    Fluorescent Calcium Imaging and Subsequent In Situ Hybridization for Neuronal Precursor Characterization in Xenopus laevis

    Spontaneous intracellular calcium activity can be observed in a variety of cell types and is proposed to play critical roles in a variety of physiological processes. In particular, appropriate regulation of calcium activity patterns during embryogenesis is necessary for many aspects of vertebrate neural development, including proper neural tube closure, synaptogenesis, and neurotransmitter phenotype specification. While the observation that calcium activity patterns can differ in both frequency and amplitude suggests a compelling mechanism by which these fluxes might transmit encoded signals to downstream effectors and regulate gene expression, existing population-level approaches have lacked the precision necessary to further explore this possibility. Furthermore, these approaches limit studies of the role of cell-cell interactions by precluding the ability to assay the state of neuronal determination in the absence of cell-cell contact. Therefore, we have established an experimental workflow that pairs time-lapse calcium imaging of dissociated neuronal explants with a fluorescence in situ hybridization assay, allowing the unambiguous correlation of calcium activity pattern with molecular phenotype on a single-cell level. We were successfully able to use this approach to distinguish and characterize specific calcium activity patterns associated with differentiating neural cells and neural progenitor cells, respectively; beyond this, however, the experimental framework described in this article could be readily adapted to investigate correlations between any time-series activity profile and expression of a gene or genes of interest

    Arp 65 interaction debris: massive HI displacement and star formation

    Context: Pre-merger interactions between galaxies can induce significant changes in the morphologies and kinematics of the stellar and ISM components. Large amounts of gas and stars are often found to be disturbed or displaced as tidal debris. This debris then evolves, sometimes forming stars and occasionally tidal dwarf galaxies. Here we present results from our HI study of Arp 65, an interacting pair hosting extended HI tidal debris. Aims: In an effort to understand the evolution of tidal debris produced by interacting pairs of galaxies, including in situ star and tidal dwarf galaxy formation, we are mapping HI in a sample of interacting galaxy pairs. The Arp 65 pair is one of them. Methods: Our resolved HI 21 cm line survey is being carried out using the Giant Metrewave Radio Telescope (GMRT). We used our HI survey data as well as available SDSS optical, Spitzer infra-red and GALEX UV data to study the evolution of the tidal debris and the correlation of HI with the star-forming regions within it. Results: In Arp 65 we see a high impact pre-merger interaction involving a pair of massive galaxies (NGC 90 and NGC 93) that have a stellar mass ratio of ~ 1:3. The interaction, which probably occurred ~ 1.0 -- 2.5 ×\times 108^8 yr ago, appears to have displaced a large fraction of the HI in NGC 90 (including the highest column density HI) beyond its optical disk. We also find extended ongoing star formation in the outer disk of NGC 90. In the major star-forming regions, we find the HI column densities to be ~ 4.7 ×\times 1020^{20} cm2^{-2} or lower. But no signature of star formation was found in the highest column density HI debris, SE of NGC 90. This indicates conditions within the highest column density HI debris remain hostile to star formation and it reaffirms that high HI column densities may be a necessary but not sufficient criterion for star formation.Comment: Accepted in A&

    Symptom Levels in Care-Seeking Bangladeshi and Nepalese Adults With Advanced Cancer

    Purpose Three-fourths of patients with advanced cancer are reported to suffer from pain. A primary barrier to provision of adequate symptom treatment is failure to appreciate the intensity of the symptoms patients are experiencing. Because data on Bangladeshi and Nepalese patients’ perceptions of their symptomatic status are limited, we sought such information using a cell phone questionnaire. Methods At tertiary care centers in Dhaka and Kathmandu, we recruited 640 and 383 adult patients, respectively, with incurable malignancy presenting for outpatient visits and instructed them for that single visit on one-time completion of a cell phone platform 15-item survey of questions about common cancer associated symptoms and their magnitudes using Likert scales of 0 to 10. The questions were taken from the Edmonton Symptom Assessment System and the Brief Pain Inventory instruments. Results All but two Bangladeshi patients recruited agreed to study participation. Two-thirds of Bangladeshi patients reported usual pain levels ≥ 5, and 50% of Nepalese patients reported usual pain levels ≤ 4 (population differences significant at P \u3c .001). Conclusion Bangladeshi and Nepalese adults with advanced cancer are comfortable with cell phone questionnaires about their symptoms and report high levels of pain. Greater attention to the suffering of these patients is warranted
