The Geological Record of Ocean Acidification
Ocean acidification may have severe consequences for marine ecosystems; however, assessing its future impact is difficult because laboratory experiments and field observations are limited by their reduced ecologic complexity and sample period, respectively. In contrast, the geological record contains long-term evidence for a variety of global environmental perturbations, including ocean acidification plus their associated biotic responses. We review events exhibiting evidence for elevated atmospheric CO2, global warming, and ocean acidification over the past ~300 million years of Earth's history, some with contemporaneous extinction or evolutionary turnover among marine calcifiers. Although similarities exist, no past event perfectly parallels future projections in terms of disrupting the balance of ocean carbonate chemistry—a consequence of the unprecedented rapidity of CO2 release currently taking place
Novel genetic loci associated with hippocampal volume
The hippocampal formation is a brain structure integrally involved in episodic memory, spatial navigation, cognition and stress responsiveness. Structural abnormalities in hippocampal volume and shape are found in several common neuropsychiatric disorders. To identify the genetic underpinnings of hippocampal structure here we perform a genome-wide association study (GWAS) of 33,536 individuals and discover six independent loci significantly associated with hippocampal volume, four of them novel. Of the novel loci, three lie within genes (ASTN2, DPP4 and MAST4) and one is found 200 kb upstream of SHH. A hippocampal subfield analysis shows that a locus within the MSRB3 gene shows evidence of a localized effect along the dentate gyrus, subiculum, CA1 and fissure. Further, we show that genetic variants associated with decreased hippocampal volume are also associated with increased risk for Alzheimer's disease (rg =-0.155). Our findings suggest novel biological pathways through which human genetic variation influences hippocampal volume and risk for neuropsychiatric illness
Gene flow in a national cross-breeding beef population
Future progress in genetic improvement and the monitoring of genetic resources in beef cattle requires a detailed understanding of the population under selection. This study examines the gene flow in the UK beef population with an uncommon breeding structure involving interaction between the beef and dairy populations. British Cattle Movement Service records were used as the primary source of information, and these data were triangulated with UK government statistics, other industry information sources and existing literature to build up a profile of the UK beef industry. Estimates were made of the breed composition of suckler cows, breeding bulls and the prime slaughter population. Cross-bred animals made up 85% and 94%, respectively, of the commercial beef breeding cow and prime slaughter populations. Holstein/Friesian (through cross-breeding) made up the largest proportion of genes in both these populations with 33% and 28%, respectively. The next five most popular breeds were specialist beef breeds: Limousin (22% and 18%), Charolais (11% and 6%), Simmental (9% and 11%), Angus (7% and 8%) and Belgian Blue (6% and 6%). Combined, the top seven beef breeds accounted for 94% of beef genetics in the prime slaughter population, and 80% of this came from non-native breeds. The influence of dairy breeds in the commercial beef breeding population was highlighted by the fact that 44% of replacement commercial beef breeding females were sourced from beef-sired crosses in the dairy herd, and in total 74% of all maternal grand dams of prime slaughter animals were Holstein/Friesian. The use of selection index technology was also investigated by analysing breeding bull sale results, with the correlation between the terminal sire index and sale price of young breeding bulls being generally moderate but significant, ranging from 0.21 to 0.38 across the major beef breeds. The most influential source of genetics in the commercial suckler beef herd was natural service breeding bulls. These were mostly sourced from pedigree breeders, and accounted for 47.8% of the genetics in the prime beef population. Artificial insemination sires were responsible for 16.6% of prime beef genetics, with the remaining 35.6% coming from dairy breeds, 95% of which was Holstein/Friesian
The Immune Landscape of Cancer
We performed an extensive immunogenomic analysis of more than 10,000 tumors comprising 33 diverse cancer types by utilizing data compiled by TCGA. Across cancer types, we identified six immune subtypes—wound healing, IFN-γ dominant, inflammatory, lymphocyte depleted, immunologically quiet, and TGF-β dominant—characterized by differences in macrophage or lymphocyte signatures, Th1:Th2 cell ratio, extent of intratumoral heterogeneity, aneuploidy, extent of neoantigen load, overall cell proliferation, expression of immunomodulatory genes, and prognosis. Specific driver mutations correlated with lower (CTNNB1, NRAS, or IDH1) or higher (BRAF, TP53, or CASP8) leukocyte levels across all cancers. Multiple control modalities of the intracellular and extracellular networks (transcription, microRNAs, copy number, and epigenetic processes) were involved in tumor-immune cell interactions, both across and within immune subtypes. Our immunogenomics pipeline to characterize these heterogeneous tumors and the resulting data are intended to serve as a resource for future targeted studies to further advance the field. Thorsson et al. present immunogenomics analyses of more than 10,000 tumors, identifying six immune subtypes that encompass multiple cancer types and are hypothesized to define immune response patterns impacting prognosis. This work provides a resource for understanding tumor-immune interactions, with implications for identifying ways to advance research on immunotherapy
Reflectivity, reflexivity and situated reflective practice
This paper describes an aspect of reflective practice referred to as ‘Situated Reflective Practice’ (SRP). The overarching theory is derived from social theories of structuration and reflexivity. In particular, from Giddens’s (1984) theory of structuration, this sees social life as interplay of agency and structure. Discussion of the research reported here centres on the nature of such situated reflection, considers related literature and presents the data collected in a recent small-scale study. The original purpose of the research was to explore the perceptions of corporate trainers on a course for Preparing to Teach in the Lifelong Learning Sector (PTLLS). As a result of this research the concept of Situated Reflective Practice was generated. It is argued that here exist some situations where a person will find themselves in a position over which they have little control, avoidance or veto. The major conclusion of this study was that key interview themes enabled the delineation of a series of five characteristics representing increasing structural distance in space and time between the reflective practitioner and the professional situation in which they work
Localization of the murine cholecystokinin A and B receptor genes
We have determined the chromosomal locations of the two cholecystokinin (CCK) receptor genes in the mouse. Genetic localization utilized an interspecific backcross panel formed from the cross (C57BL/6J x Mus spretus ) F 1 x Mus spretus . Genomic DNAs from 94 individuals in the backcross were analyzed by Southern hybridization with rat CCK A and CCK B receptor cDNA probes. Unique map positions were determined by haplotype analysis with 650 previously mapped loci in the mouse backcross. The CCK A receptor gene ( Cckar ) mapped to mouse Chromosome (Chr) 5, in tight linkage with the DNA marker D5Bir8 . The CCK B receptor gene ( Cckbr ) mapped to mouse Chr 7, tightly linked to the β-hemoglobin locus ( Hbb ). This localization places Cckbr in the same region as the mouse obesity mutation tubby ( tub ), which also maps near Hbb (2.4±1.4 cM). Since CCK can function as a satiety factor when administered to rodents, localization of Cckbr near the tub mutation identifies this receptor as a possible candidate gene for this obesity mutation.Peer Reviewedhttp://deepblue.lib.umich.edu/bitstream/2027.42/47021/1/335_2004_Article_BF00352408.pd
