    A Bridge to the Future: Cultivating Academic Leadership Today for Better Communities Tomorrow

    The purpose of this project is to develop a plan for expanding and funding the Grace E. Harris Leadership Institute (GEHLI). Currently, teams of leadership scholars selected from Virginia Commonwealth University (VCU) develop projects relating to the academic community in addition to studying the dynamics of effective leadership. Consistent with VCU\u27s vision of partnering with and service to metropolitan Richmond, we propose that future teams of leadership scholars develop outreach projects that involve and benefit the local community. The training in which scholars would participate would include learning about the Richmond community, its needs and goals, as well as the dynamics of leadership. Project teams would collaborate with community groups or leaders, thereby strengthening the bonds between VCU and the community, and providing public service, in accordance with VCU\u27s mission. Projects might include education, outreach, fundraising, mentoring, or research that would benefit Richmond and provide scholars with opportunities to network with local leaders. Financial and philanthropic resources will be investigated and a list of potential funding organizations will be generated

    Effects of Anion Identity and Concentration on Electrochemical Reduction of CO2

    The electrochemical reduction of CO2 is known to be influenced by the concentration and identity of the anionic species in the electrolyte; however, a full understanding of this phenomenon has not been developed. Here, we present the results of experimental and computational studies aimed at understanding the role of electrolyte anions on the reduction of CO2 over Cu surfaces. Experimental studies were performed to show the effects of bicarbonate buffer concentration and the composition of other buffering anions on the partial currents of the major products formed by reduction of CO2 over Cu. It was demonstrated that the composition and concentration of electrolyte anions has relatively little effect on the formation of CO, HCOO−, C2H4, and CH3CH2OH, but has a significant effect on the formation of H2 and CH4. Continuum modeling was used to assess the effects of buffering anions on the pH at the electrode surface. The influence of pH on the activity of Cu for producing H2 and CH4 was also considered. Changes in the pH near the electrode surface were insufficient to explain the differences in activity and selectivity observed with changes in anion buffering capacity observed for the formation of H2 and CH4. Therefore, it is proposed that these differences are the result of the ability of buffering anions to donate hydrogen directly to the electrode surface and in competition with water. The effectiveness of buffering anions to serve as hydrogen donors is found to increase with decreasing pKa of the buffering anion

    Anatomy and lateralization of the human corticobulbar tracts: an fMRI-guided tractography study

    The left hemisphere lateralization bias for language functions, such as syntactic processing and semantic retrieval, is well known. Although several theories and clinical data indicate a link between speech motor execution and language, the functional and structural brain lateralization for these functions has never been examined concomitantly in the same individuals. Here we used functional MRI during rapid silent syllable repetition (/lalala/, /papapa/ and /pataka/, known as oral diadochokinesis or DDK) to map the cortical representation of the articulators in 17 healthy adults. In these same participants, functional lateralization for language production was assessed using the well established verb generation task. We then used DDK-related fMRI activation clusters to guide tractography of the corticobulbar tract from diffusion-weighted MRI. Functional MRI revealed a wide inter-individual variability of hemispheric asymmetry patterns (left and right dominant, as well as bilateral) for DDK in the motor cortex, despite predominantly left hemisphere dominance for language-related activity in Broca’s area. Tractography revealed no evidence for structural asymmetry (based on fractional anisotropy) within the corticobulbar tract. To our knowledge, this study is the first to reveal that motor brain activation for syllable repetition is unrelated to functional asymmetry for language production in adult humans. In addition, we found no evidence that the human corticobulbar tract is an asymmetric white matter pathway. We suggest that the predominance of dysarthria following left hemisphere infarct is probably a consequence of disrupted feedback or input from left hemisphere language and speech planning regions, rather than structural asymmetry of the corticobulbar tract itself

