A Wide-field Imaging Approach For Simultaneous Super-resolution Surface-enhanced Raman Scattering Imaging And Spectroscopy

    Get PDF
The ability to simultaneously obtain high spatial resolution images and chemical specific information is of interest in a variety of biological and physical processes. Surface-enhanced Raman scattering (SERS) is particularly suited for this purpose due to its ability to enhance signal from Raman vibrational modes by probing molecules near the surface of plasmonic metal nanostructures. The spatial resolution in SERS imaging is limited by the diffraction limit of light, limiting the resolution to hundreds of nanometers. However, Raman reporter molecules adsorbed to single nanoparticles experience temporal intensity fluctuations that enable the SERS signal to be fit with localization algorithms, such as stochastic optical reconstruction microscopy (STORM). STORM fittings can be applied to generate images with sub-diffraction limited localization of the emitting centers from the nanoparticles. In this work, we demonstrate a wide-field spectrally resolved SERS imaging approach where a transmission diffraction grating placed before the imaging array detector captures the image and first-order diffraction on the same detector. The first-order diffraction corresponds to the SERS spectrum and can be directly correlated to the location and features of a nanoparticle. STORM fitting both the spatial and spectral response results in improved localization in the spatial response and improved peak identification compared to the measured spectra in the spectral response. We show that spatially correlated Raman spectra from multiple nanoparticles in a wide-field of view are readily obtained on a 10-100 ms time scale, which enables spatially resolved monitoring of chemical processes

The MDM2-inhibitor Nutlin-3 synergizes with cisplatin to induce p53 dependent tumor cell apoptosis in non-small cell lung cancer

The p53/MDM2 interaction has been a well-studied target for new drug design leading to the development of the small molecule inhibitor Nutlin-3. Our objectives were to combine Nutlin-3 with cisplatin (CDDP), a well-known activator of the p53 pathway, in a series of non-small cell lung cancer cell lines in order to increase the cytotoxic response to CDDP. We report that sequential treatment (CDDP followed by Nutlin-3), but not simultaneous treatment, resulted in strong synergism. Combination treatment induced p53's transcriptional activity, resulting in increased mRNA and protein levels of MDM2, p21, PUMA and BAX. In addition we report the induction of a strong p53 dependent apoptotic response and induction of G2/M cell cycle arrest. The strongest synergistic effect was observed at low doses of both CDDP and Nutlin-3, which could result in fewer (off-target) side effects while maintaining a strong cytotoxic effect. Our results indicate a promising preclinical potential, emphasizing the importance of the applied treatment scheme and the presence of wild type p53 for the combination of CDDP and Nutlin-3

Single cell fertilizer (SCF): Evidence to prove that bio-molecules are potent nutrient for plant growth

    Get PDF
Fertilizers of various kinds are used for the cultivation of crop plants for hyper production of plant based food materials. The study used bio-molecules made in a bacterial cell. The experimental results showed tremendous effect on plant growth. These cellular molecules were made by treating the bacterial cells with lysozyme and protenase K. The wet/weight was increased in multiple folds compared to that of control sets. The fold of increase was 4.79 for rice, 2.77 for wheat, 1.89 for gram and 1.89 for pea when bacterial cellular molecules were used as fertilizer

This article presents the results of an analysis of the alleged defamation case of Marissa Icha by Medina Zein. As a form of forensic linguistics study, the data in this research was analyzed descriptively-analytically using semantic and pragmatic approaches. This research found that 1) semantically, the defendants indicated that they intended to commit acts of insult and/or defamation. 2) pragmatically, the majority of these utterances fell into the form of expressive illocutions, especially to show anger or simply the personal opinion of the defendant, and thus, 3) these speeches can be said to have the potential to violate article 27 paragraph (3) of the ITE Law and articles 310 and 311 of the Criminal Cod

Modelling and Simulation of Current Source Converter based Dynamic Voltage Restorer for Voltage Regulation cum Harmonics Mitigation

    Get PDF
The modern electric power distribution system faces lots of power quality (PQ) problems such as short and long duration voltage variations, voltage imbalance, waveform distortions, impulsive and oscillatory transients and voltage flicker etc. The custom power devices (CPD) have been designed and developed with the aim of mitigating these problems. The major CPDs introduced so far include distribution-static synchronous compensator (D-STATCOM), dynamic voltage restorer (DVR), unified power quality conditioners (UPQC), active power filters (APF) etc.  However, most of these CPDs are designed and implemented using voltage source converter (VSC) topology. Although lots of research works have been carried out for realizing DVR system based on VSC topology, not much research work has been reported on the application of CSC topology in DVR system over the last one decade. Through this paper, it has been attempted to develop a model of DVR system based on CSC topology capable of performing dual power quality enhancement tasks viz. voltage regulation and harmonics mitigation at a power distribution system catering power to a variety of loads. The DVR system has been modeled and simulated in the MATLAB / Simulink platform and the simulation results reveal the effectiveness and validity of the proposed model for use in power distribution system
