"I am your mother and your father!": In vitro derived gametes and the ethics of solo reproduction
In this paper, we will discuss the prospect of human reproduction achieved with gametes originating from only one person. According to statements by a minority of scientists working on the generation of gametes in vitro, it may become possible to create eggs from men’s non-reproductive cells and sperm from women’s. This would enable, at least in principle, the creation of an embryo from cells obtained from only one individual: ‘solo reproduction’. We will consider what might motivate people to reproduce in this way, and the implications that solo reproduction might have for ethics and policy. We suggest that such an innovation is unlikely to revolutionise reproduction and parenting. Indeed, in some respects it is less revolutionary than in vitro fertilisation as a whole. Furthermore, we show that solo reproduction with in vitro created gametes is not necessarily any more ethically problematic than gamete donation—and probably less so. Where appropriate, we draw parallels with the debate surrounding reproductive cloning. We note that solo reproduction may serve to perpetuate reductive geneticised accounts of reproduction, and that this may indeed be ethically questionable. However, in this it is not unique among other technologies of assisted reproduction, many of which focus on genetic transmission. It is for this reason that a ban on solo reproduction might be inconsistent with continuing to permit other kinds of reproduction that also bear the potential to strengthen attachment to a geneticised account of reproduction. Our claim is that there are at least as good reasons to pursue research towards enabling solo reproduction, and eventually to introduce solo reproduction as an option for fertility treatment, as there are to do so for other infertility related purposes
Novel transparent nanocomposite films based on chitosan and bacterial cellulose
New nanocomposite films based on different chitosan matrices (two chitosans with different DPs and one water soluble derivative) and bacterial cellulose were prepared by a fully green procedure by casting a water based suspension of chitosan and bacterial cellulose nanofibrils. The films were characterized by several techniques, namely SEM, AFM, X-ray diffraction, TGA, tensile assays and visible spectroscopy. They were highly transparent, flexible and displayed better mechanical properties than the corresponding unfilled chitosan films. These new renewable nanocomposite materials also presented reasonable thermal stability and low O(2) permeability.FCT - SFRH/BD/41388/ 2007FCT - SFRH/BPD/38515/200
Comparative gene expression profiling of ADAMs, MMPs, TIMPs, EMMPRIN, EGF-R and VEGFA in low grade meningioma
MMPs (matrix metalloproteinases), ADAMs (a disintegrin and metalloproteinase) and TIMPs (tissue inhibitors of metalloproteinases) are implicated in invasion and angiogenesis: both are tissue remodeling processes involving regulated proteolysis of the extracellular matrix, growth factors and their receptors. The expression of these three groups and their correlations with clinical behaviour has been reported in gliomas but a similar comprehensive study in meningiomas is lacking. In the present study, we aimed to evaluate the patterns of expression of 23 MMPs, 4 TIMPs, 8 ADAMs, selective growth factors and their receptors in 17 benign meningiomas using a quantitative real-time polymerase chain reaction (qPCR). Results indicated very high gene expression of 13 proteases, inhibitors and growth factors studied: MMP2 and MMP14, TIMP-1, -2 and -3, ADAM9, 10, 12, 15 and 17, EGF-R, EMMPRIN and VEGF-A, in almost every meningioma.
Expression pattern analysis showed several positive correlations between MMPs, ADAMs, TIMPs and growth factors. Furthermore, our findings suggest that expression of MMP14, ADAM9, 10, 12, 15 and 17, TIMP-2, EGF-R and EMMPRIN reflects histological subtype of meningioma such that fibroblastic subtype had the highest mRNA expression, transitional subtype was intermediate and meningothelial type had the lowest expression. In conclusion, this is the first comprehensive study characterizing gene expression of ADAMs in meningiomas. These neoplasms, although by histological definition benign, have invasive potential. Taken together, the selected elevated gene expression pattern may serve to identify targets for therapeutic intervention or indicators of biological progression and recurrence
Thermal and mechanical properties of chitosan nanocomposites with cellulose modified in ionic liquids
In this paper, ionic liquid treatment was applied to produce nanometric cellulose particles of two polymorphic forms. A complex characterization of nanofillers including wide-angle X-ray scattering, Fourier transform infrared spectroscopy, and particle size determination was performed. The evaluated ionic liquid treatment was effective in terms of nanocrystalline cellulose production, leaving chemical and supermolecular structure of the materials intact. However, nanocrystalline cellulose II was found to be more prone to ionic liquid hydrolysis leading to formation larger amount of small particles. Each nanocrystalline cellulose was subsequently mixed with a solution of chitosan, so that composite films containing 1, 3, and 5% mass/mass of nanometric filler were obtained. Reference samples of chitosan and chitosan with micrometric celluloses were also solvent casted. Thermal, mechanical, and morphological properties of films were tested and correlated with properties of filler used. The results of both, tensile tests and thermogravimetric analysis showed a significant discrepancy between composites filled with nanocrystalline cellulose I and nanocrystalline cellulose II
