    A thermostable salmonella phage endolysin, Lys68, with broad bactericidal properties against gram-negative pathogens in presence of weak acids

    Resistance rates are increasing among several problematic Gram-negative pathogens, a fact that has encouraged the development of new antimicrobial agents. This paper characterizes a Salmonella phage endolysin (Lys68) and demonstrates its potential antimicrobial effectiveness when combined with organic acids towards Gram-negative pathogens. Biochemical characterization reveals that Lys68 is more active at pH 7.0, maintaining 76.7% of its activity when stored at 4°C for two months. Thermostability tests showed that Lys68 is only completely inactivated upon exposure to 100°C for 30 min, and circular dichroism analysis demonstrated the ability to refold into its original conformation upon thermal denaturation. It was shown that Lys68 is able to lyse a wide panel of Gram-negative bacteria (13 different species) in combination with the outer membrane permeabilizers EDTA, citric and malic acid. While the EDTA/Lys68 combination only inactivated Pseudomonas strains, the use of citric or malic acid broadened Lys68 antibacterial effect to other Gram-negative pathogens (lytic activity against 9 and 11 species, respectively). Particularly against Salmonella Typhimurium LT2, the combinatory effect of malic or citric acid with Lys68 led to approximately 3 to 5 log reductions in bacterial load/CFUs after 2 hours, respectively, and was also able to reduce stationary-phase cells and bacterial biofilms by approximately 1 log. The broad killing capacity of malic/citric acid-Lys68 is explained by the destabilization and major disruptions of the cell outer membrane integrity due to the acidity caused by the organic acids and a relatively high muralytic activity of Lys68 at low pH.     Exploring Pompeii: discovering hospitality through research synergy

    Hospitality research continues to broaden through an ever-increasing dialogue and alignment with a greater number of academic disciplines. This paper demonstrates how an enhanced understanding of hospitality can be achieved through synergy between archaeology, the classics and sociology. It focuses on classical Roman life, in particular Pompeii, to illustrate the potential for research synergy and collaboration, to advance the debate on hospitality research and to encourage divergence in research approaches. It demonstrates evidence of commercial hospitality activities through the excavation hotels, bars and taverns, restaurants and fast food sites. The paper also provides an example of the benefits to be gained from multidisciplinary analysis of hospitality and tourism

    Characterization and genome sequencing of a Citrobacter freundii phage CfP1 harboring a lysin active against multidrug-resistant isolates

    Citrobacter spp., although frequently ignored, is emerging as an important nosocomial bacterium able to cause various superficial and systemic life-threatening infections. Considered to be hard-to-treat bacterium due to its pattern of high antibiotic resistance, it is important to develop effective measures for early and efficient therapy. In this study, the first myovirus (vB_CfrM_CfP1) lytic for Citrobacter freundii was microbiologically and genomically characterized. Its morphology, activity spectrum, burst size, and biophysical stability spectrum were determined. CfP1 specifically infects C. freundii, has broad host range (>85 %; 21 strains tested), a burst size of 45 PFU/cell, and is very stable under different temperatures (20 to 50 °C) and pH (3 to 11) values. CfP1 demonstrated to be highly virulent against multidrug-resistant clinical isolates up to 12 antibiotics, including penicillins, cephalosporins, carbapenems, and fluroquinoles. Genomically, CfP1 has a dsDNA molecule with 180,219 bp with average GC content of 43.1 % and codes for 273 CDSs. The genome architecture is organized into function-specific gene clusters typical for tailed phages, sharing 46 to 94 % nucleotide identity to other Citrobacter phages. The lysin gene encoding a predicted D-Ala-D-Ala carboxypeptidase was also cloned and expressed in Escherichia coli and its activity evaluated in terms of pH, ionic strength, and temperature. The lysine optimum activity was reached at 20 mM HEPES, pH 7 at 37 °C, and was able to significantly reduce all C. freundii (>2 logs) as well as Citrobacter koseri (>4 logs) strains tested. Interestingly, the antimicrobial activity of this enzyme was performed without the need of pretreatment with outer membrane-destabilizing agents.     Research Themes, Authors, and Methodologies in the Journal of Applied Communications: A Ten-Year Overview

