Sirtuins, bioageing, and cancer
The Sirtuins are a family of orthologues of yeast Sir2 found in a wide range of organisms from bacteria to man. They display a high degree of conservation between species, in both sequence and function, indicative of their key biochemical roles. Sirtuins are heavily implicated in cell cycle, cell division, transcription regulation, and metabolism, which places the various family members at critical junctures in cellular metabolism. Typically, Sirtuins have been implicated in the preservation of genomic stability and in the prolongation of lifespan though many of their target interactions remain unknown.
Sirtuins play key roles in tumourigenesis, as some have tumour-suppressor functions and others influence tumours through their control of the metabolic state of the cell. Their links to ageing have also highlighted involvement in various age-related and degenerative diseases. Here, we discuss the current understanding of the role of Sirtuins in age-related diseases while taking a closer look at their roles and functions in maintaining genomic stability and their influence on telomerase and telomere function
Atmospheric aerosol and Doppler lidar studies
Experimental and theoretical studies were performed of atmospheric aerosol backscatter and atmospheric dynamics with Doppler lidar as a primary tool. Activities include field and laboratory measurement and analysis efforts. The primary focus of activities related to understanding aerosol backscatter is the GLObal Backscatter Experiment (GLOBE) program. GLOBE is a multi-element effort designed toward developing a global aerosol model to describe tropospheric clean background backscatter conditions that Laser Atmospheric Wind Sounder (LAWS) is likely to encounter. Two survey missions were designed and flown in the NASA DC-8 in November 1989 and May to June 1990 over the remote Pacific Ocean, a region where backscatter values are low and where LAWS wind measurements could make a major contribution. The instrument complement consisted of pulsed and continuous-wave (CW) CO2 gas and solid state lidars measuring aerosol backscatter, optical particle counters measuring aerosol concentration, size distribution, and chemical composition, a filter/impactor system collecting aerosol samples for subsequent analysis, and integrating nephelometers measuring visible scattering coefficients. The GLOBE instrument package and survey missions were carefully planned to achieve complementary measurements under clean background backscatter conditions
Multi-center Airborne Coherent Atmospheric Wind Sensor (MACAWS)
This effort involves development of a calibrated, pulsed coherent CO2 Doppler lidar, followed by a carefully-planned and -executed program of multi-dimensional wind velocity and aerosol backscatter measurements from the NASA DC-8 research aircraft. The lidar, designated as the Multi-center Airborne Coherent Atmospheric Wind Sensor (MACAWS), will be applicable to two research areas. First, MACAWS will enable specialized measurements of atmospheric dynamical processes in the planetary boundary layer and free troposphere in geographic locations and over scales of motion not routinely or easily accessible to conventional sensors. The proposed observations will contribute fundamentally to a greater understanding of the role of the mesoscale, helping to improve predictive capabilities for mesoscale phenomena and to provide insights into improving model parameterizations of sub-grid scale processes within large-scale circulation models. As such, it has the potential to contribute uniquely to major, multi-institutional field programs planned for the mid 1990's. Second, MACAWS measurements can be used to reduce the degree of uncertainty in performance assessments and algorithm development for NASA's prospective Laser Atmospheric Wind Sounder (LAWS), which has no space-based instrument heritage. Ground-based lidar measurements alone are insufficient to address all of the key issues. To minimize costs, MACAWS is being developed cooperatively by the lidar remote sensing groups of the Jet Propulsion Laboratory, NOAA Wave Propagation Laboratory, and MSFC using existing lidar hardware and manpower resources. Several lidar components have already been exercised in previous airborne lidar programs (for example, MSFC Airborne Doppler Lidar System (ADLS) used in 1981,4 Severe Storms Wind Measurement Program; JPL Airborne Backscatter Lidar Experiment (ABLE) used in 1989,90 Global Backscatter Experiment Survey Missions). Analysis of airborne Doppler lidar, Doppler radar and tall tower measurements of atmospheric flows in quiescent and stormy weather
The first experiment to combine airborne Doppler Lidar and ground-based dual Doppler Radar measurements of wind to detail the lower tropospheric flows in quiescent and stormy weather was conducted in central Oklahoma during four days in June-July 1981. Data from these unique remote sensing instruments, coupled with data from conventional in-situ facilities, i.e., 500-m meteorological tower, rawinsonde, and surface based sensors, were analyzed to enhance understanding of wind, waves and turbulence. The purposes of the study were to: (1) compare winds mapped by ground-based dual Doppler radars, airborne Doppler lidar, and anemometers on a tower; (2) compare measured atmospheric boundary layer flow with flows predicted by theoretical models; (3) investigate the kinematic structure of air mass boundaries that precede the development of severe storms; and (4) study the kinematic structure of thunderstorm phenomena (downdrafts, gust fronts, etc.) that produce wind shear and turbulence hazardous to aircraft operations. Orbitofrontal cortex mediates pain inhibition by monetary reward
