    Genomic Analysis of Drosophila Neuronal Remodeling: A Role for the RNA-Binding Protein Boule as a Negative Regulator of Axon Pruning

    Drosophila mushroom body (MB) {gamma} neurons undergo axon pruning during metamorphosis through a process of localized degeneration of specific axon branches. Developmental axon degeneration is initiated by the steroid hormone ecdysone, acting through a nuclear receptor complex composed of USP (ultraspiracle) and EcRB1 (ecdysone receptor B1) to regulate gene expression in MB {gamma} neurons. To identify ecdysone-dependent gene expression changes in MB {gamma} neurons at the onset of axon pruning, we use laser capture microdissection to isolate wild-type and mutant MB neurons in which EcR (ecdysone receptor) activity is genetically blocked, and analyze expression changes by microarray. We identify several molecular pathways that are regulated in MB neurons by ecdysone. The most striking observation is the upregulation of genes involved in the UPS (ubiquitin–proteasome system), which is cell autonomously required for {gamma} neuron pruning. In addition, we characterize the function of Boule, an evolutionarily conserved RNA-binding protein previously implicated in spermatogenesis in flies and vertebrates. boule expression is downregulated by ecdysone in MB neurons at the onset of pruning, and forced expression of Boule in MB {gamma} neurons is sufficient to inhibit axon pruning. This activity is dependent on the RNA-binding domain of Boule and a conserved DAZ (deleted in azoospermia) domain implicated in interactions with other RNA-binding proteins. However, loss of Boule does not result in obvious defects in axon pruning or morphogenesis of MB neurons, suggesting that it acts redundantly with other ecdyonse-regulated genes. We propose a novel function for Boule in the CNS as a negative regulator of developmental axon pruning

    Cross-cultural effects of color, but not morphological masculinity, on perceived attractiveness of men's faces

The role of masculinity (which for adults is thought to be a relatively stable shape cue to developmental testosterone levels) in male facial attractiveness has been examined, with mixed results. Recent work on the perception of skin color (a more variable cue to current health status) indicates that increased skin redness, yellowness, and lightness enhance apparent health. It has been suggested that stable cues such as masculinity may be less important to attractiveness judgments than short-term, more variable health cues. We examined associations between male facial attractiveness, masculinity, and skin color in African and Caucasian populations. Masculinity was not found to be associated with attractiveness in either ethnic group. However, skin color was found to be an important predictor of attractiveness judgments, particularly for own-ethnicity faces. Our results suggest that more plastic health cues, such as skin color, are more important than developmental cues such as masculinity. Further, unfamiliarity with natural skin color variation in other ethnic groups may limit observers' ability to utilize these color cues

    Dark halos acting as chaos controllers in asymmetric triaxial galaxy models

    We study the regular or chaotic character of orbits in a 3D dynamical model, describing a triaxial galaxy surrounded by a spherical dark halo component. Our numerical experiments suggest that the percentage of chaotic orbits decreases exponentially as the mass of the dark halo increases. A linear increase of the percentage of the chaotic orbits was observed as the scale length of the halo component increases. In order to distinguish between regular and chaotic motion, we chose to use the total angular momentum Ltot of the 3D orbits as a new indicator. Comparison with other, previously used, dynamical indicators, such as the Lyapunov Characteristic Exponent or the P(f) spectral method, shows that the Ltot indicator gives very fast and reliable results for characterizing the nature of orbits in galactic dynamical models.Comment: Published in Research in Astronomy and Astrophysics (RAA) journa

    Characterizing the Circumgalactic Medium of Nearby Galaxies with HST/COS and HST/STIS Absorption-Line Spectroscopy: II. Methods and Models

    We present basic data and modeling for a survey of the cool, photo-ionized Circum-Galactic Medium (CGM) of low-redshift galaxies using far-UV QSO absorption line probes. This survey consists of "targeted" and "serendipitous" CGM subsamples, originally described in Stocke et al. (2013, Paper 1). The targeted subsample probes low-luminosity, late-type galaxies at z<0.02z<0.02 with small impact parameters (ρ=71\langle\rho\rangle = 71 kpc), and the serendipitous subsample probes higher luminosity galaxies at z0.2z\lesssim0.2 with larger impact parameters (ρ=222\langle\rho\rangle = 222 kpc). HST and FUSE UV spectroscopy of the absorbers and basic data for the associated galaxies, derived from ground-based imaging and spectroscopy, are presented. We find broad agreement with the COS-Halos results, but our sample shows no evidence for changing ionization parameter or hydrogen density with distance from the CGM host galaxy, probably because the COS-Halos survey probes the CGM at smaller impact parameters. We find at least two passive galaxies with H I and metal-line absorption, confirming the intriguing COS-Halos result that galaxies sometimes have cool gas halos despite no on-going star formation. Using a new methodology for fitting H I absorption complexes, we confirm the CGM cool gas mass of Paper 1, but this value is significantly smaller than found by the COS-Halos survey. We trace much of this difference to the specific values of the low-zz meta-galactic ionization rate assumed. After accounting for this difference, a best-value for the CGM cool gas mass is found by combining the results of both surveys to obtain log(M/M)=10.5±0.3\log{(M/M_{\odot})}=10.5\pm0.3, or ~30% of the total baryon reservoir of an LLL \geq L^*, star-forming galaxy.Comment: 51 pages, 20 figures, 11 tables, accepted for publication in the Astrophysical Journal Supplement Serie

