Can individual and social patterns of resource use buffer animal populations against resource decline?
Species in many ecosystems are facing declines of key resources. If we are to understand and predict the effects of resource loss on natural populations, we need to understand whether and how the way animals use resources changes under resource decline. We investigated how the abundance of arboreal marsupials varies in response to a critical resource, hollow-bearing trees. Principally, we asked what mechanisms mediate the relationship between resources and abundance? Do animals use a greater or smaller proportion of the remaining resource, and is there a change in cooperative resource use (den sharing), as the availability of hollow trees declines? Analyses of data from 160 sites surveyed from 1997 to 2007 showed that hollow tree availability was positively associated with abundance of the mountain brushtail possum, the agile antechinus and the greater glider. The abundance of Leadbeater's possum was primarily influenced by forest age. Notably, the relationship between abundance and hollow tree availability was significantly less than 1:1 for all species. This was due primarily to a significant increase by all species in the proportional use of hollow-bearing trees where the abundance of this resource was low. The resource-sharing response was weaker and inconsistent among species. Two species, the mountain brushtail possum and the agile antechinus, showed significant but contrasting relationships between the number of animals per occupied tree and hollow tree abundance. The discrepancies between the species can be explained partly by differences in several aspects of the species' biology, including body size, types of hollows used and social behaviour as it relates to hollow use. Patient-powered research networks: building capacity for conducting patient-centered clinical outcomes research.
The Patient-Centered Outcomes Research Institute (PCORI) recently launched PCORnet to establish a single inter-operable multicenter data research network that will support observational research and randomized clinical trials. This paper provides an overview of the patient-powered research networks (PPRNs), networks of patient organizations focused on a particular health condition that are interested in sharing health information and engaging in research. PPRNs will build on their foundation of trust within the patient communities and draw on their expertise, working with participants to identify true patient-centered outcomes and direct a patient-centered research agenda. The PPRNs will overcome common challenges including enrolling a diverse and representative patient population; engaging patients in governance; designing the data infrastructure; sharing data securely while protecting privacy; prioritizing research questions; scaling small networks into a larger network; and identifying pathways to sustainability. PCORnet will be the first distributed research network to bring PCOR to national scale
Reciprocal regulation of the basic helix-loop-helix/Per-Arnt-Sim partner proteins, Arnt and Arnt2, during neuronal differentiation
Basic helix–loop–helix/Per–Arnt–Sim (bHLH/PAS) transcription factors function broadly in development, homeostasis and stress response. Active bHLH/PAS heterodimers consist of a ubiquitous signal-regulated subunit (e.g., hypoxia-inducible factors, HIF-1α/2α/3α; the aryl hydrocarbon receptor, AhR) or tissue-restricted subunit (e.g., NPAS1/3/4, Single Minded 1/2), paired with a general partner protein, aryl hydrocarbon receptor nuclear translocator (Arnt or Arnt2). We have investigated regulation of the neuron-enriched Arnt paralogue, Arnt2. We find high Arnt/Arnt2 ratios in P19 embryonic carcinoma cells and ES cells are dramatically reversed to high Arnt2/Arnt on neuronal differentiation. mRNA half-lives of Arnt and Arnt2 remain similar in both parent and neuronal differentiated cells. The GC-rich Arnt2 promoter, while heavily methylated in Arnt only expressing hepatoma cells, is methylation free in P19 and ES cells, where it is bivalent with respect to active H3K4me3 and repressive H3K27me3 histone marks. Typical of a ‘transcription poised’ developmental gene, H3K27me3 repressive marks are removed from Arnt2 during neuronal differentiation. Our data are consistent with a switch to predominant Arnt2 expression in neurons to allow specific functions of neuronal bHLH/PAS factors and/or to avoid neuronal bHLH/PAS factors from interfering with AhR/Arnt signalling.Nan Hao, Veronica L. SIRT1 Negatively Regulates the Mammalian Target of Rapamycin
The IGF/mTOR pathway, which is modulated by nutrients, growth factors, energy status and cellular stress regulates aging in various organisms. SIRT1 is a NAD+ dependent deacetylase that is known to regulate caloric restriction mediated longevity in model organisms, and has also been linked to the insulin/IGF signaling pathway. Here we investigated the potential regulation of mTOR signaling by SIRT1 in response to nutrients and cellular stress. We demonstrate that SIRT1 deficiency results in elevated mTOR signaling, which is not abolished by stress conditions. The SIRT1 activator resveratrol reduces, whereas SIRT1 inhibitor nicotinamide enhances mTOR activity in a SIRT1 dependent manner. Furthermore, we demonstrate that SIRT1 interacts with TSC2, a component of the mTOR inhibitory-complex upstream to mTORC1, and regulates mTOR signaling in a TSC2 dependent manner. These results demonstrate that SIRT1 negatively regulates mTOR signaling potentially through the TSC1/2 complex
