Transition metal-catalyzed synthesis of 13α-estrone derivatives with potential anticancer properties

Synthesis of 3-(N,N-dimethylcarbamoyl)-13α-estrone and its 17-deoxy counterpart has been carried out. The dimethylcarbamoyl directing group enabled the regioselective ortho arylation of the steroidal starting compounds. The microwave-assisted, palladium-catalyzed phenylations led to 2-substituted 13α-estrones in a one-pot, tandem process. The Suzuki-Miyaura cross coupling reactions of the carbamates with phenylboronic acid resulted in 3-phenyl-3-deoxy13α-estrone. A Highly Virulent Staphylococcus aureus: Rabbit Anterior Chamber Infection, Characterization, and Genetic Analysis

PURPOSE. To describe and characterize a Staphylococcus aureus strain with unique virulence that overcomes host defenses of the rabbit anterior chamber and mimics clinical cases of postcataract surgery endophthalmitis. METHODS. Nine isolates of S. aureus were tested to determine their viability in the rabbit anterior chamber. Growth of UMCR1 in the anterior chamber was established and expressed as log colony-forming units per milliliter of aqueous humor. Pathologic changes produced by UMCR1 were documented by photographs, slit lamp examination, histopathologic analysis, and quantification of neutrophils. UMCR1 was characterized by antibiotic susceptibility, biochemical tests, ribotyping, genome restriction mapping, and multilocus sequence typing (MLST). RESULTS. UMCR1 was the only S. aureus strain that grew within the anterior chamber, reaching log 6.97 Ϯ 0.18 CFU/mL by 16 hours after infection. Pathologic changes included conjunctival injection, chemosis, corneal edema, severe iritis, fibrin accumulation, and a 193-fold increase in neutrophils by 16 hours after infection. UMCR1 was only resistant to sulfamethoxazole and, like other S. aureus isolates, polymyxin B. UMCR1 also had biochemical reactions and a ribotype pattern typical of S. aureus. The genomic reconstruction analysis of UMCR1 was most similar to strains MW2 and MSSA476. MLST revealed a 1 in 3198 nucleotide difference between UMCR1 and strains MW2 and MSSA476. CONCLUSIONS. This study describes a unique S. aureus strain that overcomes host defenses and replicates in the anterior chamber. The survival and growth of this organism could be used for studies of S. aureus pathogenesis, host defenses, and effectiveness of antibiotics within the anterior chamber. Two-site recognition of Staphylococcus aureus peptidoglycan by lysostaphin SH3b

Lysostaphin is a bacteriolytic enzyme targeting peptidoglycan, the essential component of the bacterial cell envelope. It displays a very potent and specific activity toward staphylococci, including methicillin-resistant Staphylococcus aureus. Lysostaphin causes rapid cell lysis and disrupts biofilms, and is therefore a therapeutic agent of choice to eradicate staphylococcal infections. The C-terminal SH3b domain of lysostaphin recognizes peptidoglycans containing a pentaglycine crossbridge and has been proposed to drive the preferential digestion of staphylococcal cell walls. Here we elucidate the molecular mechanism underpinning recognition of staphylococcal peptidoglycan by the lysostaphin SH3b domain. We show that the pentaglycine crossbridge and the peptide stem are recognized by two independent binding sites located on opposite sides of the SH3b domain, thereby inducing a clustering of SH3b domains. We propose that this unusual binding mechanism allows synergistic and structurally dynamic recognition of S. aureus peptidoglycan and underpins the potent bacteriolytic activity of this enzyme

Liberação intraocular de ofloxacina associada a lente de contato biossintética em ceratite bacteriana experimental em cães

Avaliou-se a concentração de ofloxacina liberada por uma lente de contato de membrana de celulose biossintética, para tratamento de ceratite bacteriana experimental em cães, pela inoculação de Staphylococcus aureus intraestromal. Comparou-se o tratamento com a lente de contato biossintética impregnada com ofloxacina à terapia tópica convencional. Realizou-se avaliação microbiológica e dosagem de ofloxacina no humor aquoso por meio do método de cromatografia líquida de alto rendimento (HPLC). Houve diferença estatística na contagem de colônias bacterianas entre os olhos com ceratite e os demais grupos, no primeiro dia de coleta. O biomaterial, impregnado com ofloxacina, promoveu liberação gradual durante o período de avaliação, aos três e sete dias; no terceiro dia, o grupo tratado com a lente de contato obteve mediana de 3,72μg/mL, enquanto o grupo tratado com colírio resultou em 49,56μg/mL. Apesar do valor inferior, o grupo com lente de contato atingiu a concentração inibitória mínima, sendo eficaz no controle da infecção bacteriana

Influenszermarketing tevékenységek az Instagramon

Az online reklámozási formák napjainkban nagy népszerűségnek örvendenek. Dolgozatomban eme hirdetési módszer egyik fajtáját, az influenszermarketinget vizsgálom részletesebben. Azokra a kérdésekre keresem a megoldást, hogy mi okozza az influenszermarketing hatékonyságát, illetve maguknak a tartalomgyártóknak a sikerét, és hogy a vállalatoknak milyen szempontok alapján érdemes influenszerekkel való együttműködéseket szervezni. Külön táblázatban összefoglalom a tíz legnépszerűbb nemzetközi és Magyar Instagram profilt, illtve hazai viszonylatokban összehasonlítom a női és férfi influenszer tevékenységet, illetve külön veszem a kedvelések és a követők számát. A dolgozatban kiderül, hogy bár az influenszerek sikere abban nyugszik, hogy olyan sikeres és híres személyek, akik közösségi platformjukon hétköznapi emberek hatását keltik, ami miatt követőik közelebb érzik magukat hozzájuk. Ettől függetlenül, bár népszerűek, a hagyományos, ,,offline" térben is elismert sztárokat nem tudják túlszárnyalni.Magyarországon alapvető különbség a női és férfi influenszerek között, hogy előbbi csoport tagjai Instagramon keresettebbek, míg utóbbiak más közösségi platformokon (jellemzően a Youtubon). A követőszám tekintetében elmondható, hogy a ,,kevesebb néha több", tehát jobban számít az influenszerek hitelessége, a posztok igényessége, mintsem a követőszám. Ezt a szempontot érdemes a vállalatoknak is szem előtt tartani: nem a követők száma, hanem összetétele a lényeg, hiszen minél jobban illik a cég célcsoportjához, annál nagyobb az esélye a sikeres hirdetéseknek. Mivel a közösségi média marketing nem hagyományos, és viszonlag új reklámozási forma, ezért jobban elfogadott, ez az oka annak, hogy a tartalomgyártók idővonalán megjelenő szponzorációk nem zavarják a követőket, fontosabb tényező, a hitelesség és a posztok minősége.BSc/BAKommunikáció-és Médiatudomán
