    Genetic Analysis of Restriction Fragment Length Polymorphisms in Vitis

    The parents and progeny from two crosses (Cayuga White x Aurore and NY62.136.2 x Yates) were examined for the presence of DNA restriction fragment length polymorphisms (RFLPs). Seventeen independent DNA sequences were used in the analysis, 15 obtained from a grape Pstl genomic library and two heterologous probes obtained from other laboratories. Most of the low copy cloned sequences hybridized to more than two restriction fragments, possibly reflecting the polyploid nature of the Vitis genome. Nine of the probes detected RFLPs between parents. Analysis of the progenies (F1generation) revealed segregation for nine distinct polymorphisms generated by seven of the probes.Thus, a relatively high level of polymorphism among parents, as well as heterozygosity within each parent, was evident. Most RFLPs gave segregation ratios close to the 1: 1 ratio predicted for a locus heterozygous in one parent. However four differences between parental phenotypes did not segregated in the progeny, and in three instances fragments present in both parents segregated in the progeny. These peculiar results may be explained by accounting for heterozygosity or homozygosity, respectively, for the DNA segment that generates the polymorphism. We conclude that RFLP studies can be performed on the first filial generation in woody perennials such as Vitis that have a relatively high level of heterozygosity in the genom

    Towards environmentally sustainable human behaviour: targeting non-conscious and conscious processes for effective and acceptable policies.

    Meeting climate change targets to limit global warming to 2°C requires rapid and large reductions in demand for products that most contribute to greenhouse gas (GHG) emissions. These include production of bulk materials (e.g. steel and cement), energy supply (e.g. fossil fuels) and animal source foods (particularly ruminants and their products). Effective strategies to meet these targets require transformative changes in supply as well as demand, involving changes in economic, political and legal systems at local, national and international levels, building on evidence from many disciplines. This paper outlines contributions from behavioural science in reducing demand. Grounded in dual-process models of human behaviour (involving non-conscious and conscious processes) this paper considers first why interventions aimed at changing population values towards the environment are usually insufficient or unnecessary for reducing demand although they may be important in increasing public acceptability of policies that could reduce demand. It then outlines two sets of evidence from behavioural science towards effective systems-based strategies, to identify interventions likely to be effective at: (i) reducing demand for products that contribute most to GHG emissions, mainly targeting non-conscious processes and (ii) increasing public acceptability for policy changes to enable these interventions, targeting conscious processes.This article is part of the themed issue 'Material demand reduction'

    The Q2Q^2-dependence of the generalised Gerasimov-Drell-Hearn integral for the deuteron, proton and neutron

    The Gerasimov-Drell-Hearn (GDH) sum rule connects the anomalous contribution to the magnetic moment of the target nucleus with an energy-weighted integral of the difference of the helicity-dependent photoabsorption cross sections. The data collected by HERMES with a deuterium target are presented together with a re-analysis of previous measurements on the proton. This provides a measurement of the generalised GDH integral covering simultaneously the nucleon-resonance and the deep inelastic scattering regions. The contribution of the nucleon-resonance region is seen to decrease rapidly with increasing Q2Q^2. The DIS contribution is sizeable over the full measured range, even down to the lowest measured Q2Q^2. As expected, at higher Q2Q^2 the data are found to be in agreement with previous measurements of the first moment of g1g_1. From data on the deuteron and proton, the GDH integral for the neutron has been derived and the proton--neutron difference evaluated. This difference is found to satisfy the fundamental Bjorken sum rule at Q2=5Q^2 = 5 GeV2^2.Comment: 12 pages, 10 figure

    Suicides in public places: findings from one English county

    Little is known about where suicides take place. We collected data from coroners’ files on all suicides and undetermined deaths in one large English county from 2000 to 2004. The data show that >30% of suicides occurred in public places. A quarter of these involved jumping from a height and nearly a quarter involved car exhaust poisoning. Several sites were associated with multiple methods of suicide. Identifying and managing high-risk locations should be an important part of an overall suicide prevention strategy and is best tackled at local level

    Soil seed banks of remnant and degraded Swartland Shale Renosterveld

    Questions: What are the characteristics of soil seed banks in highly endangered renosterveld vegetation and adjacent degraded sites? What is the contribution of the soil seed bank to restoring renosterveld vegetation after degradation through agriculture or afforestation? Location: Tygerberg Nature Reserve – one of the largest remaining fragments of Swartland Shale Renosterveld, Cape Floristic Region, South Africa. Methods: Assessment of vegetation, soil parameters and soil seed banks in three renosterveld sites, two adjacent abandoned fields, one pine plantation and one pine plantation clear-cut site. Smoke primer (i.e. fire surrogate) was applied to soil seed bank samples to evaluate fire as a possible management and restoration tool. Results: Abandoned agricultural fields adjacent to renosterveld remnants are characterized by alien grass cover, nutrient enrichment of the soil and depletion of the indigenous soil seed bank. In contrast, pine plantations show less alien species infestation, soil nutrient alteration and have a viable soil seed bank as well as re-development of indigenous renosterveld vegetation after clearance. Seedling recruitment was not significantly influenced by application of a smoke primer compared to the magnitude of fire response in fynbos. Conclusion: Abandoned agricultural fields (previously renosterveld) at Tygerberg have a very low restoration potential. In contrast, pine plantation sites should be given priority in restoration, because soil chemistry is less significantly altered and a viable indigenous soil seed bank is still present, which can be successfully activated through clearance management

