    Operational modal analysis of a spar-type floating platform using frequency domain decomposition method

    System identification of offshore floating platforms is usually performed by testing small-scale models in wave tanks, where controlled conditions, such as still water for free decay tests, regular and irregular wave loading can be represented. However, this approach may result in constraints on model dimensions, testing time, and costs of the experimental activity. For such reasons, intermediate-scale field modelling of offshore floating structures may become an interesting as well as cost-effective alternative in a near future. Clearly, since the open sea is not a controlled environment, traditional system identification may become challenging and less precise. In this paper, a new approach based on Frequency Domain Decomposition (FDD) method for Operational Modal Analysis is proposed and validated against numerical simulations in ANSYS AQWA v.16.0 on a simple spar-type structure. The results obtained match well with numerical predictions, showing that this new approach, opportunely coupled with more traditional wave tanks techniques, proves to be very promising to perform field-site identification of the model structures

    Les communautés théâtrales du XXe siècle sont l'œuvre de projets visionnaires menés par des figures charismatiques. En 1924, lassé de l'exploitation de sa salle parisienne du Vieux- Colombier, Jacques Copeau fonde en Bourgogne, à l'abri des rumeurs citadines, une communauté d'élèves et de comédiens férus d'expérimentation. A la même époque, il retrouve la foi catholique. Si les travaux antérieurs sur les Copiaus (1924-1929) mentionnent systématiquement la " crise religieuse " de Jacques Copeau à partir de 1925, il n'existe aucune étude approfondie sur le lien entre la vie communautaire des Copiaus et la ferveur catholique de son fondateur. La référence explicite de Jacques Copeau à la Règle de saint Benoît, ainsi que l'examen de sources inexplorées, révèlent que Jacques Copeau entretient des liens étroits avec le milieu des intellectuels catholiques de l'époque. Dès octobre 1924, Jacques Copeau est profondément inspiré par la " première " Règle monastique chrétienne ainsi que par les notions religieuses d'obéissance librement consentie et de moralité de l'art. Cette étude contribue à comprendre les aspirations contradictoires et les difficultés d'entente entre un " Patron " et un groupe de jeunes gens animé par le désir de créations théâtrales collectives

    Progress on the experimental set-up for the testing of a floating offshore wind turbine scaled model in a field site

    This document describes design and realization of a small-scale field experiment on a 1:30 model of spar floating support structure for offshore wind turbines. The aim of the experiment is to investigate the dynamic behaviour of the floating wind turbine under extreme wave and parked rotor conditions. The experiment has been going on in the Natural Ocean Engineering Laboratory of Reggio Calabria (Italy). In this article, all the stages of the experimental activity are presented, and some results are shown in terms of motions and response amplitude operators. Finally, a comparison with corresponding results obtained using ANSYS AQWA software package is shown, and conclusions are drawn. The presented experimental set-up seems promising to test offshore floating structures for marine renewable energy at a relatively large scale in the Natural Ocean Engineering Laboratory field site

    Assembly of Influenza Hemagglutinin Fusion Peptides in a Phospholipid Bilayer by Coarse-grained Computer Simulations

    Membrane fusion is critical to eukaryotic cellular function and crucial to the entry of enveloped viruses such as influenza and human immunodeficiency virus. Influenza viral entry in the host cell is mediated by a 20-23 amino acid long sequence, called the fusion peptide (FP). Recently, possible structures for the fusion peptide (ranging from an inverted V shaped α-helical structure to an α-helical hairpin, or to a complete α-helix) and their implication in the membrane fusion initiation have been proposed. Despite the large number of studies devoted to the structure of the FP, the mechanism of action of this peptide remains unclear with several mechanisms having been suggested, including the induction of local disorder, promoting membrane curvature, and/or altering local membrane composition. In recent years, several research groups have employed atomistic and/or coarse-grained molecular dynamics (MD) simulations to investigate the matter. In all previous works, the behavior of a single FP monomer was studied, while in this manuscript, we use a simplified model of a tripeptide (TP) monomer of FP (TFP) instead of a single FP monomer because each Influenza Hemagglutinin contains three FP molecules in the biological system. In this manuscript we report findings targeted at understanding the fusogenic properties and the collective behavior of these trimers of FP peptides on a 1-palmitoyl-2-oleoyl-sn-glycero-3-phosphocholine model membrane. Here we show how the TFP monomers self-assemble into differently sized oligomers in the presence of the membrane. We measure the perturbation to the structure of the phospholipid membrane caused by the presence of these TFP oligomers. Our work (i) shows how self-assembly of TFP in the presence of the membrane induces non negligible deformation to the membrane and (ii) could be a useful starting point to stimulate discussion and further work targeted to fusion pore formation.</p

