Etest® versus broth microdilution for ceftaroline MIC determination with Staphylococcus aureus: results from PREMIUM, a European multicentre study
Objectives: To compare the concordance of ceftaroline MIC values 24 by reference broth microdilution (BMD) and Etest (BioMérieux, France) for MSSA and MRSA isolates, respectively, in isolates from PREMIUM (D372SL00001), a European multi-centre study. Methods: Ceftaroline MICs were determined by reference BMD and by Etest for 1,242 MSSA and MRSA from adult patients with community-acquired pneumonia or complicated skin and soft tissue infections collected between February and May 2012; tests were performed across six European laboratories. Selected isolates with ceftaroline resistance in broth (MIC >1 mg/L) were retested in three central laboratories to confirm their behaviour. Results: Overall concordance between BMD and Etest was good, with >97% essential agreement and >95% categorical agreement. Nevertheless, 12 of the 26 MRSA isolates found resistant by BMD scored as susceptible by Etest, with MICs ≤1 mg/L, thus counting as very major errors, whereas only five of 380 MRSA found ceftaroline susceptible in BMD were mis-categorised as resistant by Etest. Twenty-one of the 26 isolates with MICs of 2 mg/L by BMD were then re-tested twice by each of three central laboratories: BMD MICs of 2 mg/L were consistently found for 19 of the 21 isolates. Among 147 Etest results for these 21 isolates (original plus six repeats per isolate) 112 were >1 mg/L. Conclusions: BMD and Etest have good overall agreement for ceftaroline against Staphylococcus aureus; nevertheless, reliable Etest-based discrimination of the minority of ceftaroline-resistant (MIC 2 mg/L) MRSA is extremely challenging, requiring careful reading of strips, ideally with duplicate testing

Dust in Supernovae and Supernova Remnants I : Formation Scenarios
Supernovae are considered as prime sources of dust in space. Observations of local supernovae over the past couple of decades have detected the presence of dust in supernova ejecta. The reddening of the high redshift quasars also indicate the presence of large masses of dust in early galaxies. Considering the top heavy IMF in the early galaxies, supernovae are assumed to be the major contributor to these large amounts of dust. However, the composition and morphology of dust grains formed in a supernova ejecta is yet to be understood with clarity. Moreover, the dust masses inferred from observations in mid-infrared and submillimeter wavelength regimes differ by two orders of magnitude or more. Therefore, the mechanism responsible for the synthesis of molecules and dust in such environments plays a crucial role in studying the evolution of cosmic dust in galaxies. This review summarises our current knowledge of dust formation in supernova ejecta and tries to quantify the role of supernovae as dust producers in a galaxy.Peer reviewe
A thermal-driven graupel generation process to explain dry-season convective vigor over the Amazon
Large-eddy simulations (LESs) are conducted for each day of the intensive observation periods (IOPs) of the Green Ocean Amazon (GoAmazon) field campaign to characterize the updrafts and microphysics within deep convective cores while contrasting those properties between Amazon wet and dry seasons. Mean Doppler velocity (Vdop) values simulated using LESs are compared with 2-year measurements from a radar wind profiler (RWP) as viewed by statistical composites separated according to wet- and dry-season conditions. In the observed RWP and simulated LES Vdop composites, we find more intense low-level updraft velocity, vigorous graupel generation, and intense surface rain during the dry periods compared with the wet periods. To investigate coupled updraft–microphysical processes further, single-day golden cases are selected from the wet and dry periods to conduct detailed cumulus thermal tracking analysis. Tracking analysis reveals that simulated dry-season environments generate more droplet-loaded low-level thermals than wet-season environments. This tendency correlates with seasonal contrasts in buoyancy and vertical moisture advection profiles in large-scale forcing. Employing a normalized time series of mean thermal microphysics, the simulated cumulus thermals appear to be the primary generator of cloud droplets. When subsequent thermals penetrate the ice crystal layer, droplets within the thermals interact with entrained ice crystals, which enhances riming in the thermals. This appears to be a production pathway of graupel/hail particles within simulated deep convective cores. In addition, less-diluted dry-case thermals tend to be elevated higher, and graupel grows further during sedimentation after spilling out from thermals. Therefore, greater concentrations of low-level moist thermals likely result in more graupel/hail production and associated dry-season convective vigor.</p

Tracking the progressive spread of the SARS-CoV-2 Omicron variant in Italy, December 2021 to January 2022
Background: The SARS-CoV-2 variant of concern Omicron was first detected in Italy in November 2021. Aim: To comprehensively describe Omicron spread in Italy in the 2 subsequent months and its impact on the overall SARS-CoV-2 circulation at population level. Methods: We analyse data from four genomic surveys conducted across the country between December 2021 and January 2022. Combining genomic sequencing results with epidemiological records collated by the National Integrated Surveillance System, the Omicron reproductive number and exponential growth rate are estimated, as well as SARS-CoV-2 transmissibility. Results: Omicron became dominant in Italy less than 1 month after its first detection, representing on 3 January 76.9-80.2% of notified SARS-CoV-2 infections, with a doubling time of 2.7-3.3 days. As of 17 January 2022, Delta variant represented < 6% of cases. During the Omicron expansion in December 2021, the esti-mated mean net reproduction numbers respectively rose from 1.15 to a maximum of 1.83 for symptomatic cases and from 1.14 to 1.36 for hospitalised cases, while remaining relatively stable, between 0.93 and 1.21, for cases needing intensive care. Despite a reduc-tion in relative proportion, Delta infections increased in absolute terms throughout December contributing to an increase in hospitalisations. A significant repro-duction numbers' decline was found after mid-January, with average estimates dropping below 1 between 10 and 16 January 2022. Conclusion: Estimates suggest a marked growth advantage of Omicron compared with Delta variant, but lower disease severity at popula-tion level possibly due to residual immunity against severe outcomes acquired from vaccination and prior infection