    The inherent room temperature mending and self-healing properties of saloplastic PAA/PAH CoPECs are studied. After ultracentrifugation of PAA/PAH polyelectrolyte complexes, tough, elastic materials are obtained that undergo self-healing facilitated by salt.     Trajectories of objectively measured physical activity and childhood overweight: longitudinal analysis of the IDEFICS/I.Family cohort

    Background: Since only few longitudinal studies with appropriate study designs investigated the relationship between objectively measured physical activity (PA) and overweight, the degree PA can prevent excess weight gain in children, remains unclear. Moreover, evidence is limited on how childhood overweight determines PA during childhood. Therefore, we analyzed longitudinal trajectories of objectively measured PA and their bi-directional association with weight trajectories of children at 2- and 6-year follow-ups. Methods: Longitudinal data of three subsequent measurements from the IDEFICS/I.Family cohort study were used to analyze the bi-directional association between moderate-to-vigorous PA (MVPA) and weight status by means of multilevel regression models. Analyses comprised 3393 (2-year follow-up) and 1899 (6-year follow-up) children aged 2–15.9 years from eight European countries with valid accelerometer data and body mass index (BMI) measurements. For categorized analyses, children’s weight status was categorized as normal weight or overweight (cutoff: 90th percentile of BMI) and children’s PA as (in-) sufficiently active (cutoffs: 30, 45 and 60 min of MVPA per day). Results: Children engaging in at least 60 min MVPA daily at baseline and follow-ups had a lower odds of becoming overweight (odds ratio [OR] at 2-year follow-up: 0.546, 95% CI: 0.378, 0.789 and 6-year follow-up: 0.393, 95% CI: 0.242, 0.638), compared to less active children. Similar associations were found for 45 min MVPA daily. On the other side, children who became overweight had the lowest odds to achieve 45 or 60 min MVPA daily (ORs: 0.459 to 0.634), compared to normal weight children. Conclusions: Bi-directional associations between MVPA and weight status were observed. In summary, at least 60 min MVPA are still recommended for the prevention of childhood overweight. To prevent excess weight gain, 45 min MVPA per day also showed preventive effects

    Haplotyping the Vitis collinear core genome with rhAmpSeq improves marker transferability in a diverse genus

    Transferable DNA markers are essential for breeding and genetics. Grapevine (Vitis) breeders utilize disease resistance alleles from congeneric species ~20 million years divergent, but existing Vitis marker platforms have cross-species transfer rates as low as 2%. Here, we apply a marker strategy targeting the inferred Vitis core genome. Incorporating seven linked-read de novo assemblies and three existing assemblies, the Vitis collinear core genome is estimated to converge at 39.8 Mb (8.67% of the genome). Adding shotgun genome sequences from 40 accessions enables identification of conserved core PCR primer binding sites flanking polymorphic haplotypes with high information content. From these target regions, we develop 2,000 rhAmpSeq markers as a PCR multiplex and validate the panel in four biparental populations spanning the diversity of the Vitis genus, showing transferability increases to 91.9%. This marker development strategy should be widely applicable for genetic studies in many taxa, particularly those ~20 million years divergent

    Primary treatment of acromegaly with high-dose lanreotide: a case series

    <p>Abstract</p> <p>Introduction</p> <p>The first-line treatment for acromegaly is transsphenoidal surgery. In approximately 50% of patients, however, a cure is not possible with surgery and alternatives are needed. Somatostatin analog therapy is the recommended first-line treatment in patients with such cases. Here we provide the first report of a high-dose lanreotide primary therapy in patients with acromegaly.</p> <p>Case presentation</p> <p>Six patients who were not suitable for surgery were given 60 mg of lanreotide (Autogel<sup>®</sup>) every four weeks. All patients were German nationals and Caucasian.</p> <p>When the response of our patients was unsatisfactory, the dose was increased sequentially to 90 mg every four weeks, 120 mg every four weeks, 120 mg every three weeks and 180 mg every three weeks. Treatment duration was 12 to 24 months. In all cases, the lanreotide dose was 120 mg every 4 weeks or higher. In five of our patients, growth hormone (GH) levels were successfully reduced (in three patients GH <2.5 ng/ml was achieved). Insulin-like growth factor 1 levels were normalized in three patients and decreased in two patients. One patient failed to show a biochemical response to lanreotide therapy or pegvisomant therapy.</p> <p>Tumor shrinkage or degeneration was observed in the five responding patients. No drug-related adverse events were noted.</p> <p>Conclusions</p> <p>These results suggest that lanreotide at high doses of 120 mg every four weeks or more is an effective first-line therapy for patients with acromegaly that surgery alone cannot treat.</p
