    Annotation Graphs and Servers and Multi-Modal Resources: Infrastructure for Interdisciplinary Education, Research and Development

    Annotation graphs and annotation servers offer infrastructure to support the analysis of human language resources in the form of time-series data such as text, audio and video. This paper outlines areas of common need among empirical linguists and computational linguists. After reviewing examples of data and tools used or under development for each of several areas, it proposes a common framework for future tool development, data annotation and resource sharing based upon annotation graphs and servers.Comment: 8 pages, 6 figure

    Flow transitions in two-dimensional foams

    For sufficiently slow rates of strain, flowing foam can exhibit inhomogeneous flows. The nature of these flows is an area of active study in both two-dimensional model foams and three dimensional foam. Recent work in three-dimensional foam has identified three distinct regimes of flow [S. Rodts, J. C. Baudez, and P. Coussot, Europhys. Lett. {\bf 69}, 636 (2005)]. Two of these regimes are identified with continuum behavior (full flow and shear-banding), and the third regime is identified as a discrete regime exhibiting extreme localization. In this paper, the discrete regime is studied in more detail using a model two dimensional foam: a bubble raft. We characterize the behavior of the bubble raft subjected to a constant rate of strain as a function of time, system size, and applied rate of strain. We observe localized flow that is consistent with the coexistence of a power-law fluid with rigid body rotation. As a function of applied rate of strain, there is a transition from a continuum description of the flow to discrete flow when the thickness of the flow region is approximately 10 bubbles. This occurs at an applied rotation rate of approximately 0.07s10.07 {\rm s^{-1}}

    ATPase mechanism of the 5'-3' DNA helicase, RecD2: evidence for a pre-hydrolysis conformation change

    The superfamily 1 helicase, RecD2, is a monomeric, bacterial enzyme with a role in DNA repair, but with 5'-3' activity unlike most enzymes from this superfamily. Rate constants were determined for steps within the ATPase cycle of RecD2 in the presence of ssDNA. The fluorescent ATP analog, mantATP (2'(3')-O-(N-methylanthraniloyl)ATP), was used throughout to provide a complete set of rate constants and determine the mechanism of the cycle for a single nucleotide species. Fluorescence stopped-flow measurements were used to determine rate constants for adenosine nucleotide binding and release, quenched-flow measurements were used for the hydrolytic cleavage step, and the fluorescent phosphate biosensor was used for phosphate release kinetics. Some rate constants could also be measured using the natural substrate, ATP, and these suggested a similar mechanism to that obtained with mantATP. The data show that a rearrangement linked to Mg(2+) coordination, which occurs before the hydrolysis step, is rate-limiting in the cycle and that this step is greatly accelerated by bound DNA. This is also shown here for the PcrA 3'-5' helicase and so may be a general mechanism governing superfamily 1 helicases. The mechanism accounts for the tight coupling between translocation and ATPase activity

    A challenging redox neutral Cp*Co(III)-catalysed alkylation of acetanilides with 3-buten-2-one: synthesis and key insights into the mechanism through DFT calculations

    Traditional, established palladium cross-coupling procedures are widely applied in complex molecule synthesis; however, there is a significant disadvantage in the requirement for pre-functionalised substrates (commonly halides/triflates). Direct C–H activation protocols provide the opportunity for a novel approach to synthesis, although this field is still in its relative infancy and often transferability between substrate classes remains unresolved and limitations not fully understood. This study focuses on the translation of an established Cp*Co(III)-catalysed alkylation of benzamides to related acetanilides using 3-buten-2-one as coupling partner. The developed procedure provides a wide substrate scope in terms of substituted acetanilides, although the optimised conditions were found to be more forcing than those for the corresponding benzamide substrates. Interestingly, density functional theory (DFT) studies reveal that the major impediment in the mechanism is not the C–H activation step, but instead and unexpectedly, effective competition with more stable compounds (resting states) not involved in the catalytic cycle

    This paper reviews the literature on taxation of the informal economy, taking stock of key debates and drawing attention to recent innovations. Conventionally, the debate on whether to tax has frequently focused on the limited revenue potential, high cost of collection, and potentially adverse impact on small firms. Recent arguments have increasingly emphasised the more indirect benefits of informal taxation in relation to economic growth, broader tax compliance, and governance. More research is needed, we argue, into the relevant costs and benefits for all, including quasi-voluntary compliance, political and administrative incentives for reform, and citizen-state bargaining over taxation

    Monomeric PcrA helicase processively unwinds plasmid lengths of DNA in the presence of the initiator protein RepD

    The helicase PcrA unwinds DNA during asymmetric replication of plasmids, acting with an initiator protein, in our case RepD. Detailed kinetics of PcrA activity were measured using bulk solution and a single-molecule imaging technique to investigate the oligomeric state of the active helicase complex, its processivity and the mechanism of unwinding. By tethering either DNA or PcrA to a microscope coverslip surface, unwinding of both linear and natural circular plasmid DNA by PcrA/RepD was followed in real-time using total internal reflection fluorescence microscopy. Visualization was achieved using a fluorescent single-stranded DNA-binding protein. The single-molecule data show that PcrA, in combination with RepD, can unwind plasmid lengths of DNA in a single run, and that PcrA is active as a monomer. Although the average rate of unwinding was similar in single-molecule and bulk solution assays, the single-molecule experiments revealed a wide distribution of unwinding speeds by different molecules. The average rate of unwinding was several-fold slower than the PcrA translocation rate on single-stranded DNA, suggesting that DNA unwinding may proceed via a partially passive mechanism. However, the fastest dsDNA unwinding rates measured in the single-molecule unwinding assays approached the PcrA translocation speed measured on ssDNA