    Snapshot Provisioning of Cloud Application Stacks to Face Traffic Surges

    Traffic surges, like the Slashdot effect, occur when a web application is overloaded by a huge number of requests, potentially leading to unavailability. Unfortunately, such traffic variations are generally totally unplanned, of great amplitude, within a very short period, and a variable delay to return to a normal regime. In this report, we introduce PeakForecast as an elastic middleware solution to detect and absorb a traffic surge. In particular, PeakForecast can, from a trace of queries received in the last seconds, minutes or hours, to detect if the underlying system is facing a traffic surge or not, and then estimate the future traffic using a forecast model with an acceptable precision, thereby calculating the number of resources required to absorb the remaining traffic to come. We validate our solution by experimental results demonstrating that it can provide instantaneous elasticity of resources for traffic surges observed on the Japanese version of Wikipedia during the Fukushima Daiichi nuclear disaster in March 2011.Les pics de trafic, tels que l'effet Slashdot, apparaissent lorsqu'une application web doit faire face un nombre important de requêtes qui peut potentiellement entraîner une mise hors service de l'application. Malheureusement, de telles variations de traffic sont en général totalement imprévues et d'une grande amplitude, arrivent pendant une très courte période de temps et le retour à un régime normal prend un délai variable. Dans ce rapport, nous présentons PeakForecast qui est une solution intergicielle élastique pour détecter et absorber les pics de trafic. En particulier, PeakForecast peut, à partir des traces de requêtes reçues dans les dernières secondes, minutes ou heures, détecter si le système sous-jacent fait face ou non à un pic de trafic, estimer le trafic futur en utilisant un modèle de prédiction suffisamment précis, et calculer le nombre de ressources nécessaires à l'absorption du trafic restant à venir. Nous validons notre solution avec des résultats expérimentaux qui démontrent qu'elle fournit une élasticité instantanée des ressources pour des pics de trafic qui ont été observés sur la version japonaise de Wikipedia lors de l'accident nucléaire de Fukushima Daiichi en mars 2011

    Phenotypic microarrays suggest Escherichia coli ST131 is not a metabolically distinct lineage of extra-intestinal pathogenic E. coli

    Extraintestinal pathogenic E. coli (ExPEC) are the major aetiological agent of urinary tract infections (UTIs) in humans. The emergence of the CTX-M producing clone E. coli ST131 represents a major challenge to public health worldwide. A recent study on the metabolic potential of E. coli isolates demonstrated an association between the E. coli ST131 clone and enhanced utilisation of a panel of metabolic substrates. The studies presented here investigated the metabolic potential of ST131 and other major ExPEC ST isolates using 120 API test reagents and found that ST131 isolates demonstrated a lower metabolic activity for 5 of 120 biochemical tests in comparison to non-ST131 ExPEC isolates. Furthermore, comparative phenotypic microarray analysis showed a lack of specific metabolic profile for ST131 isolates countering the suggestion that these bacteria are metabolically fitter and therefore more successful human pathogens

    First Steps towards Underdominant Genetic Transformation of Insect Populations

    The idea of introducing genetic modifications into wild populations of insects to stop them from spreading diseases is more than 40 years old. Synthetic disease refractory genes have been successfully generated for mosquito vectors of dengue fever and human malaria. Equally important is the development of population transformation systems to drive and maintain disease refractory genes at high frequency in populations. We demonstrate an underdominant population transformation system in Drosophila melanogaster that has the property of being both spatially self-limiting and reversible to the original genetic state. Both population transformation and its reversal can be largely achieved within as few as 5 generations. The described genetic construct {Ud} is composed of two genes; (1) a UAS-RpL14.dsRNA targeting RNAi to a haploinsufficient gene RpL14 and (2) an RNAi insensitive RpL14 rescue. In this proof-of-principle system the UAS-RpL14.dsRNA knock-down gene is placed under the control of an Actin5c-GAL4 driver located on a different chromosome to the {Ud} insert. This configuration would not be effective in wild populations without incorporating the Actin5c-GAL4 driver as part of the {Ud} construct (or replacing the UAS promoter with an appropriate direct promoter).     Better Workers, Better Elections? Electoral Management Body Workforces and Electoral Integrity Worldwide

