    'Chardonel' resulted from the cross, 'SeyvaT x 'Chardonnay,' made in 1953. Fruit were first observed in 1958, and the original vine was propagated in 1960 under the number NY 45010.     Functional Divergence in the Genus Oenococcus as Predicted by Genome Sequencing of the Newly-Described Species, Oenococcus kitaharae

    Oenococcus kitaharae is only the second member of the genus Oenococcus to be identified and is the closest relative of the industrially important wine bacterium Oenococcus oeni. To provide insight into this new species, the genome of the type strain of O. kitaharae, DSM 17330, was sequenced. Comparison of the sequenced genomes of both species show that the genome of O. kitaharae DSM 17330 contains many genes with predicted functions in cellular defence (bacteriocins, antimicrobials, restriction-modification systems and a CRISPR locus) which are lacking in O. oeni. The two genomes also appear to differentially encode several metabolic pathways associated with amino acid biosynthesis and carbohydrate utilization and which have direct phenotypic consequences. This would indicate that the two species have evolved different survival techniques to suit their particular environmental niches. O. oeni has adapted to survive in the harsh, but predictable, environment of wine that provides very few competitive species. However O. kitaharae appears to have adapted to a growth environment in which biological competition provides a significant selective pressure by accumulating biological defence molecules, such as bacteriocins and restriction-modification systems, throughout its genome

    Autogene cevumeran with or without atezolizumab in advanced solid tumors: a phase 1 trial

    Effective targeting of somatic cancer mutations to enhance the efficacy of cancer immunotherapy requires an individualized approach. Autogene cevumeran is a uridine messenger RNA lipoplex-based individualized neoantigen-specific immunotherapy designed from tumor-specific somatic mutation data obtained from tumor tissue of each individual patient to stimulate T cell responses against up to 20 neoantigens. This ongoing phase 1 study evaluated autogene cevumeran as monotherapy (n = 30) and in combination with atezolizumab (n = 183) in pretreated patients with advanced solid tumors. The primary objective was safety and tolerability; exploratory objectives included evaluation of pharmacokinetics, pharmacodynamics, preliminary antitumor activity and immunogenicity. Non-prespecified interim analysis showed that autogene cevumeran was well tolerated and elicited poly-epitopic neoantigen-specific responses, encompassing CD4 + and/or CD8 + T cells, in 71% of patients, most of them undetectable at baseline. Responses were detectable up to 23 months after treatment initiation. CD8 + T cells specific for several neoantigens constituted a median of 7.3% of circulating CD8 + T cells, reaching up to 23% in some patients. Autogene cevumeran-induced T cells were found within tumor lesions constituting up to 7.2% of tumor-infiltrating T cells. Clinical activity was observed, including one objective response in monotherapy dose escalation and in two patients with disease characteristics unfavorable for response to immunotherapy treated in combination with atezolizumab. These findings support the continued development of autogene cevumeran in earlier treatment lines. ClinicalTrials.gov registration: NCT03289962

    Chemiosmotic energy from malolactic fermentation.

    By using the luciferase-luciferin ATP assay and whole cells of Leuconostoc oenos, we have demonstrated that malolactic fermentation does yield ATP. This energy-yielding mechanism did not occur in a cell extract and was inhibited in the presence of dicyclohexylcarbodiimide or an ionophore such as monensin. A lactate:proton efflux mechanism for this proposed pathway is presented

    Evaluation of Malolactic Bacteria Isolated from Oregon Wines

    Oregon is a cool wine-producing region where grapes characteristically contain high concentrations of organic acids. To reduce the natural acidity and increase the microbiological stability and flavor complexity of the wine, malolactic fermentation is encouraged. In this study, strains of Leuconostoc oenos indigenous to Oregon wines were evaluated for their suitability to conduct malolactic fermentation in Oregon wines. Tests determined the malolactic activity of the Oregon isolates in comparison with commercial strains ML-34, PSU-1, MLT-kli, and ens 44-40 under various temperature and pH conditions. Sensitivities to sulfur dioxide, ethanol, and fumaric acid also were determined. Two Oregon strains, Er-1a and Ey-2d, were selected for commercial winemaking tests because they had greater malolactic activity under conditions of low pH (3.0) and low temperature (15 and 8°C), respectively. </jats:p

    Os tricomas são importantes estruturas ligadas à resistência de plantas a insetos. Na cultura da mandioca alguns trabalhos mostram que maior número de tricomas influência negativamente a viabilidade de mosca branca. Porém, há uma carência na descrição dos tipos de tricomas observados nas folhas de mandioca. Com isso, o presente trabalho teve como objetivo quantificar e caracterizar os tricomas observados em diferentes variedades de mandioca. Para isso, foram coletadas folhas de três genótipos (2011-02-12, 2011-02-43 e Santa Helena), das quais foi destacado para análise o folíolo central, dividido em três regiões: basal, mediana, apical. Após a dissociação da epiderme foi realizada contagem e medição dos tricomas de cinco campos de 1,37mm2 de cada região. Os tricomas que foram observados nas diferentes variedades de mandioca são do tipo tectores simples. Não foi observada diferença estatística entre as regiões analisadas na mesma variedade, porém, há diferença conforme a variedade avaliada. Foi observado 41,8; 1,5 e 0,2 tricomas na região mediana, com média de 415,4; 357,5 e 245,4?m de comprimento para 2011-02-12, Santa Helena e 2011-02-43, respectivamente.