    Describing and prescribing the constitutive response of yield stress fluids using large amplitude oscillatory shear stress (LAOStress)

    Large amplitude oscillatory shear (LAOS) is used as a tool to probe the nonlinear rheological response of a model elasto-viscoplastic material (a Carbopol microgel). In contrast to most recent studies, these large amplitude measurements are carried out in a stress-controlled manner. We outline a descriptive framework of characterization measures for nonlinear rheology under stress-controlled LAOS, and this is contrasted experimentally to the strain-controlled framework that is more commonly used. We show that this stress-controlled methodology allows for a physically intuitive interpretation of the yielding behavior of elasto-viscoplastic materials. The insight gained into the material behavior through these nonlinear measures is then used to develop two constitutive models that prescribe the rheological response of the Carbopol microgel. We show that these two successively more sophisticated constitutive models, which are based on the idea of strain decomposition, capture in a compact manner the important features of the nonlinear rheology of the microgel. The second constitutive model, which incorporates the concept of kinematic hardening, embodies all of the essential behaviors exhibited by Carbopol. These include elasto-viscoplastic creep and time-dependent viscosity plateaus below a critical stress, a viscosity bifurcation at the critical stress, and Herschel–Bulkley flow behavior at large stresses

    Integrating detectors and their application to infrared astronomy

    The work contained in this thesis is concerned with the performance of infrared integrating detector arrays, within the context of astronomical spectroscopy.A linear array of thirty-two InSb photo diodes is investigated. It is found to exhibit good capacitance and dark current uniformity across the array. By applying the principle of charge conservation to the multiplexed readout arrangement of this device, the signal response of the detector to different levels of illumination is derived. It is found from this, and confirmed experimentally, that the device has a highly linear radiation response over a range of reverse biases.The interaction between dark current and photo-current is studied,primarily by the application of a simple model. The results indicate that the effective signal gain of a detector can vary in the situation where dark current dominates the discharge processes, since in this instance accurate dark current subtraction becomes difficult.The predictions of the model are compared with experiments performed on two integrating arrays; one under study in the laboratory,and the other installed in the low background environment of a cooled grating spectrometer. Finally, suggestions are presented of ways of avoiding this problem, the simplest of which involves utilizing, where possible, low dark current detector materials.The importance of achieving good dark current uniformity with arrays is stressed, since this will improve the ability to flat-field faint object spectra.To illustrate the importance of these devices, infrared spectra obtained with array detectors, covering a range of astronomical objects,are presented and discussed

    Measuring the capability to raise revenue process and output dimensions and their application to the Zambia revenue authority

    The worldwide diffusion of the good governance agenda and new public management has triggered a renewed focus on state capability and, more specifically, on the capability to raise revenue in developing countries. However, the analytical tools for a comprehensive understanding of the capability to raise revenue remain underdeveloped. This article aims at filling this gap and presents a model consisting of the three process dimensions ‘information collection and processing’, ‘merit orientation’ and ‘administrative accountability’. ‘Revenue performance’ constitutes the fourth capability dimension which assesses tax administration’s output. This model is applied to the case of the Zambia Revenue Authority. The dimensions prove to be valuable not only for assessing the how much but also the how of collecting taxes. They can be a useful tool for future comparative analyses of tax administrations’ capabilities in developing countries.Die weltweite Verbreitung der Good-Governance- und New-Public-Management-Konzepte hat zu einer zunehmenden Konzentration auf staatliche Leistungsfähigkeit und, im Besonderen, auf die Leistungsfähigkeit der Steuererhebung in Entwicklungsländern geführt. Allerdings bleiben die analytischen Werkzeuge für ein umfassendes Verständnis von Leistungsfähigkeit unterentwickelt. Dieser Artikel stellt hierfür ein Modell vor, das die drei Prozess-Dimensionen „Sammeln und Verarbeiten von Informationen“, „Leistungsorientierung der Mitarbeiter“ und „Verantwortlichkeit der Verwaltung“ beinhaltet. „Einnahmeperformanz“ ist die vierte Dimension und erfasst den Output der Steuerverwaltung. Das mehrdimensionale Modell wird für die Analyse der Leistungsfähigkeit der Steuerbehörde Zambias (Zambia Revenue Authority) genutzt. Es erweist sich nicht nur für die Untersuchung des Wieviel, sondern auch des Wie des Erhebens von Steuern als wertvoll. Die vier Dimensionen können in Zukunft zur umfassenden und vergleichenden Analyse der Leistungsfähigkeit verschiedener Steuerverwaltungen in Entwicklungsländern genutzt werden

    Multi-Task Time Series Analysis applied to Drug Response Modelling

    Time series models such as dynamical systems are frequently fitted to a cohort of data, ignoring variation between individual entities such as patients. In this paper we show how these models can be personalised to an individual level while retaining statistical power, via use of multi-task learning (MTL). To our knowledge this is a novel development of MTL which applies to time series both with and without control inputs. The modelling framework is demonstrated on a physiological drug response problem which results in improved predictive accuracy and uncertainty estimation over existing state-of-the-art models.Comment: To appear in AISTATS 201