Pleasurable stimuli, including reward, inhibit pain, but the level of the neuraxis at which they do so and the cerebral
processes involved are unknown. Here, we characterized a brain circuitry mediating pain inhibition by reward. Twenty-four
healthy participants underwent functional magnetic resonance imaging while playing a wheel of fortune game with simultaneous thermal pain stimuli and monetary wins or losses. As expected, winning decreased pain perception compared to
losing. Inter-individual differences in pain modulation by monetary wins relative to losses correlated with activation in the
medial orbitofrontal cortex (mOFC). When pain and reward occured simultaneously, mOFCs functional connectivity
changed: the signal time course in the mOFC condition-dependent correlated negatively with the signal time courses in the
rostral anterior insula, anterior-dorsal cingulate cortex and primary somatosensory cortex, which might signify momentto-moment down-regulation of these regions by the mOFC. Monetary wins and losses did not change the magnitude of
pain-related activation, including in regions that code perceived pain intensity when nociceptive input varies and/or receive
direct nociceptive input. Pain inhibition by reward appears to involve brain regions not typically involved in nociceptive intensity coding but likely mediate changes in the significance and/or value of pain
GOES-R Geostationary Lightning Mapper Performance Specifications and Algorithms
The Geostationary Lightning Mapper (GLM) is a single channel, near-IR imager/optical transient event detector, used to detect, locate and measure total lightning activity over the full-disk. The next generation NOAA Geostationary Operational Environmental Satellite (GOES-R) series will carry a GLM that will provide continuous day and night observations of lightning. The mission objectives for the GLM are to: (1) Provide continuous, full-disk lightning measurements for storm warning and nowcasting, (2) Provide early warning of tornadic activity, and (2) Accumulate a long-term database to track decadal changes of lightning. The GLM owes its heritage to the NASA Lightning Imaging Sensor (1997- present) and the Optical Transient Detector (1995-2000), which were developed for the Earth Observing System and have produced a combined 13 year data record of global lightning activity. GOES-R Risk Reduction Team and Algorithm Working Group Lightning Applications Team have begun to develop the Level 2 algorithms and applications. The science data will consist of lightning "events", "groups", and "flashes". The algorithm is being designed to be an efficient user of the computational resources. This may include parallelization of the code and the concept of sub-dividing the GLM FOV into regions to be processed in parallel. Proxy total lightning data from the NASA Lightning Imaging Sensor on the Tropical Rainfall Measuring Mission (TRMM) satellite and regional test beds (e.g., Lightning Mapping Arrays in North Alabama, Oklahoma, Central Florida, and the Washington DC Metropolitan area) are being used to develop the prelaunch algorithms and applications, and also improve our knowledge of thunderstorm initiation and evolution
Protocols for the field testing
The COMMON SENSE project has been designed and planned in order to meet the general and specific scientific and technical objectives mentioned in its Description of Work (page 77).
In an overall strategy of the work plan, work packages (11) can be grouped into 3 key phases: (1) RD basis for cost-effective sensor development, (2) Sensor development, sensor web platform and integration, and (3) Field testing. In the first two phases WP1 and WP2 partners have provided a general understanding and integrated basis for a cost effective sensors development. Within the following WPs 4 to 8 the new sensors are created and integrated into different identified platforms. During the third phase 3, characterized by WP9, partners will deploy precompetitive prototypes at chosen platforms (e.g. research vessels, oil platforms, buoys and submerged moorings, ocean racing yachts, drifting buoys). Starting from August 2015 (month 22; task 9.2), these platforms will allow the partnership to test the adaptability and performance of the in-situ sensors and verify if the transmission of data is properly made, correcting deviations.
In task 9.1 all stakeholders identified in WP2, and other relevant agents, have been contacted in order to close a coordinated agenda for the field testing phase for each of the platforms. Field testing procedures (WP2) and deployment specificities, defined during sensor development in WPs 4 to 8, are closely studied by all stakeholders involved in field testing activities in order for everyone to know their role, how to proceed and to provide themselves with the necessary material and equipment (e.g. transport of instruments). All this information will provide the basis for designing and coordinating field testing activities.
