    Efficiency of transient transformation in tobacco protoplasts is independent of plasmid amount

    An optimized protocol for the transient transformation of tobacco protoplasts mediated by polyethylene\u2013glycol (PEG) is here described. As expected, the quantitative b\u2013glucuronidase (Gus) activity driven by pCaMVGus was dependent on the amount of plasmid used. Nevertheless, we demonstrate by an immunodetection method that transformation efficiency did not depend on the amount of plasmid used but on the limitation imposed by cell competence. In fact, we obtained the same percentage of transformed cells (about 60%) using a wide range of plasmid concentrations (0.1\u201310 mg per test). Finally, we show that, when we used two plasmid types in a mixture at a concentration ranging from 0.1 to 10 mg for each, all transformed cells expressed proteins encoded by both plasmids. Transient expression and co-transformation experiments are routinely used methods and, probably, the major results from this work were assumed by many researchers in this field, but our data experimentally support this assumption

    A candidate protostellar object in the L1457 / MBM12 cloud

    The association of young T Tauri stars, MBM12A, indicates that L1457 was forming stars not too long ago. With our study we want to find out whether or not there are still signs for ongoing star formation in that cloud. Using the Max-Planck-Millimeter-Bolometer MAMBO at the IRAM 30m telescope we obtained a map of about 8' by 8' centered on L1457 in the dust continuum emission at 230 GHz. Towards the most intense regions in our bolometer map we obtained spectra at high angular resolution in the CS (2-1) and the N2H+(1-0) lines using the IRAM 30m telescope. We find that the cold dust in L1457 is concentrated in several small cores with high H2 column densities and solar masses. The density profiles of the cores are inconsistent with a sphere with constant density. These cores are closer to virial equilibrium than the cloud as a whole. Data from the VLA and Spitzer archives reveal two point sources in the direction of one dust core. One of the sources is probably a distant quasar, whereas the other source is projected right on a local maximum of our dust map and shows characteristics of a protostellar object.Comment: 4 pages, 4 figures, accepted by Astronomy & Astrophysic

    Thermal Expansion of the Heavy-fermion Superconductor PuCoGa5_{5}

    We have performed high-resolution powder x-ray diffraction measurements on a sample of 242^{242}PuCoGa5_{5}, the heavy-fermion superconductor with the highest critical temperature TcT_{c} = 18.7 K. The results show that the tetragonal symmetry of its crystallographic lattice is preserved down to 2 K. Marginal evidence is obtained for an anomalous behaviour below TcT_{c} of the aa and cc lattice parameters. The observed thermal expansion is isotropic down to 150 K, and becomes anisotropic for lower temperatures. This gives a c/ac/a ratio that decreases with increasing temperature to become almost constant above \sim150 K. The volume thermal expansion coefficient αV\alpha_{V} has a jump at TcT_{c}, a factor \sim20 larger than the change predicted by the Ehrenfest relation for a second order phase transition. The volume expansion deviates from the curve expected for the conventional anharmonic behaviour described by a simple Gr\"{u}neisen-Einstein model. The observed differences are about ten times larger than the statistical error bars but are too small to be taken as an indication for the proximity of the system to a valence instability that is avoided by the superconducting state.Comment: 6 pages, 4 figure

    Extinction Curves, Distances, and Clumpiness of Diffuse Interstellar Dust Clouds

    We present CCD photometry in UBVRI of several thousand Galactic field stars in four large (>1 degree^2) regions centered on diffuse interstellar dust clouds, commonly referred to as ``cirrus'' clouds (with optical depth A_V less than unity). Our goal in studying these stars is to investigate the properties of the cirrus clouds. A comparison of the observed stellar surface density between on-cloud and off-cloud regions as a function of apparent magnitude in each of the five bands effectively yields a measure of the extinction through each cloud. For two of the cirrus clouds, this method is used to derive UBVRI star counts-based extinction curves, and U-band counts are used to place constraints on the cloud distance. The color distribution of stars and their location in (U-B, B-V) and (B-V, V-I) color-color space are analyzed in order to determine the amount of selective extinction (reddening) caused by the cirrus. The color excesses, A_lambda-A_V, derived from stellar color histogram offsets for the four clouds, are better fit by a reddening law that rises steeply towards short wavelengths [R_V==A_V/E(B-V)<=2] than by the standard law (R_V=3.1). This may be indicative of a higher-than-average abundance of small dust grains relative to larger grains in diffuse cirrus clouds. The shape of the counts-based effective extinction curve and a comparison of different estimates of the dust optical depth (extinction optical depth derived from background star counts/colors; emission optical depth derived from far infrared measurements), are used to measure the degree of clumpiness in clouds. The set of techniques explored in this paper can be readily adapted to the Sloan Digital Sky Survey data set in order to carry out a systematic, large-scale study of cirrus clouds.Comment: 22 pages, 13 figures (postscript, gif, jpg).