    The Journal of Applied Communications ( JAC) has been a primary outlet of agricultural communications research and professional scholarship—a claim validated by a survey of professionals in the field. The purpose of this study was to assess ten years of JAC to determine primary and secondary research themes, frequent primary and secondary research themes by year, prolific authorship, and research methods and types reported, using a mixed-methods design. Analyzed in the study were 91 research and/or professional articles with research methodologies published from 1997 through 2006. The research identified twenty-one primary research theme areas and 28 secondary research theme areas. A compiled list of primary and secondary research theme areas and frequent themes identified by year are reported. JAC authors were identified totaling 222 contributors; Tracy Irani and Ricky Telg (13.2%) were the most prolific authors. A majority of the articles (65.9%) employed quantitative research methods, and survey methodology (47.3%) was the most common data collection measure. Research themes appear cyclic, with specific themes moving in and out of primary and secondary areas, which may contribute to research theme diversity. Research must continue to determine whether cycles exist; if cycles do indeed exist then focus should be placed on determining cycle depth and the influence on research in agricultural communications as an integrated specialization area of agricultural education. This research should be used comparatively with priority areas identified in the National Research Agenda: Agricultural Education and Communication, 2007–2010, to determine where future research might be focused

    Acute black tea consumption improves cutaneous vascular function in healthy middle-aged humans.

    BACKGROUND & AIMS: Dietary flavonoids, such as those present in black tea, are associated with reduced risk of cardiovascular disease (CVD), possibly through improving nitric oxide (NO) mediated vascular function. The aim of this study was to examine the effect of acute black tea ingestion on cutaneous microvascular function. METHODS: Twenty healthy participants (58 ± 5 y, 9 men) attended two experimental trials (tea, placebo), 7-days apart in a randomised, controlled, double-blind, cross-over design. Participants ingested a single dose of 200 ml black tea or placebo, followed by assessment of forearm cutaneous perfusion using laser-Doppler flowmetry (LDF) using three distinct heating protocols, enabling us to distinguish between axon- and endothelium-dependent vasodilation: 1. rapid 42°C, 2. rapid 39°C and 3. gradual 42°C. On the contralateral arm, full-field laser perfusion imaging (FLPI) was used to assess forearm perfusion during gradual 42°C. Data were presented as cutaneous vascular conductance (CVC; flux/mean arterial pressure, MAP) and CVC expressed as a percentage of maximal CVC (%CVCmax). RESULTS: Rapid local heating to 39°C or 42°C demonstrated no effect of tea for flux, CVC or %CVCmax (all P > 0.05). Gradual local heating to 42 °C, however, produced a higher skin blood flow following black tea ingestion for absolute CVC (P = 0.04) when measured by LDF, and higher absolute flux (P < 0.001) and CVC (P < 0.001) measured with FLPI. No effect of tea was found for %CVCmax when assessed by either LDF or FLPI. CONCLUSIONS: Acute tea ingestion enhanced cutaneous vascular responses to gradual local heating to 42 °C in healthy, middle-aged participants, possibly through a mechanism related to activation of endothelium-derived chemical mediators, such as NO. These improvements may contribute to the cardiovascular health benefits of regular tea ingestion

    Time-reversed adapted-perturbation (TRAP) optical focusing onto dynamic objects inside scattering media

    The ability to steer and focus light inside scattering media has long been sought for a multitude of applications. At present, the only feasible strategy to form optical foci inside scattering media is to guide photons by using either implanted or virtual guide stars, which can be inconvenient and limits the potential applications. Here we report a scheme for focusing light inside scattering media by employing intrinsic dynamics as guide stars. By adaptively time-reversing the perturbed component of the scattered light, we show that it is possible to focus light to the origin of the perturbation. Using this approach, we demonstrate non-invasive dynamic light focusing onto moving targets and imaging of a time-variant object obscured by highly scattering media. Anticipated applications include imaging and photoablation of angiogenic vessels in tumours, as well as other biomedical uses

    Study on the performance of different craniofacial superimposition approaches (II): Best practices proposal

