Cluster J Mycobacteriophages: Intron Splicing in Capsid and Tail Genes
Bacteriophages isolated on Mycobacterium smegmatis mc2155 represent many distinct genomes sharing little or no DNA sequence similarity. The genomes are architecturally mosaic and are replete with genes of unknown function. A new group of genomes sharing substantial nucleotide sequences constitute Cluster J. The six mycobacteriophages forming Cluster J are morphologically members of the Siphoviridae, but have unusually long genomes ranging from 106.3 to 117 kbp. Reconstruction of the capsid by cryo-electron microscopy of mycobacteriophage BAKA reveals an icosahedral structure with a triangulation number of 13. All six phages are temperate and homoimmune, and prophage establishment involves integration into a tRNA-Leu gene not previously identified as a mycobacterial attB site for phage integration. The Cluster J genomes provide two examples of intron splicing within the virion structural genes, one in a major capsid subunit gene, and one in a tail gene. These genomes also contain numerous free-standing HNH homing endonuclease, and comparative analysis reveals how these could contribute to genome mosaicism. Twenty-one lessons: preventing domestic violence in the Caribbean
Domestic violence infringes the basic right to security of the individual and affects society as a whole – it impacts on demography, education and health systems, the economy, political participation and the overall security of a country. Domestic violence is a universal problem which affects all parts of the social fabric; it transcends ethnic, gender, religious, generational and economic lines. In Barbados and the Eastern Caribbean, beyond the ratification of a number of international instruments, domestic violence is widely recognised as a persistent challenge. On average, more than one in three women in the Caribbean experience gender-based violence in their lifetime; child sexual abuse is also a concern. The causes behind domestic violence in Barbados and the Eastern Caribbean are complex and deeply entrenched in cultural and behavioural norms. The prevention of domestic violence is vital if protection rights are to be effectively safeguarded. To this end, a sustainable and community driven approach -involving civil society and the various stakeholders - is crucial in effecting changes in cultural attitudes towards domestic violence. Early and continuous education about gender based violence and a focus on the prosecution and rehabilitation of perpetrators are both necessary to address the problem at its root. In addition, it is important that the relevant legal framework - and accompanying measures - is fortified and effectively enforced, so that the safety of victims and their families is adequately provided for. A co-ordinated approach between the various stakeholders is important to allow public and private mitigation and response mechanisms toward both protective and restorative safety nets (European Instrument for Democracy and Human Rights (EIDHR), ‘Towards a Future Free from Domestic Violence’, Guidelines for grant applicants 2014)
Modeling the cosmological co-evolution of supermassive black holes and galaxies: II. The clustering of quasars and their dark environment
We use semi-analytic modeling on top of the Millennium simulation to study
the joint formation of galaxies and their embedded supermassive black holes.
Our goal is to test scenarios in which black hole accretion and quasar
activity are triggered by galaxy mergers, and to constrain different models for
the lightcurves associated with individual quasar events. In the present work
we focus on studying the spatial distribution of simulated quasars. At all
luminosities, we find that the simulated quasar two-point correlation function
is fit well by a single power-law in the range 0.5 < r < 20 h^{-1} Mpc, but its
normalization is a strong function of redshift. When we select only quasars
with luminosities within the range typically accessible by today's quasar
surveys, their clustering strength depends only weakly on luminosity, in
agreement with observations. This holds independently of the assumed lightcurve
model, since bright quasars are black holes accreting close to the Eddington
limit, and are hosted by dark matter haloes with a narrow mass range of a few
10^12 h^{-1} M_sun. Therefore the clustering of bright quasars cannot be used
to disentangle lightcurve models, but such a discrimination would become
possible if the observational samples can be pushed to significantly fainter
limits.
Overall, our clustering results for the simulated quasar population agree
rather well with observations, lending support to the conjecture that galaxy
mergers could be the main physical process responsible for triggering black

Necessary and sufficient conditions of solution uniqueness in minimization
This paper shows that the solutions to various convex minimization
problems are \emph{unique} if and only if a common set of conditions are
satisfied. This result applies broadly to the basis pursuit model, basis
pursuit denoising model, Lasso model, as well as other models that
either minimize or impose the constraint , where
is a strictly convex function. For these models, this paper proves that,
given a solution and defining I=\supp(x^*) and s=\sign(x^*_I),
is the unique solution if and only if has full column rank and there
exists such that and for . This
condition is previously known to be sufficient for the basis pursuit model to
have a unique solution supported on . Indeed, it is also necessary, and
applies to a variety of other models. The paper also discusses ways to

The role of childhood social position in adult type 2 diabetes: Evidence from the English Longitudinal Study of Ageing

The main aim of the present study is to examine the role of childhood social position and later inflammatory markers and health behaviours in developing type 2 diabetes at older ages using a pathway analytic approach. Methods. Data on childhood and adult life circumstances of 2,994 men and 4,021 women from English Longitudinal Study of Ageing (ELSA) were used to evaluate their association with diabetes at age 50 years and more. The cases of diabetes were based on having increased blood levels of glycated haemoglobin and/or self-reported medication for diabetes and/or being diagnosed with type 2 diabetes. Father's job when ELSA participants were aged 14 years was used as the measure of childhood social position. Current social characteristics, health behaviours and inflammatory biomarkers were used as potential mediators in the statistical analysis to assess direct and indirect effects of childhood circumstances on diabetes in later life. Results: 12.6 per cent of participants were classified as having diabetes. A disadvantaged social position in childhood, as measured by father's manual occupation, was associated at conventional levels of statistical significance with an increased risk of type 2 diabetes in adulthood, both directly and indirectly through inflammation, adulthood social position and a risk score constructed from adult health behaviours including tobacco smoking and limited physical activity. The direct effect of childhood social position was reduced by mediation analysis (standardised coefficient decreased from 0.089 to 0.043) but remained statistically significant (p = 0.035). All three indirect pathways made a statistically significantly contribution to the overall effect of childhood social position on adulthood type 2 diabetes. In-depth study of moderately young but extremely red, very dusty substellar companion HD206893B

