Exact Fermi coordinates for a class of spacetimes
We find exact Fermi coordinates for timelike geodesic observers for a class
of spacetimes that includes anti-de Sitter spacetime, de Sitter spacetime, the
constant density interior Schwarzschild spacetime with positive, zero, and
negative cosmological constant, and the Einstein static universe. Maximal
charts for Fermi coordinates are discussed.Comment: 15 page
Does operational oceanography address the needs of fisheries and applied environmental scientists?
Although many oceanographic data products are now considered operational, continued dialogue between data producers and their user communities is still needed. The fisheries and environmental science communities have often been criticized for their lack of multidisciplinarity, and it is not clear whether recent developments in operational oceanographic products are addressing these needs. The International Council for the Exploration of the Sea (ICES) Working Group on Operational Oceanographic products for Fisheries and Environment (WGOOFE) identified a potential mismatch between user requirements and the perception of requirements by the providers. Through a questionnaire (98 respondents), WGOOFE identified some of these issues. Although products of physical variables were in higher demand, several biological parameters scored in the top 10 rankings. Users placed specific focus on historic time series products with monthly or annual resolution and updating on similar time scales. A significant percentage requested access to numerical data rather than graphical output. While the outcomes of this survey challenge our views of operational oceanography, several initiatives are already attempting to close the gap between user requirements and products available
Patient-specific image-based computer simulation for theprediction of valve morphology and calcium displacement after TAVI with the Medtronic CoreValve and the Edwards SAPIEN valve
AIMS:
Our aim was to validate patient-specific software integrating baseline anatomy and biomechanical properties of both the aortic root and valve for the prediction of valve morphology and aortic leaflet calcium displacement after TAVI.
METHODS AND RESULTS:
Finite element computer modelling was performed in 39 patients treated with a Medtronic CoreValve System (MCS; n=33) or an Edwards SAPIEN XT (ESV; n=6). Quantitative axial frame morphology at inflow (MCS, ESV) and nadir, coaptation and commissures (MCS) was compared between multislice computed tomography (MSCT) post TAVI and a computer model as well as displacement of the aortic leaflet calcifications, quantified by the distance between the coronary ostium and the closest calcium nodule. Bland-Altman analysis revealed a strong correlation between the observed (MSCT) and predicted frame dimensions, although small differences were detected for, e.g., Dmin at the inflow (mean±SD MSCT vs.
MODEL:
21.6±2.4 mm vs. 22.0±2.4 mm; difference±SD: -0.4±1.3 mm, p<0.05) and Dmax (25.6±2.7 mm vs. 26.2±2.7 mm; difference±SD: -0.6±1.0 mm, p<0.01). The observed and predicted calcium displacements were highly correlated for the left and right coronary ostia (R2=0.67 and R2=0.71, respectively p<0.001).
CONCLUSIONS:
Dedicated software allows accurate prediction of frame morphology and calcium displacement after valve implantation, which may help to improve outcome
Extended Fermi coordinates
We extend the notion of Fermi coordinates to a generalized definition in
which the highest orders are described by arbitrary functions. From this
definition rises a formalism that naturally gives coordinate transformation
formulae. Some examples are developped in which the extended Fermi coordinates

Evaluating the suitability of coupled biophysical models for fishery management
The potential role of coupled biophysical models in enhancing the conservation, management, and recovery of fish stocks is assessed, with emphasis on anchovy, cod, herring, and sprat in European waters. The assessment indicates that coupled biophysical models are currently capable of simulating transport patterns, along with temperature and prey fields within marine ecosystems; they therefore provide insight into the variability of early-life-stage dynamics and connectivity within stocks. Moreover, the influence of environmental variability on potential recruitment success may be discerned from model hindcasts. Based on case studies, biophysical modelling results are shown to be capable of shedding light on whether stock management frameworks need re-evaluation. Hence, key modelling products were identified that will contribute to the development of viable stock recovery plans and management strategies. The study also suggests that approaches combining observation, process knowledge, and numerical modelling could be a promising way forward in understanding and simulating the dynamics of marine fish populations
DEVELOPING NEW APPROACHES TO GLOBAL STOCK STATUS ASSESSMENT AND FISHERY PRODUCTION POTENTIAL OF THE SEAS
Stock status is a key parameter for evaluating the sustainability of fishery resources and developing
corresponding management plans. However, the majority of stocks are not assessed, often as a result of
insufficient data and a lack of resources needed to execute formal stock assessments. The working
group involved in this publication focused on two approaches to estimating fisheries status: one based

