Putting hydrodynamic interactions to work: tagged particle separation
Separation of magnetically tagged cells is performed by attaching markers to
a subset of cells in suspension and applying fields to pull from them in a
variety of ways. The magnetic force is proportional to the field gradient, and
the hydrodynamic interactions play only a passive, adverse role. Here we
propose using a homogeneous rotating magnetic field only to make tagged
particles rotate, and then performing the actual separation by means of
hydrodynamic interactions, which thus play an active role. The method, which we
explore here theoretically and by means of numerical simulations, lends itself

Propagation of a Solitary Fission Wave
Reaction-diffusion phenomena are encountered in an astonishing array of natural systems. Under the right conditions, self stabilizing reaction waves can arise that will propagate at constant velocity. Numerical studies have shown that fission waves of this type are also possible and that they exhibit soliton like properties. Here, we derive the conditions required for a solitary fission wave to propagate at constant velocity. The results place strict conditions on the shapes of the flux, diffusive, and reactive profiles that would be required for such a phenomenon to persist, and this condition would apply to other reaction diffusion phenomena as well. Numerical simulations are used to confirm the results and show that solitary fission waves fall into a bistable class of reaction diffusion phenomena. (C) 2012 American Institute of Physics. Influence of boundaries on pattern selection in through-flow
The problem of pattern selection in absolutely unstable open flow systems is
investigated by considering the example of Rayleigh-B\'{e}nard convection. The
spatiotemporal structure of convection rolls propagating downstream in an
externally imposed flow is determined for six different inlet/outlet boundary
conditions. Results are obtained by numerical simulations of the Navier-Stokes
equations and by comparison with the corresponding Ginzburg-Landau amplitude
equation. A unique selection process is observed being a function of the
control parameters and the boundary conditions but independent of the history
and the system length. The problem can be formulated in terms of a nonlinear
eigen/boundary value problem where the frequency of the propagating pattern is

Pattern selection as a nonlinear eigenvalue problem
A unique pattern selection in the absolutely unstable regime of driven,
nonlinear, open-flow systems is reviewed. It has recently been found in
numerical simulations of propagating vortex structures occuring in
Taylor-Couette and Rayleigh-Benard systems subject to an externally imposed
through-flow. Unlike the stationary patterns in systems without through-flow
the spatiotemporal structures of propagating vortices are independent of
parameter history, initial conditions, and system length. They do, however,
depend on the boundary conditions in addition to the driving rate and the
through-flow rate. Our analysis of the Ginzburg-Landau amplitude equation
elucidates how the pattern selection can be described by a nonlinear eigenvalue
problem with the frequency being the eigenvalue. Approaching the border between
absolute and convective instability the eigenvalue problem becomes effectively
linear and the selection mechanism approaches that of linear front propagation.

A unique pattern selection in the absolutely unstable regime of a driven,
nonlinear, open-flow system is analyzed: The spatiotemporal structures of
rotationally symmetric vortices that propagate downstream in the annulus of the
rotating Taylor-Couette system due to an externally imposed axial through-flow
are investigated for two different axial boundary conditions at the in- and
outlet. Unlike the stationary patterns in systems without through-flow the
spatiotemporal structures of propagating vortices are independent of parameter
history, initial conditions, and system's length. They do, however, depend on
the axial boundary conditions, the driving rate of the inner cylinder and the
through-flow rate. Our analysis of the amplitude equation shows that the
pattern selection can be described by a nonlinear eigenvalue problem with the
frequency being the eigenvalue. Approaching the border between absolute and
convective instability the eigenvalue problem becomes effectively linear and
the selection mechanism approaches that one of linear front propagation.

Age-Specific ADME Gene Expression in Infant Intestinal Enteroids
In childhood, developmental changes and environmental interactions highly affect orally dosed drug disposition across the age range. To optimize dosing regimens and ensure safe use of drugs in pediatric patients, understanding this age-dependent biology is necessary. In this proof-of-concept study, we aimed to culture age-specific enteroids from infant tissue which represent its original donor material, specifically for drug transport and metabolism. Enteroid lines from fresh infant tissues ( n = 8, age range: 0.3-45 postnatal weeks) and adult tissues ( n = 3) were established and expanded to 3D self-organizing enteroids. The gene expression of drug transporters P-gp ( ABCB1), BCRP ( ABCG2), MRP2 ( ABCC2), and PEPT1 ( SLC15A1) and drug metabolizing enzymes CYP3A4, CYP2C18, and UGT1A1 was determined with RT-qPCR in fresh tissue and its derivative differentiated enteroids. Expression levels of P-gp, BCRP, MRP2, and CYP3A4 were similar between tissues and enteroids. PEPT1 and CYP2C18 expression was lower in enteroids compared to that in the tissue. The expression of UGT1A1 in the tissue was lower than that in enteroids. The gene expression did not change with the enteroid passage number for all genes studied. Similar maturational patterns in tissues and enteroids were visually observed for P-gp, PEPT1, MRP2, CYP3A4, CYP2C18, and VIL1. In this explorative study, interpatient variability was high, likely due to the diverse patient characteristics of the sampled population (e.g., disease, age, and treatment). To summarize, maturational patterns of clinically relevant ADME genes in tissue were maintained in enteroids. These findings are an important step toward the potential use of pediatric enteroids in pediatric drug development, which in the future may lead to improved pediatric safety predictions during drug development. We reason that such an approach can contribute to a potential age-specific platform to study and predict drug exposure and intestinal safety in pediatrics. </p

'OH MY GOD! BUY IT!' A multimodal discourse analysis of the discursive strategies used by Chinese ecommerce live-streamer Austin Li
Ecommerce livestreaming, also known as live commerce or social commerce, has taken off over the past two years in East Asia and is showing the tendency of going global. Intrigued by the phenomenal success of ecommerce livestream, we concentrate on analyzing the most prominent and illustrative ex- ample of Chinese ecommerce live-streamer Austin Li. Through this individual case study, we aim to investigate discursive strategies employed in ecommerce livestreaming and reveal resources specific to this new media genre. Guided by multimodal discourse analysis, our research first accommodates the socio-eco- nomic context of Li’s success to warrant social situatedness in interpreting data. After that we move into analyzing his discourse employed in livestreaming. Re- search findings suggest that in attention economy, Li strategically utilizes his male gender as a resource in trying on lipsticks for female customers. His dis- course in multiple modes serves to build consumer trust and propagate products. An in-depth analysis of his discursive strategies indicates that, ecommerce livestreaming as a new form of advertising not only shares commonalities with traditional advertisement discourse but also embodies affordances that are spe- cific to livestreaming platforms. To be more specific, livestreaming is featured with delimitation of time, real-time interactivity, and video-aided communica- tion. These affordances enable Li to adopt more interactive and personalized per- suasive discourse than conventional advertisement