Improving water and hand-washing services in rural health care facilities in Kitui County, Kenya
This study draws upon a diagnostic survey of WASH in health care facilities in Kitui County, Kenya, in 2019, and a pilot intervention providing professional delivery of water and soap services to 11 rural dispensaries in 2020. Three objectives guide the work. First, to estimate the annualised cost of providing safe water and soap in dispensaries. Second, to understand how these costs vary by location. Third, to chart the relative difference in costs of water from piped water schemes and vended water. The results estimate an annualised cost of water and soap services of around USD 1 per health care facility visit. Costs vary by remoteness and are higher for dispensaries relying on vended water provision rather than piped water schemes. The authors propose that county government include these cost estimates in future budget and investment planning, support professional service delivery models, and exploit synergies to manage water services to schools and communities
Digit Ratio Predicts Sense of Direction in Women
The relative length of the second-to-fourth digits (2D:4D) has been linked with prenatal androgen in humans. The 2D:4D is sexually dimorphic, with lower values in males than females, and appears to correlate with diverse measures of behavior. However, the relationship between digit ratio and cognition, and spatial cognition in particular, has produced mixed results. In the present study, we hypothesized that spatial tasks separating cue conditions that either favored female or male strategies would examine this structure-function correlation with greater precision. Previous work suggests that males are better in the use of directional cues than females. In the present study, participants learned a target location in a virtual landscape environment, in conditions that contained either all directional (i.e., distant or compass bearing) cues, or all positional (i.e., local, small objects) cues. After a short delay, participants navigated back to the target location from a novel starting location. Males had higher accuracy in initial search direction than females in environments with all directional cues. Lower digit ratio was correlated with higher accuracy of initial search direction in females in environments with all directional cues. Mental rotation scores did not correlate with digit ratio in either males or females. A multi-agent system framework for dialogue games in the group decision-making context
Dialogue games have been applied to various contexts in computer science and artificial intelligence, particularly to define interactions between autonomous software agents. However, in order to implement dialogue games, the developers need to deal with other important details besides what is presented in the model’s definition. This is a complex work, mostly when it is expected that the agents’ interactions correctly represent a human group behavior. In this work, we present a multi-agent system framework specifically designed to facilitate the implementation of dialogue games under the context of group decision-making in which agents interact as the humans do in face-to-face meetings. The proposed framework, named MAS4GDM, encapsulates the JADE framework and provides a layer that allows developers to easily implement their dialogue models without being concerned with some complex implementation details, such as: the communication model, the agents’ life cycle, among others. Diversity of Staphylococcus aureus Isolates in European Wildlife
Staphylococcus aureus is a well-known colonizer and cause of infection among
animals and it has been described from numerous domestic and wild animal
species. The aim of the present study was to investigate the molecular
epidemiology of S. aureus in a convenience sample of European wildlife and to
review what previously has been observed in the subject field. 124 S. aureus
isolates were collected from wildlife in Germany, Austria and Sweden; they
were characterized by DNA microarray hybridization and, for isolates with
novel hybridization patterns, by multilocus sequence typing (MLST). The
isolates were assigned to 29 clonal complexes and singleton sequence types
(CC1, CC5, CC6, CC7, CC8, CC9, CC12, CC15, CC22, CC25, CC30, CC49, CC59, CC88,
CC97, CC130, CC133, CC398, ST425, CC599, CC692, CC707, ST890, CC1956, ST2425,
CC2671, ST2691, CC2767 and ST2963), some of which (ST2425, ST2691, ST2963)
were not described previously. Resistance rates in wildlife strains were
rather low and mecA-MRSA isolates were rare (n = 6). mecC-MRSA (n = 8) were
identified from a fox, a fallow deer, hares and hedgehogs. The common cattle-
associated lineages CC479 and CC705 were not detected in wildlife in the
present study while, in contrast, a third common cattle lineage, CC97, was
found to be common among cervids. No Staphylococcus argenteus or
Staphylococcus schweitzeri-like isolates were found. Systematic studies are
required to monitor the possible transmission of human- and livestock-
associated S. aureus/MRSA to wildlife and vice versa as well as the possible
transmission, by unprotected contact to animals. The prevalence of S.
aureus/MRSA in wildlife as well as its population structures in different
wildlife host species warrants further investigation
