Calcium-independent contraction in lysed cell models of teleost retinal cones: activation by unregulated myosin light chain kinase or high magnesium and loss of cAMP inhibition.
The retinal cones of teleost fish contract at dawn and elongate at dusk. We have previously reported that we can selectively induce detergent-lysed models of cones to undergo either reactivated contraction or reactivated elongation, with rates and morphology comparable to those observed in vivo. Reactivated contraction is ATP dependent, activated by Ca2+, and inhibited by cAMP. In addition, reactivated cone contraction exhibits several properties that suggest that myosin phosphorylation plays a role in mediating Ca2+-activation (Porrello, K., and B. Burnside, 1984, J. Cell Biol., 98:2230-2238). We report here that lysed cone models can be induced to contract in the absence of Ca2+ by incubation with trypsin-digested, unregulated myosin light chain kinase (MLCK) obtained from smooth muscle. This observation provides further evidence that MLCK plays a role in regulating cone contraction. We also report here that lysed cone models can be induced to contract in the absence of Ca2+ by incubation with high concentrations of MgCl2 (10-20 mM). Mg2+-induced reactivated contraction is supported by inosine triphosphate (ITP) just as well as by ATP. Because ITP will not serve as a substrate for MLCK, this finding suggests that Mg2+-activation of contraction does not require myosin phosphorylation. Although Ca2+-induced contraction is completely blocked by cAMP at concentrations less than 10 microM, cAMP has no effect on cone contraction activated by unregulated MLCK or by high Mg2+ in the absence of Ca2+. Because trypsin digestion of MLCK cleaves off not only the Ca2+/calmodulin-binding site but also the site phosphorylated by cAMP-dependent protein kinase, and because Mg2+ activation of cone contraction circumvents MLCK action altogether, both these observations would be expected if cAMP inhibits reactivated cone contraction by catalyzing the phosphorylation of MLCK and thus reducing its affinity for Ca2+, as has been described for smooth muscle. Together our results suggest that in lysed cone models, myosin phosphorylation is sufficient for activating cone contraction, even in the absence of other Ca2+-mediated events, that cAMP inhibition of contraction is mediated by cAMP-dependent phosphorylation of MLCK, and that 10-20 mM Mg2+ can activate actin-myosin interaction to produce contraction in the absence of myosin phosphorylation
A very wide frequency band pulsed/IF radar system
A pulsed/IF radar for compact range radar cross section measurements has been developed which converts RF returns to a fixed IF, so that amplification and grating may be performed at one frequency. This permits the use of components which have optimal performance at this frequency which results in a corresponding improvement in performance. Sensitivity and dynamic range are calculated for this system and compared with our old radar, and the effect of pulse width on clutter level is also studied. Sensitivity and accuracy tests are included to verify the performance of the radar
Regulation of reactivated elongation in lysed cell models of teleost retinal cones by cAMP and calcium.
Teleost retinal cones elongate in the dark and contract in the light. In isolated retinas of the green sunfish Lepomis cyanellus, cone myoids undergo microtubule-dependent elongation from 5 to 45 micron. We have previously shown that cone contraction can be reactivated in motile models of cones lysed with Brij-58. Reactivated contraction is both actin and ATP dependent, activated by calcium, and inhibited by cAMP. We report here that we have obtained reactivated cone elongation in lysed models prepared by the same procedures. Reactivated elongation is ATP dependent, activated by cAMP, and inhibited by calcium. The rate of reactivated elongation is proportional to the cAMP concentration between 10 microM and 0.5 mM, but is constant between 10 microM and 1.0 mM Mg-ATP. No elongation occurs if cAMP or Mg-ATP concentration is less than or equal to 5 microM. Mg-ATP is required for both cAMP-dependent and cAMP-independent processes, suggesting that Mg-ATP is required both for a regulatory process entailing cAMP-dependent phosphorylation and for a force-producing process. Free calcium concentrations greater than or equal to 10(-7) reduce the elongation rate by 78% or more, completely inhibiting elongation at 10(-5) M. This inhibition is not due to competition from calcium-activated contraction. Cytochalasin D blocks reactivated contraction, but does not abolish calcium inhibition of reactivated elongation. Thus calcium directly affects the elongation mechanism. Calcium inhibition is calmodulin dependent. The calmodulin inhibitor trifluoperazine abolishes calcium inhibition of elongation. Furthermore, calcium blocks elongation only if present during the lysis step; subsequent calcium addition has no effect. However, if calcium plus exogenous calmodulin are subsequently added, elongation is again inhibited. Thus calcium inhibition appears to require a soluble calmodulin which is lost shortly after lysis
A very efficient RCS data compression and reconstruction technique, volume 4
A very efficient compression and reconstruction scheme for RCS measurement data was developed. The compression is done by isolating the scattering mechanisms on the target and recording their individual responses in the frequency and azimuth scans, respectively. The reconstruction, which is an inverse process of the compression, is granted by the sampling theorem. Two sets of data, the corner reflectors and the F-117 fighter model, were processed and the results were shown to be convincing. The compression ratio can be as large as several hundred, depending on the target's geometry and scattering characteristics

Computational Fluid Dynamics in Small Airway Models of the Human Lung
The promise of gene replacement therapy for cystic fibrosis, the administration of drugs via inhalation therapy, and die deposition location of man-made airborne particulates all involve a more complete understanding of the fluid dynamics in the human lung. Flow in the larger airways may be measured through life-sized models directly, but the airways in the peripheral lung are too small and the flows are too complex to be studied in this manner. Computational models can be developed which will accurately represent both the geometric nature of the central airways and the fluid dynamics with in them. Two-dimensional and three-dimensional models of central lung airway bifurcations were developed based on morphometry. These models were used as the spatial basis upon which the differential equations that describe incompressible flow, the Navier Stokes equations, are solved. Flow solutions have been computed at Reynolds numbers from 1000 down to 100. Solutions for single and double bifurcations agree with the experimental data for flow in a branching tube. These studies are being extended to multiple bifurcations in three dimensions
A Model for the Analysis of Caries Occurrence in Primary Molar Tooth Surfaces
Recently methods of caries quantification in the primary dentition have moved away from summary ‘whole mouth’ measures at the individual level to methods based on generalised linear modelling (GLM) approaches or survival analysis approaches. However, GLM approaches based on logistic transformation fail to take into account the time-dependent process of tooth/surface survival to caries. There may also be practical difficulties associated with casting parametric survival-based approaches in a complex multilevel hierarchy and the selection of an optimal survival distribution, while non-parametric survival methods are not generally suitable for the assessment of supplementary information recorded on study participants. In the current investigation, a hybrid semi-parametric approach comprising elements of survival-based and GLM methodologies suitable for modelling of caries occurrence within fixed time periods is assessed, using an illustrative multilevel data set of caries occurrence in primary molars from a cohort study, with clustering of data assumed to occur at surface and tooth levels. Inferences of parameter significance were found to be consistent with previous parametric survival-based analyses of the same data set, with gender, socio-economic status, fluoridation status, tooth location, surface type and fluoridation status-surface type interaction significantly associated with caries occurrence. The appropriateness of the hierarchical structure facilitated by the hybrid approach was also confirmed. Hence the hybrid approach is proposed as a more appropriate alternative to primary caries modelling than non-parametric survival methods or other GLM-based models, and as a practical alternative to more rigorous survival-based methods unlikely to be fully accessible to most researchers
On uniformization of Burnside's curve
Main objects of uniformization of the curve are studied: its

parameters. As a result we obtain the first examples of solvable Fuchsian
equations on torus and exhibit number-theoretic integer -series for
uniformizing functions, relevant modular forms, and analytic series for
holomorphic Abelian integrals. A conjecture of Whittaker for hyperelliptic
curves and its hypergeometric reducibility are discussed. We also consider the

Does shear wave ultrasound independently predict axillary lymph node metastasis in women with invasive breast cancer?
Shear wave elastography (SWE) shows promise as an adjunct to greyscale ultrasound examination in assessing breast masses. In breast cancer, higher lesion stiffness on SWE has been shown to be associated with features of poor prognosis. The purpose of this study was to assess whether lesion stiffness at SWE is an independent predictor of lymph node involvement. Patients with invasive breast cancer treated by primary surgery, who had undergone SWE examination were eligible. Data were retrospectively analysed from 396 consecutive patients. The mean stiffness values were obtained using the Aixplorer(®) ultrasound machine from SuperSonic Imagine Ltd. Measurements were taken from a region of interest positioned over the stiffest part of the abnormality. The average of the mean stiffness value obtained from each of two orthogonal image planes was used for analysis. Associations between lymph node involvement and mean lesion stiffness, invasive cancer size, histologic grade, tumour type, ER expression, HER-2 status and vascular invasion were assessed using univariate and multivariate logistic regression. At univariate analysis, invasive size, histologic grade, HER-2 status, vascular invasion, tumour type and mean stiffness were significantly associated with nodal involvement. Nodal involvement rates ranged from 7 % for tumours with mean stiffness <50 kPa to 41 % for tumours with a mean stiffness of >150 kPa. At multivariate analysis, invasive size, tumour type, vascular invasion, and mean stiffness maintained independent significance. Mean stiffness at SWE is an independent predictor of lymph node metastasis and thus can confer prognostic information additional to that provided by conventional preoperative tumour assessment and staging
On the trace of the antipode and higher indicators
We introduce two kinds of gauge invariants for any finite-dimensional Hopf
algebra H. When H is semisimple over C, these invariants are respectively, the
trace of the map induced by the antipode on the endomorphism ring of a
self-dual simple module, and the higher Frobenius-Schur indicators of the
regular representation. We further study the values of these higher indicators
in the context of complex semisimple quasi-Hopf algebras H. We prove that these
indicators are non-negative provided the module category over H is modular, and
that for a prime p, the p-th indicator is equal to 1 if, and only if, p is a
factor of dim H. As an application, we show the existence of a non-trivial
self-dual simple H-module with bounded dimension which is determined by the