Spatial Regulation of Membrane Fusion Controlled by Modification of Phosphoinositides
Membrane fusion plays a central role in many cell processes from vesicular
transport to nuclear envelope reconstitution at mitosis but the mechanisms that
underlie fusion of natural membranes are not well understood. Studies with
synthetic membranes and theoretical considerations indicate that accumulation of
lipids characterised by negative curvature such as diacylglycerol (DAG)
facilitate fusion. However, the specific role of lipids in membrane fusion of
natural membranes is not well established. Nuclear envelope (NE) assembly was
used as a model for membrane fusion. A natural membrane population highly
enriched in the enzyme and substrate needed to produce DAG has been isolated and
is required for fusions leading to nuclear envelope formation, although it
contributes only a small amount of the membrane eventually incorporated into the
NE. It was postulated to initiate and regulate membrane fusion. Here we use a
multidisciplinary approach including subcellular membrane purification,
fluorescence spectroscopy and Förster resonance energy transfer
(FRET)/two-photon fluorescence lifetime imaging microscopy (FLIM) to demonstrate
that initiation of vesicle fusion arises from two unique sites where these
vesicles bind to chromatin. Fusion is subsequently propagated to the endoplasmic
reticulum-derived membranes that make up the bulk of the NE to ultimately
enclose the chromatin. We show how initiation of multiple vesicle fusions can be
controlled by localised production of DAG and propagated bidirectionally.
Phospholipase C (PLCγ), GTP hydrolysis and
(phosphatidylinsositol-(4,5)-bisphosphate (PtdIns(4,5)P2) are

Influence of ovarian hormones deprivation on gene expression in the lower urinary tract of rats
OBJECTIVE: Identify the influence of ovarian hormone deprivation in expression genes on the lower urinary tract of rats. MATERIALS AND METHODS: This study deals with gene screening on lower urinary tract of rats. Fifty isogenic rats divided in two groups of twenty-five animals have their lower urinary tract surgically removed: group I, ovariectomized rats 30 days prior to surgery; group II, non-ovariectomized rats. Total RNA was isolated from bladder and urethra, and differential expression of genes was analyzed quantitative, qualitative and comparatively by array technology and RT-PCR. RESULTS: A total of 76 candidate genes were identified as differentially expressed between the groups, 26 being lower expressed in group II, and 50 in group I. Among them, differential expression validation was confirmed by RT-PCR for three lower expressed genes in group I: Vascular Endothelial Growth Factor (VEGF), Beta-2 Microglobulin (B2M) and Cytochrome c Oxidase subunit I (COX I). CONCLUSION: Ovarian hormone deprivation influences the expression genes on lower urinary tract. We demonstrated that a 30-day period of castration down regulate the expression of VEGF, B2M and COX I in adult rats which are involved in activities of angiogenesis, immune responses and cellular metabolism respectively.Federal University of São Paulo Department of GynecologyUNIFESP, Department of GynecologySciEL
The Role of Paracrine and Autocrine Signaling in the Early Phase of Adipogenic Differentiation of Adipose-derived Stem Cells.
INTRODUCTION: High cell density is known to enhance adipogenic differentiation of mesenchymal stem cells, suggesting secretion of signaling factors or cell-contact-mediated signaling. By employing microfluidic biochip technology, we have been able to separate these two processes and study the secretion pathways. METHODS AND RESULTS: Adipogenic differentiation of human adipose-derived stem cells (ASCs) cultured in a microfluidic system was investigated under perfusion conditions with an adipogenic medium or an adipogenic medium supplemented with supernatant from differentiating ASCs (conditioned medium). Conditioned medium increased adipogenic differentiation compared to adipogenic medium with respect to accumulation of lipid-filled vacuoles and gene expression of key adipogenic markers (C/EBPα, C/EBPβ, C/EBPδ, PPARγ, LPL and adiponectin). The positive effects of conditioned medium were observed early in the differentiation process. CONCLUSIONS: Using different cell densities and microfluidic perfusion cell cultures to suppress the effects of cell-released factors, we have demonstrated the significant role played by auto- or paracrine signaling in adipocyte differentiation. The cell-released factor(s) were shown to act in the recruitment phase of the differentiation process