    Craniofacial superimposition, although existing for one century, is still a controversial technique within the scientific community. Objective and unbiased validation studies over a significant number of cases are required to establish a more solid picture on the reliability. However, there is lack of protocols and standards in the application of the technique leading to contradictory information concerning reliability. Instead of following a uniform methodology, every expert tends to apply his own approach to the problem, based on the available technology and deep knowledge on human craniofacial anatomy, soft tissues, and their relationships. The aim of this study was to assess the reliability of different craniofacial superimposition methodologies and the corresponding technical approaches to this type of identification. With all the data generated, some of the most representative experts in craniofacial identification joined in a discussion intended to identify and agree on the most important issues that have to be considered to properly employ the craniofacial superimposition technique. As a consequence, the consortium has produced the current manuscript, which can be considered the first standard in the field; including good and bad practices, sources of error and uncertainties, technological requirements and desirable features, and finally a common scale for the craniofacial matching evaluation. Such a document is intended to be part of a more complete framework for craniofacial superimposition, to be developed during the FP7-founded project MEPROCS, which will favour and standardize its proper application

    Relative abundance of a mesocarnivore in a human-dominated, semi-arid rangeland in Namibia

    Mesocarnivores fill important roles in ecological communities globally, but their distribution and abundance are often understudied. Many species have historically been regarded as vermin and subject to lethal control due to their role in livestock predation. Identifying the factors influencing mesocarnivore populations can help disentangle their relationship within ecological communities and inform conflict mitigation and conservation priorities. To help identify these factors, we used camera traps to study the community of medium and large mammals in four communal conservancies of northeastern Namibia covering the wet and dry seasons using 99 and 97 camera trap stations, respectively. We modelled black-backed jackal (Lupulella mesomelas) abundance using the robust Royle-Nichols model. Black-backed jackal were widespread, with a mean per site abundance of 2.01 (SD=0.66) in the wet season and 2.41 (SD=0.49) in the dry season. Black-backed jackal showed seasonally contrasting covariate associations, with lower abundance in areas with medium and large-sized wild prey during the wet season, and higher abundance in areas with more villages and close to African wild dog (Lycaon pictus) dens in the dry season. We identified localized hotspots of black-backed jackal abundance during the dry season, which may indicate that when resources are scarce, black-backed jackals rely on anthropogenic food sources despite an elevated risk for conflict, and on carcass remains from African wild dog kills. These findings highlight potential drivers of mesocarnivore abundance that would be obscured in a conventional occurrence modelling framework, and illustrate how local abundance may be influenced by seasonal variability, wild and anthropogenic food sources, and a likely facilitative relationship with a large carnivore. Further investigations in areas with more complex carnivore guilds and higher density of dominant predators are needed to understand black-backed jackal-African wild dog interactions and impacts on population dynamics

    Characterization of Modular Bacteriophage Endolysins from Myoviridae Phages OBP, 201ϕ2-1 and PVP-SE1

    Peptidoglycan lytic enzymes (endolysins) induce bacterial host cell lysis in the late phase of the lytic bacteriophage replication cycle. Endolysins OBPgp279 (from Pseudomonas fluorescens phage OBP), PVP-SE1gp146 (Salmonella enterica serovar Enteritidis phage PVP-SE1) and 201ϕ2-1gp229 (Pseudomonas chlororaphis phage 201ϕ2-1) all possess a modular structure with an N-terminal cell wall binding domain and a C-terminal catalytic domain, a unique property for endolysins with a Gram-negative background. All three modular endolysins showed strong muralytic activity on the peptidoglycan of a broad range of Gram-negative bacteria, partly due to the presence of the cell wall binding domain. In the case of PVP-SE1gp146, this domain shows a binding affinity for Salmonella peptidoglycan that falls within the range of typical cell adhesion molecules (Kaff = 1.26×106 M−1). Remarkably, PVP-SE1gp146 turns out to be thermoresistant up to temperatures of 90°C, making it a potential candidate as antibacterial component in hurdle technology for food preservation. OBPgp279, on the other hand, is suggested to intrinsically destabilize the outer membrane of Pseudomonas species, thereby gaining access to their peptidoglycan and exerts an antibacterial activity of 1 logarithmic unit reduction. Addition of 0.5 mM EDTA significantly increases the antibacterial activity of the three modular endolysins up to 2–3 logarithmic units reduction. This research work offers perspectives towards elucidation of the structural differences explaining the unique biochemical and antibacterial properties of OBPgp279, PVP-SE1gp146 and 201ϕ2-1gp229. Furthermore, these endolysins extensively enlarge the pool of potential antibacterial compounds used against multi-drug resistant Gram-negative bacterial infections