Type and characteristics of the system (vessel or mooring, surface or deep, open sea or coastal area, duration, etc.), used for the field testing activities, are planned comprising the indicators included in the above-mentioned descriptors, taking into account that they must of interest for eutrophication, concentration of contaminants, marine litter and underwater noise.
In order to obtain the necessary information, two tables were realized starting from the information acquired for D2.2 delivered in June 2014. One table was created for sensor developers and one for those partners that will test the sensors at sea.
The six developers in COMMON SENSE have provided information on the seven sensors: CEFAS and IOPAN for underwater noise; IDRONAUT and LEITAT for microplastics; CSIC for an innovative piro and piezo resistive polymeric temperature and pressure and for heavy metal; DCU for the eutrophication sensor.
This information is anyway incomplete because in most cases the novel sensors are still far to be ready and will be developed over the course of COMMON SENSE. So the sensors cannot be clearly designed yet and, consequently, technical characteristics cannot still be perfectly defined. This produces some lag in the acquired information and, consequently, in the planning of their testing on specific platforms that will be solved in the near future.
In the table for Testers, partners have provided information on fifteen available platforms. Specific answers have been given on number and type of sensors on each platforms, their availability and technical characteristics, compatibility issues and, very important when new sensors are tested, comparative measurements to be implemented to verify them.
Finally IOPAN has described two more platforms, a motorboat not listed in the DoW, but already introduced in D2.2, and their oceanographic buoy in the Gdansk Bay that was previously unavailable. The same availability now is present for the OBSEA Underwater observatory from CSIC, while their Aqualog undulating mooring is still not ready for use.
In the following months, new information on sensors and platforms will be provided and the planning of testing activities will improve. Further updates of this report will be therefore necessary in order to individuate the most suitable platforms to test each kind of sensor.

Subsequently, the available new sensors have been tested since August 2015 till mid-October of the current year (2016) as part of task 9.2, following the indications defined in D9.1, such as the intercomparison of the new sensors with commercial ones, when possible.
The availability of new sensors was quite different in time starting with the first tests in September and October 2015 on noise, nutrient and heavy metals sensors and closing with pCO2 in late September 2016.
Sensors are technically fully described in the deliverables of WPs 3 to 8 and are here just mentioned where necessary. For further details, please consider those reports.
Objectives and rationale
The protocols prepared in D9.1 have been verified during the field testing activities of the innovative sensors on platforms. These can be summarized into 3 categories: (1) Research vessels (regular cruises); (2) Fixed platforms; (3) Ocean racing yachts. An exhaustive analysis of the different data obtained during field testing activities has been carried on in order to set possible optimization actions for prototypes design and performances. The data from each platform have been analyzed to verify limits and optimal installations or possible improvements. Finally a set of possible optimization actions has been defined. Data and observations collected during the course of field testing have been used to iteratively optimize the design and performance of the precompetitive prototypes
Analysis of relevant technical issues and deficiencies of the existing sensors and related initiatives currently set and working in marine environment. New generation technologies for cost-effective sensors
The last decade has seen significant growth in the field of sensor networks, which are currently collecting large amounts of environmental data. This data needs to be collected, processed, stored and made available for analysis and interpretation in a manner which is meaningful and accessible to end users and stakeholders with a range of requirements, including government agencies, environmental agencies, the research community, industry users and the public.
The COMMONSENSE project aims to develop and provide cost-effective, multi-functional innovative sensors to perform reliable in-situ measurements in the marine environment. The sensors will be easily usable across several platforms, and will focus on key parameters including eutrophication, heavy metal contaminants, marine litter (microplastics) and underwater noise descriptors of the MSFD.
The aims of Tasks 2.1 and 2.2 which comprise the work of this deliverable are:
• To obtain a comprehensive understanding and an up-to-date state of the art of existing sensors.
• To provide a working basis on “new generation” technologies in order to develop cost-effective sensors suitable for large-scale production.
This deliverable will consist of an analysis of state-of-the-art solutions for the different sensors and data platforms related with COMMONSENSE project. An analysis of relevant technical issues and deficiencies of existing sensors and related initiatives currently set and working in marine environment will be performed. Existing solutions will be studied to determine the main limitations to be considered during novel sensor developments in further WP’s.

The objectives of deliverable 2.1 are:
