    Anti-phage islands force their target phage to directly mediate island excision and spread.

    Vibrio cholerae, the causative agent of the diarrheal disease cholera, is antagonized by the lytic phage ICP1 in the aquatic environment and in human hosts. Mobile genetic elements called PLEs (phage-inducible chromosomal island-like elements) protect V. cholerae from ICP1 infection and initiate their anti-phage response by excising from the chromosome. Here, we show that PLE 1 encodes a large serine recombinase, Int, that exploits an ICP1-specific protein as a recombination directionality factor (RDF) to excise PLE 1 in response to phage infection. We show that this phage-encoded protein is sufficient to direct Int-mediated recombination in vitro and that it is highly conserved in all sequenced ICP1 genomes. Our results uncover an aspect of the molecular specificity underlying the conflict between a single predatory phage and V. cholerae PLE and contribute to our understanding of long-term evolution between phage and their bacterial hosts

    Non-stationarity in daily and sub-daily intense rainfall – Part 1: Sydney, Australia

    This study was driven by a need to clarify how variations in climate might affect intense rainfall and the potential for flooding. Sub-daily durations are of particular interest for urban applications. Worldwide, few such observation-based studies exist, which is mainly due to limitations in data. While there are still large discrepancies between precipitation data sets from observations and models, both show that there is a tendency for moist regions to become wetter and for dry regions to become drier. However, changes in extreme conditions may show the opposite sign to those in average conditions. Where changes in observed intense precipitation have been studied, this has typically been for daily durations or longer. <br><br> The purpose of this two-part study is to examine daily and sub-daily rainfall extremes for evidence of non-stationarity. Here the problem was addressed by supplementing one long record (Part 1) by a set of shorter records for a 30-yr concurrent period (Part 2). Variations in frequency and magnitude of rainfall extremes across durations from 6 min to 72 h were assessed using data from sites in the south-east of Australia. For the analyses presented in this paper, a peaks-over-threshold approach was chosen since it allows investigating changes in frequency as well as magnitude. Non-parametric approaches were used to assess changes in frequency, magnitude, and quantile estimates as well as the statistical significance of changes for one station (Sydney Observatory Hill) for the period 1921 to 2005. Deviations from the long-term average vary with season, duration, and threshold. The effects of climate variations are most readily detected for the highest thresholds. Deviations from the long-term average tend to be larger for frequencies than for magnitudes, and changes in frequency and magnitude may have opposite signs. <br><br> Investigations presented in this paper show that variations in frequency and magnitude of events at daily durations are a poor indicator of changes at sub-daily durations. Studies like the one presented here should be undertaken for other regions to allow the identification of regions with significant increase/decrease in intense rainfall, whether there are common features with regards to duration and season exhibiting most significant changes (which in turn could lead to establishing a theoretical framework), and assist in validation of projections of rainfall extremes

    Development and validation of a measure of health literacy in the UK: the newest vital sign

    Background: Health literacy (HL) is an important public health issue. Current measures have drawbacks in length and/ or acceptability. The US-developed Newest Vital Sign (NVS) health literacy instrument measures both reading comprehension and numeracy skills using a nutrition label, takes 3 minutes to administer, and has proven to be acceptable to research subjects. This study aimed to amend and validate it for the UK population.Methods: We used a three-stage process; (1) a Delphi study with academic and clinical experts to amend the NVS label to reflect UK nutrition labeling (2) community-based cognitive testing to assess and improve ease of understanding and acceptability of the test (3) validation of the NVS-UK against an accepted standard test of health literacy, the Test of Functional Health Literacy in Adults (TOFHLA) (Pearson's r and the area under the Receiver Operating Characteristic (ROC) curve) and participant educational level. A sample size calculation indicated that 250 participants would be required. Inclusion criteria were age 18-75 years and ability to converse in English. We excluded people working in the health field and those with impaired vision or inability to undertake the interview due to cognitive impairment or inability to converse in English.Results: In the Delphi study, 28 experts reached consensus (3 cycles). Cognitive testing (80 participants) yielded an instrument that needed no further refinement. Validation testing (337 participants) showed high internal consistency (Cronbach's Alpha = 0.74). Validation against the TOFHLA demonstrated a Pearson's r of 0.49 and an area under the ROC curve of 0.81.Conclusions: The NVS-UK is a valid measure of HL. Its acceptability and ease of application makes it an ideal tool for use in the UK. It has potential uses in public health research including epidemiological surveys and randomized controlled trials, and in enabling practitioners to tailor care to patient need.</p

    Let us conserve and exchange seeds: celebrating traditional crop diversity of the Nepali lowlands

    A seed fair is an activity to create awareness about and appreciate local crop diversity, exchange seed and related knowledge, and celebrate farmers’ efforts to conserve agrobiodiversity. It takes considerable time and effort to organize a seed fair. This brief describes the seed fair organized at the Agyauli Community Seedbank, Nawalparasi in the southern region of Nepal. About 30 members of 10 community seedbanks from the terai (the southern lowland) region of Nepal came together for this. Apart from exchanging seeds of traditional crop varieties, they also shared stories about the socio-cultural, religious, spiritual, nutritional and medicinal values of their varieties. The recent formal registration of the Community Seed Banks Association of Nepal (CSBAN) was also celebrated

    Disrupted Maturation of the Microbiota and Metabolome among Extremely Preterm Infants with Postnatal Growth Failure

    Growth failure during infancy is a major global problem that has adverse effects on long-term health and neurodevelopment. Preterm infants are disproportionately affected by growth failure and its effects. Herein we found that extremely preterm infants with postnatal growth failure have disrupted maturation of the intestinal microbiota, characterized by persistently low diversity, dominance of pathogenic bacteria within the Enterobacteriaceae family, and a paucity of strictly anaerobic taxa including Veillonella relative to infants with appropriate postnatal growth. Metabolomic profiling of infants with growth failure demonstrated elevated serum acylcarnitines, fatty acids, and other byproducts of lipolysis and fatty acid oxidation. Machine learning algorithms for normal maturation of the microbiota and metabolome among infants with appropriate growth revealed a pattern of delayed maturation of the microbiota and metabolome among infants with growth failure. Collectively, we identified novel microbial and metabolic features of growth failure in preterm infants and potentially modifiable targets for intervention