Reproduced with permission from Astronomy & Astrophysics. © 2018 ESO.The substellar companion HD206893b has recently been discovered by direct imaging of its disc-bearing host star with the SPHERE instrument. We investigate the atypical properties of the companion, which has the reddest near-infrared colours among all known substellar objects, either orbiting a star or isolated, and we provide a comprehensive characterisation of the host star-disc-companion system. We conducted a follow-up of the companion with adaptive optics imaging and spectro-imaging with SPHERE, and a multiinstrument follow-up of its host star. We obtain a R=30 spectrum from 0.95 to 1.64 micron of the companion and additional photometry at 2.11 and 2.25 micron. We carried out extensive atmosphere model fitting for the companions and the host star in order to derive their age, mass, and metallicity. We found no additional companion in the system in spite of exquisite observing conditions resulting in sensitivity to 6MJup (2MJup) at 0.5" for an age of 300 Myr (50 Myr). We detect orbital motion over more than one year and characterise the possible Keplerian orbits. We constrain the age of the system to a minimum of 50 Myr and a maximum of 700 Myr, and determine that the host-star metallicity is nearly solar. The comparison of the companion spectrum and photometry to model atmospheres indicates that the companion is an extremely dusty late L dwarf, with an intermediate gravity (log g 4.5-5.0) which is compatible with the independent age estimate of the system. Though our best fit corresponds to a brown dwarf of 15-30 MJup aged 100-300 Myr, our analysis is also compatible with a range of masses and ages going from a 50 Myr 12MJup planetary-mass object to a 50 MJup Hyades-age brown dwarf...Peer reviewedFinal Accepted Versio
Experimental Assessment of the Water Quality Influence on the Phosphorus Uptake of an Invasive Aquatic Plant: Biological Responses throughout Its Phenological Stage
International audienceUnderstanding how an invasive plant can colonize a large range of environments is still a great challenge in freshwater ecology. For the first time, we assessed the relative importance of four factors on the phosphorus uptake and growth of an invasive macrophyte Elodea nuttallii (Planch.) St. John. This study provided data on its phenotypic plasticity, which is frequently suggested as an important mechanism but remains poorly investigated. The phosphorus uptake of two Elodea nuttallii subpopulations was experimentally studied under contrasting environmental conditions. Plants were sampled in the Rhine floodplain and in the Northern Vosges mountains, and then maintained in aquaria in hard (Rhine) or soft (Vosges) water. Under these conditions, we tested the influence of two trophic states (eutrophic state, 100 mu g.l(-1) P-PO43- and hypertrophic state, 300 mu g.l(-1) P-PO43-) on the P metabolism of plant subpopulations collected at three seasons (winter, spring and summer). Elodea nuttallii was able to absorb high levels of phosphorus through its shoots and enhance its phosphorus uptake, continually, after an increase of the resource availability (hypertrophic > eutrophic). The lowest efficiency in nutrient use was observed in winter, whereas the highest was recorded in spring, what revealed thus a storage strategy which can be beneficial to new shoots. This experiment provided evidence that generally, the water trophic state is the main factor governing P uptake, and the mineral status (softwater > hardwater) of the stream water is the second main factor. The phenological stage appeared to be a confounding factor to P level in water. Nonetheless, phenology played a role in P turnover in the plant. Finally, phenotypic plasticity allows both subpopulations to adapt to a changing environment
Identification of Degraded Land in the Canary Islands; Tests and Reviews
Degraded Land is an area that either by natural causes (fires, floods, storms or volcanic eruptions) or more by direct or indirect causes of human action, has been altered or modified from its natural state. Restoration is an activity that initiates or accelerates the recovery of an ecosystem. It can be defined as the set of actions taken in order to reverse or reduce the damage caused in the territory. In the case of the Canary Islands there is a high possibility for the territory to suffer processes that degrade the environment, given that the islands are very fragile ecosystems. Added to this they are territories isolated from the continent, which complicates the process of restoring them. In this paper, the different types of common degraded areas in the Canary Islands are identified, as well as the proposed solutions for remediation, such as afforestation of agricultural land or landfill closure and restoration
The cytotoxic T cell proteome and its shaping by the kinase mTOR
High-resolution mass spectrometry maps the cytotoxic T lymphocyte (CTL) proteome and the impact of mammalian target of rapamycin complex 1 (mTORC1) on CTLs. The CTL proteome was dominated by metabolic regulators and granzymes and mTORC1 selectively repressed and promoted expression of subset of CTL proteins (~10%). These included key CTL effector molecules, signaling proteins and a subset of metabolic enzymes. Proteomic data highlighted the potential for mTORC1 negative control of phosphatidylinositol (3,4,5)-trisphosphate (PtdIns(3,4,5)P(3)) production in CTL. mTORC1 was shown to repress PtdIns(3,4,5)P(3) production and to determine the mTORC2 requirement for activation of the kinase Akt. Unbiased proteomic analysis thus provides a comprehensive understanding of CTL identity and mTORC1 control of CTL function
