    Evaluation of enzymatic techniques for screening amphetamines and alcohol in oral fluid

    Evaluations of the Immunalysis enzyme linked immunosorbent assay (ELISA) and Immunalysis enzymatic assay for ethyl alcohol were undertaken to evaluate their suitability for screening drugs of abuse (namely amphetamine and methamphetamine) and alcohol in oral fluid samples collected with the Quantisal Collection Device. Multi-analyte controls were prepared for the drugs of abuse screen and diluted with Quantisal buffer prior to analysis to match the dilution in the Quantisal Oral Fluid Collection Device that was used to collect the samples. These samples were analysed over time to evaluate stability and case samples were analysed to evaluate sensitivity and specificity. Alcohol calibrators and controls were evaluated for linearity and stability before being applied to case samples to evaluate the sensitivity and specificity of the method. The amphetamine assay was found to be highly sensitive and specific. The methamphetamine assay was found to be highly specific but no positive samples were analysed so the sensitivity could not be evaluated. The multi-analyte controls were found to be stable over a fourteen month period. The Immunalysis ELISA assays were found to be suitable for screening oral fluid samples. The alcohol assay was found to be linear over the 0 – 300mg/dL range and the calibrators and controls were found to be stable over time. The assay was found to be highly sensitive and specific and best suited to high throughput laboratories expecting mainly negative samples. However, it would not be cost effective for smaller laboratories or those expecting a high number of positives, where going straight to confirmation by head-space gas chromatography with flame ionisation detection would be recommended

    Soil-sebum partition coefficients for high molecular weight polycyclic aromatic hydrocarbons (HMW-PAH)

    In vitro high molecular weight polycyclic aromatic hydrocarbons (HMW-PAH) soil-sebum partition coefficients (KSS) were measured for twelve soils collected from former UK gasworks. Concentrations of ∑16 USEPA PAH in the soils ranged from 51 to 1440 mg/kg. Time series extractions (0.5, 1, 2, 4, 8 and 24 h) at skin temperature (32 °C) of HMW-PAH from sebum to soil for two samples were conducted to determine the maximum release time-step. The maximum HMW-PAH release time-step was determined as 4 h, which was subsequently used as the extraction time for the remaining samples. The 4 h extraction for all samples showed HMW-PAH KSS ranging from 0.06 to 1.4 (median = 0.59; mean = 0.59; standard deviation = 0.27; median absolute deviation = 0.29). Evaluation of KSS data for the 4 h extractions showed that soil type and selected HMW-PAH properties (literature based molecular weight and octanol-carbon partition coefficients) affect the amount of HMW-PAH released from soil into sebum

    Pilot study assessing pathophysiology and healing of digital ulcers in patients with systemic sclerosis using laser Doppler imaging and thermography

    OBJECTIVES: Systemic sclerosis (SSc)-related digital ulcers (DU) cause significant pain and disability and are often a primary endpoint in clinical trials. However, their pathophysiology has been little studied. The objectives of this prospective study were to determine whether laser Doppler imaging (LDI) and thermography can identify ischaemic components in both fingertip and extensor surface DU and assess ulcer healing.METHODS: Patients prospectively reported new DU over a year. Patients' DU underwent imaging until the ulcer had healed. Ischaemia was defined as lower blood flow or skin temperature (and inflammation as higher) within the ulcer, compared to a non-affected site.RESULTS: 53 ulcers (19 fingertip, 18 extensor, 16 'other' sites) in 17 patients were imaged (53 with LDI, 52 with thermography). For LDI data 32 (60%) ulcers were ischaemic; median perfusion ulcer/unaffected area; 0.79 (range 0.11-2.9). For thermography data 35 (66%) were ischaemic; 0.98 (0.89 to 1.1). Inflammation in the surrounding area was identified for all ulcers by LDI but not thermography. In the 36 ulcers with repeat imaging, LDI showed trends (with healing) towards increased ulcer perfusion (p=0.23) and decreased hyperaemia in adjacent areas (p=0.59). Skin temperature at the ulcer site showed no significant change (p=0.13) but adjacent area showed decreased temperature (p=0.04 signifying decreased blood flow).CONCLUSIONS: LDI and thermography are sufficiently sensitive to measure ischaemia in both fingertip and extensor ulcers. LDI was better suited to monitoring change in perfusion with healing (due to higher imaging resolution, or vascular changes occurring in more superficial skin layers).</p

    An overview of research and development themes in the measurement and occurrences of polyaromatic hydrocarbons in dusts and particulates

    Polycyclic aromatic hydrocarbons (PAHs) are a group of organic compounds consisting of two or more fused aromatic rings and are probably one of the most studied groups of organic chemicals in environmental research. PAHs originate mainly from anthropogenic processes, particularly from incomplete combustion of organic fuels. PAHs are distributed widely in particulate matter. Due to widespread sources and persistent characteristics, PAHs disperse through atmospheric transport and exist almost everywhere. Human beings are exposed to PAH mixtures in gaseous or particulate phases in ambient air. Long-term exposure to high concentrations of PAHs is associated with adverse health problems. This review identifies the main research and development themes in the measurement and occurrences of PAHs in dusts and particulates using a new approach to carrying out a literature review where many peer-review publications have been produced. The review extracts the most important research themes from a literature search using a combination of text mining and a more detailed review of selected papers from within the identified theme