    Metal Abundances in the Magellanic Stream

    We report on the first metallicity determination for gas in the Magellanic Stream, using archival HST GHRS data for the background targets Fairall 9, III Zw 2, and NGC 7469. For Fairall 9, using two subsequent HST revisits and new Parkes Multibeam Narrowband observations, we have unequivocally detected the MSI HI component of the Stream (near its head) in SII1250,1253 yielding a metallicity of [SII/H]=-0.55+/-0.06(r)+/-0.2(s), consistent with either an SMC or LMC origin and with the earlier upper limit set by Lu et al. (1994). We also detect the saturated SiII1260 line, but set only a lower limit of [SiII/H]>-1.5. We present serendipitous detections of the Stream, seen in MgII2796,2803 absorption with column densities of (0.5-1)x10^13 cm^-2 toward the Seyfert galaxies III Zw 2 and NGC 7469. These latter sightlines probe gas near the tip of the Stream (80 deg down-Stream of Fairall 9). For III Zw 2, the lack of an accurate HI column density and the uncertain MgIII ionization correction limits the degree to which we can constrain [Mg/H]; a lower limit of [MgII/HI]>-1.3 was found. For NGC 7469, an accurate HI column density determination exists, but the extant FOS spectrum limits the quality of the MgII column density determination, and we conclude that [MgII/HI]>-1.5. Ionization corrections associated with MgIII and HII suggest that the corresponding [Mg/H] may range lower by 0.3-1.0 dex. However, an upward revision of 0.5-1.0 dex would be expected under the assumption that the Stream exhibits a dust depletion pattern similar to that seen in the Magellanic Clouds.     Prefrontal cortex stimulation does not affect emotional bias, but may slow emotion identification

    Transcranial direct current stimulation (tDCS) has recently garnered attention as a putative depression treatment. However, the cognitive mechanisms by which it exerts an antidepressant effect are unclear: tDCS may directly alter ‘hot’ emotional processing biases, or alleviate depression through changes in ‘cold’ (non-emotional) cognitive function. Here, 75 healthy participants performed a facial emotion identification task during 20 minutes of anodal or sham tDCS over the left dorsolateral prefrontal cortex (DLPFC) in a double-blind, within-subject crossover design. A subset of 31 participants additionally completed a task measuring attentional distraction during stimulation. Compared to sham stimulation, anodal tDCS of the left DLPFC resulted in an increase in response latency across all emotional conditions. Bayesian analysis showed definitively that tDCS exerted no emotion-dependent effect on behaviour. Thus, we demonstrate that anodal tDCS produces a general, rather than an emotion-specific, effect. We also report a preliminary finding in the subset of participants who completed the distractibility task: increased distractibility during active stimulation correlated significantly with the degree to which tDCS slowed emotion identification. Our results provide insight into the possible mechanisms by which DLPFC tDCS may treat symptoms of depression, suggesting that it may not alter emotional biases, but instead may affect ‘cold’ cognitive processes

    Tracing baryons in the warm-hot intergalactic medium with broad Ly alpha absorption

    We discuss physical properties and baryonic content of broad Ly alpha absorbers (BLAs) at low redshift. These absorption systems, recently discovered in high-resolution, high-signal to noise quasar absorption line spectra, possibly trace the warm-hot intergalactic medium (WHIM) in the temperature range between 10^5 and 10^6 K. To extend previous BLA measurements we have analyzed STIS data of the two quasars H 1821+643 and PG 0953+415 and have identified 13 BLA candidates along a total (unblocked) redshift path of dz=0.440. Combining our measurements with previous results for the lines of sight toward PG 1259+593 and PG 1116+215, the resulting new BLA sample consists of 20 reliably detected systems as well as 29 additional tentative cases, implying a BLA number density of dN/dz=22-53. We estimate that the contribution of BLAs to the baryon density at z=0 is Omega_b(BLA)>0.0027 h_70^-1 for absorbers with log (N/b)>11.3. This number indicates that WHIM broad Ly alpha absorbers contain a substantial fraction of the baryons in the local Universe.     Body odor quality predicts behavioral attractiveness in humans

    Growing effort is being made to understand how different attractive physical traits co-vary within individuals, partly because this might indicate an underlying index of genetic quality. In humans, attention has focused on potential markers of quality such as facial attractiveness, axillary odor quality, the second-to-fourth digit (2D:4D) ratio and body mass index (BMI). Here we extend this approach to include visually-assessed kinesic cues (nonverbal behavior linked to movement) which are statistically independent of structural physical traits. The utility of such kinesic cues in mate assessment is controversial, particularly during everyday conversational contexts, as they could be unreliable and susceptible to deception. However, we show here that the attractiveness of nonverbal behavior, in 20 male participants, is predicted by perceived quality of their axillary body odor. This finding indicates covariation between two desirable traits in different sensory modalities. Depending on two different rating contexts (either a simple attractiveness rating or a rating for long-term partners by 10 female raters not using hormonal contraception), we also found significant relationships between perceived attractiveness of nonverbal behavior and BMI, and between axillary odor ratings and 2D:4D ratio. Axillary odor pleasantness was the single attribute that consistently predicted attractiveness of nonverbal behavior. Our results demonstrate that nonverbal kinesic cues could reliably reveal mate quality, at least in males, and could corroborate and contribute to mate assessment based on other physical traits