    Invertases are a widespread group of enzymes that catalyse the conversion of sucrose into fructose and glucose. Plants invertases and their substrates are essential factors that play an active role in primary metabolism and in cellular differentiation and by these activities they sustain development and growth. Being naturally present in multiple isoforms, invertases are known to be highly differentiated and tissue specific in such a way that every isoform is characteristic of a specific part of the plant. In this work, we report the identification of the invertase RhVI1 that was found to be highly expressed in rose petals. A characterization of this protein revealed that RhVI1 is a glycosylated membrane-anchored protein associated with the cytosolic side of the vacuolar membrane which occurs in vivo in a monomeric form. Purification yields have shown that the levels of expression decreased during the passage of petals from buds to mature and pre-senescent flowers. Moreover, the activity assay indicates RhVI1 to be an acidic vacuolar invertase. The physiological implications of these findings are discussed, suggesting a possible role of this protein during anthesis

    Brain-derived neurotrophic factor (BDNF) plays a crucial role in neurodevelopment, synaptic plasticity and neuronal function and survival. Serum and plasma BDNF levels are moderately, but consistently, decreased in patients with schizophrenia (SCZ) compared with healthy controls. There is a lack of knowledge, however, on the temporal manifestation of this decline. Clinical, illness course and treatment factors might influence the variation of BDNF serum levels in patients with psychosis. In this context, we propose a longitudinal study of a cohort of SCZ and schizophrenic and schizoaffective disorder (SAD) Sardinian patients with the aim of disentangling the relationship between peripheral BDNF serum levels and changes of psychopathology, cognition and drug treatments

    Output-only identification of rigid body motions of floating structures: a case study

    In order to identify rigid body motions of floating offshore structures, output-only techniques are very useful for developing low-cost intermediate-scale experimental activities directly into the sea, instead of wave tanks. A crucial parameter, however, is the length of the response records used as input for the identification process, since short records may result in significant loss of accuracy, while long ones may be incompatible with the assumption of stationarity of the sea state. This work presents a sensitivity study conducted on a numerical model of a spar structure, identified by means of Enhanced Frequency Domain Decomposition method. An overview on the efficiency of the method is given for various lengths of response record, along with practical indications on the minimum values acceptable

    High-density lipoprotein contribute to G0-G1/S transition in Swiss NIH/3T3 fibroblasts

    High density lipoproteins (HDLs) play a crucial role in removing excess cholesterol from peripheral tissues. Although their concentration is lower during conditions of high cell growth rate (cancer and infections), their involvement during cell proliferation is not known. To this aim, we investigated the replicative cycles in synchronized Swiss 3T3 fibroblasts in different experimental conditions: i) contact-inhibited fibroblasts re-entering cell cycle after dilution; ii) scratch-wound assay; iii) serum deprived cells induced to re-enter G1 by FCS, HDL or PDGF. Analyses were performed during each cell cycle up to quiescence. Cholesterol synthesis increased remarkably during the replicative cycles, decreasing only after cells reached confluence. In contrast, cholesteryl ester (CE) synthesis and content were high at 24h after dilution and then decreased steeply in the successive cycles. Flow cytometry analysis of DiO-HDL, as well as radiolabeled HDL pulse, demonstrated a significant uptake of CE-HDL in 24h. DiI-HDL uptake, lipid droplets (LDs) and SR-BI immunostaining and expression followed the same trend. Addition of HDL or PDGF partially restore the proliferation rate and significantly increase SR-BI and pAKT expression in serum-deprived cells. In conclusion, cell transition from G0 to G1/S requires CE-HDL uptake, leading to CE-HDL/SR-BI pathway activation and CEs increase into LDs

    Mucuna pruriens (Velvet bean) Rescues Motor, Olfactory, Mitochondrial and Synaptic Impairment in PINK1(B9) Drosophila melanogaster Genetic Model of Parkinson's Disease

    The fruit fly Drosophila melanogaster (Dm) mutant for PTEN-induced putative kinase 1 (PINK1B9) gene is a powerful tool to investigate physiopathology of Parkinson's disease (PD). Using PINK1B9 mutant Dm we sought to explore the effects of Mucuna pruriens methanolic extract (Mpe), a L-Dopa-containing herbal remedy of PD. The effects of Mpe on PINK1B9 mutants, supplied with standard diet to larvae and adults, were assayed on 3–6 (I), 10–15 (II) and 20–25 (III) days old flies. Mpe 0.1% significantly extended lifespan of PINK1B9 and fully rescued olfactory response to 1-hexanol and improved climbing behavior of PINK1B9 of all ages; in contrast, L-Dopa (0.01%, percentage at which it is present in Mpe 0.1%) ameliorated climbing of only PINK1B9 flies of age step II. Transmission electron microscopy analysis of antennal lobes and thoracic ganglia of PINK1B9 revealed that Mpe restored to wild type (WT) levels both T-bars and damaged mitochondria. Western blot analysis of whole brain showed that Mpe, but not L-Dopa on its own, restored bruchpilot (BRP) and tyrosine hydroxylase (TH) expression to age-matched WT control levels. These results highlight multiple sites of action of Mpe, suggesting that its effects cannot only depend upon its L-Dopa content and support the clinical observation of Mpe as an effective medication with intrinsic ability of delaying the onset of chronic L-Dopa-induced long-term motor complications. Overall, this study strengthens the relevance of using PINK1B9 Dm as a translational model to study the properties of Mucuna pruriens for PD treatment
