The src-family kinase feline gardner-rasheed is implicated in chronic lymphocytic leukemia cellular metabolic regulation

Alterations intrinsic to Chronic Lymphocytic Leukemia (CLL) cells affect many signaling pathways, including those relating to proliferation and evasion of apoptosis. Interactions with the microenvironment further alter survival, proliferative, and metabolic signaling of CLL cells. Feline Gardner-Rasheed (Fgr) is a Src-family kinase implicated in B cell receptor signaling and metabolism. Its expression is variable and its function is unknown in CLL cells. The ability of CLL cells to regulate their metabolism in response to energy demand is key to their ability to sustain proliferation and survive cellular stress. We hypothesize that Fgr expression correlates with clinical parameters, and contributes to signaling that enhances cell survival and metabolic adaptation to energy demand. In a transiently-transfected BJAB cell line, we found that Fgr over expression increased glycolysis and mitochondrial respiratory capacity in BJAB cells. This correlated with the ability of Fgr to activate complex II in the mitochondria. Furthermore, Fgr expression provided cell survival signaling following treatment with metabolic toxins. Finally, we found that Fgr expression correlates positively with favourable prognosis markers in CLL cells. This suggests that Fgr regulates metabolism and cell survival in CLL cells.October 201

The impact of prescription pain reliever misuse and heroin use on morbidity and mortality by level of urbanicity: 2002-2014

Non-medical use of prescription pain relievers (PPRs), heroin, and more recently fentanyl, continue to have major public health consequences in the United States. This article analyzes trends in PPR and heroin use, emergency department and hospital stays, substance use treatment services, and mortality to assess the relative impact of the opioid crisis on rural versus more urbanized counties in the United States. Our findings suggest that while more urbanized counties have had greater increases in opioid use, rural and less urbanized counties tended to be more negatively impacted than larger and non-rural counties. Disparities in service availability highlight the need for a serious discussion on how resources are allocated in counties that have lower tax bases. Based on these results, we conclude that rural and less urbanized counties can benefit from real increases in resources for substance use prevention and treatment services, including the expansion of prescribers trained to screen and treat opioid use. Understanding the unique challenges of rural and less urbanized counties may help decrease the disparity in consequences found in this study

Desde marzo de 2020 hasta la actualidad como país y de manera global, hemos vivido un fenómeno particular que es la pandemia por el Covid -19. Todas las áreas del sector Salud han tenido que modificar su trabajo, dada las condiciones y protocolos de la autoridad sanitaria que exige esta situación. El control de niño sano, no ha quedado exento de lo antes mencionado. Dado lo anterior, es de interés profundizar en la vivencia de los profesionales de enfermería que en pandemia realizan estos controles, para generar conocimiento comprensivo y con ello generar próximas estrategias para abordar esta labor de gran importancia en el desarrollo de los niños

Identification of novel risk loci for restless legs syndrome in genome-wide association studies in individuals of European ancestry : a meta-analysis

Background Restless legs syndrome is a prevalent chronic neurological disorder with potentially severe mental and physical health consequences. Clearer understanding of the underlying pathophysiology is needed to improve treatment options. We did a meta-analysis of genome-wide association studies (GWASs) to identify potential molecular targets. Methods In the discovery stage, we combined three GWAS datasets (EU-RLS GENE, INTERVAL, and 23andMe) with diagnosis data collected from 2003 to 2017, in face-to-face interviews or via questionnaires, and involving 15126 cases and 95 725 controls of European ancestry. We identified common variants by fixed-effect inverse-variance meta-analysis. Significant genome-wide signals (p Findings We identified and replicated 13 new risk loci for restless legs syndrome and confirmed the previously identified six risk loci. MEIS1 was confirmed as the strongest genetic risk factor for restless legs syndrome (odds ratio 1.92, 95% CI 1 85-1.99). Gene prioritisation, enrichment, and genetic correlation analyses showed that identified pathways were related to neurodevelopment and highlighted genes linked to axon guidance (associated with SEMA6D), synapse formation (NTNG1), and neuronal specification (HOXB cluster family and MYT1). Interpretation Identification of new candidate genes and associated pathways will inform future functional research. Advances in understanding of the molecular mechanisms that underlie restless legs syndrome could lead to new treatment options.
