A dynamical trichotomy for structured populations experiencing positive density-dependence in stochastic environments
Positive density-dependence occurs when individuals experience increased
survivorship, growth, or reproduction with increased population densities.
Mechanisms leading to these positive relationships include mate limitation,
saturating predation risk, and cooperative breeding and foraging. Individuals
within these populations may differ in age, size, or geographic location and
thereby structure these populations. Here, I study structured population models
accounting for positive density-dependence and environmental stochasticity i.e.
random fluctuations in the demographic rates of the population. Under an
accessibility assumption (roughly, stochastic fluctuations can lead to
populations getting small and large), these models are shown to exhibit a
dynamical trichotomy: (i) for all initial conditions, the population goes
asymptotically extinct with probability one, (ii) for all positive initial
conditions, the population persists and asymptotically exhibits unbounded
growth, and (iii) for all positive initial conditions, there is a positive
probability of asymptotic extinction and a complementary positive probability
of unbounded growth. The main results are illustrated with applications to
spatially structured populations with an Allee effect and age-structured
populations experiencing mate limitation
'A Monstrous Failure of Justice'?:Guantanamo Bay and National Security Challenges to Fundamental Human Rights
This article considers challenges to the existing international human rights regime in the post-9/11 era. It uses an interdisciplinary approach that brings together issues of politics and law by focussing on international legal provisions and setting them into the context of International Relations theory. The article examines the establishment of Guantanamo Bay as a detention centre for suspected terrorists captured in the 'war on terror' and focuses on violations of international human rights and humanitarian law in the name of national security. This article demonstrates that the wrangling over Guantanamo Bay is an important illustration of the complex interaction between interests and norms as well as law and politics in US policy making. The starting point is that politics and law are linked and cannot be seen in isolation from each other; the question that then arises is what kind of politics law can maintain. Access to Principal Treatment Centres and survival rates for children and young people with cancer in Yorkshire, UK
Background: Principal Treatment Centres (PTC) were established to provide age-appropriate care as well as clinical expertise for children and young people with cancer. However, little is known about the effects of specialist treatment centres on survival outcomes especially for teenagers and young adults. This population-based study aimed to describe access to PTC and the associated trends in survival for 0–24 year olds accounting for stage of disease at presentation and treatment. Methods: Patients diagnosed from 1998–2009 aged 0–24 years were extracted from the Yorkshire Specialist Register of Cancer in Children and Young People, including information on all treating hospitals, followed-up until 31st December 2014. The six commonest cancer types were included: leukaemia (n = 684), lymphoma (n = 558), CNS tumours (n = 547), germ cell tumours (n = 364), soft tissue sarcomas (n = 171) and bone tumours (n = 163). Treatment was categorised into three groups: ‘all’, ‘some’ or ‘no’ treatment received at a PTC. Treatment at PTC was examined by diagnostic group and patient characteristics. Overall survival was modelled using Cox regression adjusting for case-mix including stage, treatment and other socio-demographic and clinical characteristics. Results: Overall 72% of patients received all their treatment at PTC whilst 13% had no treatment at PTC. This differed by diagnostic group and age at diagnosis. Leukaemia patients who received no treatment at PTC had an increased risk of death which was partially explained by differences in patient case-mix (adjusted Hazard Ratio (HR) = 1.73 (95%CI 0.98–3.04)). Soft tissue sarcoma patients who had some or no treatment at PTC had better survival outcomes, which remained after adjustment for patient case-mix (adjusted HR = 0.48 (95%CI 0.23–0.99)). There were no significant differences in outcomes for other diagnostic groups (lymphoma, CNS tumours, bone tumours and germ cell tumours). For leukaemia patients survival outcomes for low risk patients receiving no treatment at PTC were similar to high risk patients who received all treatment at PTC, implying a benefit for care at the PTC. Conclusion: This study demonstrates that for leukaemia patients receiving treatment at a PTC is associated with improved survival that may compensate for a poorer prognosis presentation. However, further information on risk factors is needed for all diagnostic groups in order to fully account for differences in patient case-mix
Mechanical Stress Inference for Two Dimensional Cell Arrays
Many morphogenetic processes involve mechanical rearrangement of epithelial
tissues that is driven by precisely regulated cytoskeletal forces and cell
adhesion. The mechanical state of the cell and intercellular adhesion are not
only the targets of regulation, but are themselves likely signals that
coordinate developmental process. Yet, because it is difficult to directly
measure mechanical stress {\it in vivo} on sub-cellular scale, little is
understood about the role of mechanics of development. Here we present an
alternative approach which takes advantage of the recent progress in live
imaging of morphogenetic processes and uses computational analysis of high
resolution images of epithelial tissues to infer relative magnitude of forces
acting within and between cells. We model intracellular stress in terms of bulk
pressure and interfacial tension, allowing these parameters to vary from cell
to cell and from interface to interface. Assuming that epithelial cell layers
are close to mechanical equilibrium, we use the observed geometry of the two
dimensional cell array to infer interfacial tensions and intracellular
pressures. Here we present the mathematical formulation of the proposed
Mechanical Inverse method and apply it to the analysis of epithelial cell
layers observed at the onset of ventral furrow formation in the {\it
Drosophila} embryo and in the process of hair-cell determination in the avian
cochlea. The analysis reveals mechanical anisotropy in the former process and
mechanical heterogeneity, correlated with cell differentiation, in the latter
process. The method opens a way for quantitative and detailed experimental
tests of models of cell and tissue mechanics
Spatially Resolved Magnetic Field Structure in the Disk of a T Tauri Star
Magnetic fields in accretion disks play a dominant role during the star
formation process but have hitherto been observationally poorly constrained.
