’ language policy is currently implemented in Hong Kong primary schools. 155 Hong Kong primary schools participated in a questionnaire survey on how trilingual education is implemented in the schools. The findings suggest that the implementation of trilingual education varied significantly from school to school, and the effectiveness of the trilingual education models varied as well. It is hoped that the findings will help us to gain a better understanding of trilingual education in Hong Kong, and the study could lead to some insightful and theoretical contributions to multilingual education in general.Full Tex