Human-environment interaction during the Holocene along the shoreline of the Ancient Lake Ladoga: A case study based on palaeoecological and archaeological material from the Karelian Isthmus, Russia
This paper presents the results of pollen, diatom, charcoal, and sediment analyses from Lake Bol'shoye Zavetnoye, situated between the Gulf of Finland and Lake Ladoga on the Karelian Isthmus, north-western Russia. The main goal is to contribute to the discussion of Neolithic land use in north-eastern Europe. The article aims to answer questions related to Stone Age hunter-gatherer economy, ecology, and anthropogenic environmental impact through a comprehensive combination of multiple types of palaeoecological data and archaeological material. According to diatom data, Lake Bol'shoye Zavetnoye was influenced by the water level oscillations of Ancient Lake Ladoga during much of the Holocene. Intensified human activity and prolonged human occupation become visible in the Lake Bol'shoye Zavetnoye pollen data between 4480 BC and 3250 BC. During the final centuries of the Stone Age, a new phase of land use began, as several anthropogenic indicators, such asTriticum, Cannabis, andPlantago lanceolataappear in the pollen data and a decrease inPinusvalues is recorded. In general, the results indicate that socio-cultural transformations could have taken place already from the mid-5th millennium BC onwards, including new ways of utilizing the environment, perhaps also in the field of subsistence, even though the livelihood was based on foraging throughout the period
Adapting the Sustainable Development Goals and the New Urban Agenda to the city level : initial reflections from a comparative research project
The Sustainable Development Goals and the New Urban Agenda recognise the role of cities in achieving sustainable development. However, these agendas were agreed and signed by national governments and thus implementing them at the local level requires a process of adaptation or localisation. In this paper, we analyse five aspects that practitioners and researchers need to consider when localising them: (1) delimitation of the urban boundary; (2) integrated governance; (3) actors; (4) synergies and trade-offs and (5) indicators. These considerations are interrelated, and while not exhaustive, provide an important initial step for reflection on the challenges and opportunities of working with these global agendas at the local level. The paper draws on the inception phase of an international comparative transdisciplinary research project in seven cities on four continents: Buenos Aires (Argentina), Cape Town (South Africa), Gothenburg (Sweden), Kisumu (Kenya), Malmö (Sweden), Sheffield (UK) and Shimla (India)

Infrastructural requirements for local implementation of safety policies: the discordance between top-down and bottom-up systems of action
<p>Abstract</p> <p>Background</p> <p>Safety promotion is planned and practised not only by public health organizations, but also by other welfare state agencies, private companies and non-governmental organizations. The term 'infrastructure' originally denoted the underlying resources needed for warfare, e.g. roads, industries, and an industrial workforce. Today, 'infrastructure' refers to the physical elements, organizations and people needed to run projects in different societal arenas.</p> <p>The aim of this study was to examine associations between infrastructure and local implementation of safety policies in injury prevention and safety promotion programs.</p> <p>Methods</p> <p>Qualitative data on municipalities in Sweden designated as Safe Communities were collected from focus group interviews with municipal politicians and administrators, as well as from policy documents, and materials published on the Internet. Actor network theory was used to identify weaknesses in the present infrastructure and determine strategies that can be used to resolve these.</p> <p>Results</p> <p>The weakness identification analysis revealed that the factual infrastructure available for effectuating national strategies varied between safety areas and approaches, basically reflecting differences between bureaucratic and network-based organizational models. At the local level, a contradiction between safety promotion and the existence of quasi-markets for local public service providers was found to predispose for a poor local infrastructure diminishing the interest in integrated inter-agency activities. The weakness resolution analysis showed that development of an adequate infrastructure for safety promotion would require adjustment of the legal framework regulating injury data exchange, and would also require rational financial models for multi-party investments in local infrastructures.</p> <p>Conclusion</p> <p>We found that the "silo" structure of government organization and assignment of resources was a barrier to collaborative action for safety at a community level. It may therefore be overly optimistic to take for granted that different approaches to injury control, such as injury prevention and safety promotion, can share infrastructure. Similarly, it may be unrealistic to presuppose that safety promotion can reach its potential in terms of injury rate reductions unless the critical infrastructure for this is in place. Such an alignment of the infrastructure to organizational processes requires more than financial investments.</p
