    Hornwort (Ceratophyllum demersum L.) and other aquatic macrophytes have historically been mechanically removed from the Rangiriri drain and Churchill East drain to maintain drain efficiency. As an alternative control method for the high plant biomass that accumulates at the end of summer, the effect of stocking diploid grass carp (Ctenopharyngodon idella L.) on the aquatic vegetation was evaluated in these Waikato drainage systems. At the start of the trial, both drains had a low diversity of aquatic macrophytes, and of the nine species (including the emergents), seven were exotic. Two months after grass carp were released to Churchill East drain (the treated drain) the four submerged and floating macrophyte species became scarce in the main drain. Over the same period, these species increased in biomass in Rangiriri drain (the untreated drain), where hornwort became dense and surface-reaching and remained so for the duration of the trial. However, grass carp did not control submerged vegetation in smaller side drains or the shallow, upper parts of the main drain, or the marginal sprawling species and emergent species. The cost of leasing the grass carp was similar to the cost of clearing the drains mechanically, but grass carp provided continuous weed control. However, subsequent to this trial, 62 dead grass carp were found in Churchill East drain in February 2001, and weed cover subsequently increased. This illustrates that grass carp management in New Zealand agricultural drains can be problematic due to periodic fish kills

    * Few dental professionals with child protection training have experience of making referrals. * There is a wide gap in practice between recognising signs of child abuse and neglect and responding effectively. * This may indicate missed opportunities to save children from continuing abuse. * There is a need for improved child protection information, support and training for dental professionals. Abstract Following several highly publicised inquiries into the deaths of children from abuse and neglect, there has been much recent interest in the role and responsibility of all health professionals to protect children at risk of maltreatment. The findings of a postal questionnaire, sent in March 2005 to 789 dentists and dental care professionals with an interest in paediatric dentistry working in varied settings in the UK, are presented in a two-part report and discussed in the context of current multi-agency good practice in safeguarding and promoting the welfare of children. This first part explores reported child protection training, experience and practice. There was a significant gap between recognising signs of abuse and responding effectively: 67% of respondents had suspected abuse or neglect of a child patient at some time in their career but only 29% had ever made a child protection referral. The dental profession is alerted to the need to ensure necessary appropriate action to safeguard children is always taken when child abuse or neglect are suspected

    Reduction in BACE1 decreases body weight, protects against diet-induced obesity and enhances insulin sensitivity in mice

    Insulin resistance and impaired glucose homoeostasis are important indicators of Type 2 diabetes and are early risk factors of AD (Alzheimer's disease). An essential feature of AD pathology is the presence of BACE1 (β-site amyloid precursor protein-cleaving enzyme 1), which regulates production of toxic amyloid peptides. However, whether BACE1 also plays a role in glucose homoeostasis is presently unknown. We have used transgenic mice to analyse the effects of loss of BACE1 on body weight, and lipid and glucose homoeostasis. BACE1−/− mice are lean, with decreased adiposity, higher energy expenditure, and improved glucose disposal and peripheral insulin sensitivity than wild-type littermates. BACE1−/− mice are also protected from diet-induced obesity. BACE1-deficient skeletal muscle and liver exhibit improved insulin sensitivity. In a skeletal muscle cell line, BACE1 inhibition increased glucose uptake and enhanced insulin sensitivity. The loss of BACE1 is associated with increased levels of UCP1 (uncoupling protein 1) in BAT (brown adipose tissue) and UCP2 and UCP3 mRNA in skeletal muscle, indicative of increased uncoupled respiration and metabolic inefficiency. Thus BACE1 levels may play a critical role in glucose and lipid homoeostasis in conditions of chronic nutrient excess. Therefore strategies that ameliorate BACE1 activity may be important novel approaches for the treatment of diabetes

    Moving beyond fitting fish into equations: Progressing the fish passage debate in the Anthropocene

    Realization of the importance of fish passage for migratory species has led to the development of innovative and creative solutions (‘fishways’) to mitigate the effects of artificial barriers in freshwater systems in the last few decades. In many instances, however, the first move has been to attempt to engineer a solution to the problem, thus attempting to ‘fit fish into an equation’. These fishways are often derived from designs targeting salmonids in the Northern Hemisphere. They are rarely adequate, even for these strong-swimming fish, and certainly appear to be unsuitable for most other species, not least for those of tropical regions. Fishway design criteria do not adequately account for natural variation among individuals, populations and species. Moreover, engineered solutions cannot reinstate the natural habitat and geomorphological properties of the river, objectives that have been largely ignored. This article discusses the most prominent issues with the current management and conservation of freshwater ecosystems as it pertains to fish passage. It is not intended as a review on fish passage, but rather a perspective on the issues related to fishways, as seen by practitioners.</p

    Enantiopure titanocene complexes: direct evidence for paraptosis in cancer cells

    Tolerated by normal tissues, anti-cancer therapies based on titanium compounds are limited by low efficacy/selectivity and lack of understanding of their mode(s) of action. In vitro antitumour activity and mode of cell death incurred by enantiopure TiCl2{n-C5H4CHEt(2 MeOPh)}2 (abbreviated CpR 2TiCl2) has been investigated. The in vitro anti-tumour activity of CpR 2TiCl2 is selective for cancer cells; in clonogenic assays, (S,S)-CpR 2TiCl2 was twice as effective at inhibiting colony formation than other stereoisomers after 24 h exposure. HPLC, MS and NMR techniques determined hydrolysis of CpR 2TiCl2; data strongly correlate with soluble [CpR 2Ti(OH (OH2)]+ being the biological trigger. Treatment of cells with CpR 2TiCl2 provoked extensive cytoplasmic vacuolization, endoplasmic reticulum (ER) swelling and activation of MAPKinase signal transduction, consistent with ligand-induced paraptosis, type III cell death, which is morphologically distinct from, and independent of apoptosis. Indeed, distinct from cisplatin, CpR 2TiCl2 failed to perturb cell cycle dynamics, induce γH2AX foci or evoke apoptosis in MDA-MB-468 and HCT-116 cells

    Using titanium complexes to defeat cancer: the view from the shoulders of titans

    When the first titanium complex with anticancer activity was identified in the 1970s, it was attractive, based on the presence of the dichloride unit in TiCl2Cp2 (Cp = η-C5H5)2, to assume its mode of biological action was closely aligned with cisplatin [cis-PtCl2(NH3)2]. Over the intervening 40 years however a far more complicated picture has arisen indicating multiple cellular mechanisms of cellular action can be triggered by titanium anti-cancer agents. This tutorial review aims to unpick the historical data and provide new researchers, without an explicit cancer biology background, a contemporary interpretation of both older and newer literature and to review the best techniques for attaining the identities of the biologically active titanium species and how these interact with the cancer cellular machinery