Field strengths have been inferred on T Tauri stars themselves and possibly in
the innermost part of the accretion disk, but the strength and morphology of
the field in the bulk of the disk have not been observed. Unresolved
measurements of polarized emission (arising from elongated dust grains aligned
perpendicular to the field) imply average fields aligned with the disks.
Theoretically, the fields are expected to be largely toroidal, poloidal, or a
mixture of the two, which imply different mechanisms for transporting angular
momentum in the disks of actively accreting young stars such as HL Tau. Here we
report resolved measurements of the polarized 1.25 mm continuum emission from
HL Tau's disk. The magnetic field on a scale of 80 AU is coincident with the
major axis (~210 AU diameter) of the disk. From this we conclude that the
magnetic field inside the disk at this scale cannot be dominated by a vertical
component, though a purely toroidal field does not fit the data well either.
The unexpected morphology suggests that the magnetic field's role for the
accretion of a T Tauri star is more complex than the current theoretical

Proteomics: in pursuit of effective traumatic brain injury therapeutics
Effective traumatic brain injury (TBI) therapeutics remain stubbornly elusive. Efforts in the field have been challenged by the heterogeneity of clinical TBI, with greater complexity among underlying molecular phenotypes than initially conceived. Future research must confront the multitude of factors comprising this heterogeneity, representing a big data challenge befitting the coming informatics age. Proteomics is poised to serve a central role in prescriptive therapeutic development, as it offers an efficient endpoint within which to assess post-TBI biochemistry. We examine rationale for multifactor TBI proteomic studies and the particular importance of temporal profiling in defining biochemical sequences and guiding therapeutic development. Lastly, we offer perspective on repurposing biofluid proteomics to develop theragnostic assays with which to prescribe, monitor and assess pharmaceutics for improved translation and outcome for TBI patients
Neuroactive steroids in depression and anxiety disorders: Clinical studies
Certain neuroactive steroids modulate ligand-gated ion channels via non-genomic mechanisms. Especially 3 alpha-reduced pregnane steroids are potent positive allosteric modulators of the gamma-aminobutyric acid type A (GABA(A)) receptor. During major depression, there is a disequilibrium of 3 alpha-reduced neuroactive steroids, which is corrected by clinically effective pharmacological treatment. To investigate whether these alterations are a general principle of successful antidepressant treatment, we studied the impact of nonpharmacological treatment options on neuroactive steroid concentrations during major depression. Neither partial sleep deprivation, transcranial magnetic stimulation, nor electroconvulsive therapy affected neuroactive steroid levels irrespectively of the response to these treatments. These studies suggest that the changes in neuroactive steroid concentrations observed after antidepressant pharmacotherapy more likely reflect distinct pharmacological properties of antidepressants rather than the clinical response. In patients with panic disorder, changes in neuroactive steroid composition have been observed opposite to those seen in depression. However, during experimentally induced panic induction either with cholecystokinine-tetrapeptide or sodium lactate, there was a pronounced decline in the concentrations of 3 alpha-reduced neuroactive steroids in patients with panic disorder, which might result in a decreased GABAergic tone. In contrast, no changes in neuroactive steroid concentrations could be observed in healthy controls with the exception of 3 alpha,5 alpha-tetrahydrodeoxycorticosterone. The modulation of GABA(A) receptors by neuroactive steroids might contribute to the pathophysiology of depression and anxiety disorders and might offer new targets for the development of novel anxiolytic compounds.