    Building better elections is a central task for the study of democracy and democratisation. Despite this, there have been no cross-national studies on the staff who manage and implement elections: electoral management body (EMB) workforces. This article provides the first macroscopic worldwide picture of workforce characteristics, human resource management practices and employee outcomes, and analyses the effects they have on electoral integrity, based on original international surveys of electoral management bodies (EMBs) (n = 51) and electoral officials (n = 2029). Drawing from the human resource management literature, a framework is developed to explain how these factors might interact with EMB performance. Analysis demonstrates them to be highly related. Adding data on human resource management practices and employee outcomes improves explanatory models designed to predict the performance of EMBs. Chiefly, EMBs that enable greater opportunities for employees to be involved in decision-making processes perform better. Recruitment practices, job satisfaction and levels of stress are also important

    Contribution of Matrix Metalloproteinase-9 to Cerebral Edema and Functional Outcome following Experimental Subarachnoid Hemorrhage

    Background: Cerebral edema is an important risk factor for death and poor outcome following subarachnoid hemorrhage (SAH). However, underlying mechanisms are still poorly understood. Matrix metalloproteinase (MMP)-9 is held responsible for the degradation of microvascular basal lamina proteins leading to blood-brain barrier dysfunction and, thus, formation of vasogenic cerebral edema. The current study was conducted to clarify the role of MMP-9 for the development of cerebral edema and for functional outcome after SAH. Methods: SAH was induced in FVB/N wild-type (WT) or MMP-9 knockout (MMP-9(-/-)) mice by endovascular puncture. Intracranial pressure (ICP), regional cerebral blood flow (rCBF), and mean arterial blood pressure (MABP) were continuously monitored up to 30 min after SAH. Mortality was quantified for 7 days after SAH. In an additional series neurological function and body weight were assessed for 3 days after SAH. Subsequently, ICP and brain water content were quantified. Results: Acute ICP, rCBF, and MABP did not differ between WT and MMP-9(-/-) mice, while 7 days' mortality was lower in MMP-9(-/-) mice (p = 0.03; 20 vs. 60%). MMP-9(-/-) mice also exhibited better neurological recovery, less brain edema formation, and lower chronic ICP. Conclusions: The results of the current study suggest that MMP-9 contributes to the development of early brain damage after SAH by promoting cerebral edema formation.     Vascular responses of the extremities to transdermal application of vasoactive agents in Caucasian and African descent individuals

Purpose: Individuals of African descent (AFD) are more susceptible to non-freezing cold injury than Caucasians (CAU) which may be due, in part, to differences in the control of skin blood flow. We investigated the skin blood flow responses to transdermal application of vasoactive agents. Methods: Twenty-four young males (12 CAU and 12 AFD) undertook three tests in which iontophoresis was used to apply acetylcholine (ACh 1 w/v %), sodium nitroprusside (SNP 0.01 w/v %) and noradrenaline (NA 0.5 mM) to the skin. The skin sites tested were: volar forearm, non-glabrous finger and toe, and glabrous finger (pad) and toe (pad). Results: In response to SNP on the forearm, AFD had less vasodilatation for a given current application than CAU (P = 0.027–0.004). ACh evoked less vasodilatation in AFD for a given application current in the non-glabrous finger and toe compared with CAU (P = 0.043–0.014) with a lower maximum vasodilatation in the non-glabrous finger (median [interquartile], AFD n = 11, 41[234] %, CAU n = 12, 351[451] %, P = 0.011) and non-glabrous toe (median [interquartile], AFD n = 9, 116[318] %, CAU n = 12, 484[720] %, P = 0.018). ACh and SNP did not elicit vasodilatation in the glabrous skin sites of either group. There were no ethnic differences in response to NA. Conclusion: AFD have an attenuated endothelium-dependent vasodilatation in non-glabrous sites of the fingers and toes compared with CAU. This may contribute to lower skin temperature following cold exposure and the increased risk of cold injuries experienced by AFD.Published versio
