Analysing and Improving Business Processes Through Hybrid Simulation Model: A Case Study
The increasing amount of process execution data, i.e. the event logs stored by the company, can be exploited using Business Process Simulation (BPS). BPS serves as a valuable tool for business analysts, enabling them to analyze and compare business processes and identify changes that optimize key performance measures. Especially when evaluating alternative scenarios, it is crucial to start with an accurate simulation of the current process. Recent research in the field of BPS has demonstrated that Hybrid Simulation Model (HSM) approaches reliably replicates business process behaviour, overcoming the unrealistic or oversimplified assumptions often found in traditional discrete event simulators. In this paper, we present a case study conducted in collaboration with EY, where we apply the HSM to a real-life business process log. This study demonstrates the benefits of the HSM for business process analysis and its potential to improve process performance
COPZ1: an example of non-oncogene addiction in human tumors
Non-oncogene addiction (NOA) indicates that tumor cell growth/survival requires the activity of genes/pathways not oncogenic per sé, and dispensable for normal cells. NOA genes provide a wide repertoire of novel therapeutic exploitable tumor vulnerabilities. A large body of evidence demonstrates the dependency of several tumors such as breast, prostate, ovary, thyroid, glioblastoma and LUAD, on the activity of COPZ1, a component of the heptameric COPI complex. Thus, COPZ1 is emerging as a potential novel therapeutic target for tumors of different origin. In different tumor models COPZ1 inhibition was found implicated in abortive autophagy, ER stress and activation of ferroptosis. In this review we summarize the different studies characterizing COPZ1 as a NOA gene in different tumor types, and discuss potential issues related to its targeting
ERIS: revitalising an adaptive optics instrument for the VLT
ERIS is an instrument that will both extend and enhance the fundamental
diffraction limited imaging and spectroscopy capability for the VLT. It will
replace two instruments that are now being maintained beyond their operational
lifetimes, combine their functionality on a single focus, provide a new
wavefront sensing module that makes use of the facility Adaptive Optics System,
and considerably improve their performance. The instrument will be competitive
with respect to JWST in several regimes, and has outstanding potential for
studies of the Galactic Center, exoplanets, and high redshift galaxies. ERIS
had its final design review in 2017, and is expected to be on sky in 2020. This
contribution describes the instrument concept, outlines its expected

Lipid Cubic Mesophases Combined with Superparamagnetic Iron Oxide Nanoparticles: A Hybrid Multifunctional Platform with Tunable Magnetic Properties for Nanomedical Applications
Hybrid materials composed of superparamagnetic iron oxide nanoparticles (SPIONs) and lipid self-assemblies possess considerable applicative potential in the biomedical field, specifically, for drug/nutrient delivery. Recently, we showed that SPIONs-doped lipid cubic liquid crystals undergo a cubic-to-hexagonal phase transition under the action of temperature or of an alternating magnetic field (AMF). This transition triggers the release of drugs embedded in the lipid scaffold or in the water channels. In this contribution, we address this phenomenon in depth, to fully elucidate the structural details and optimize the design of hybrid multifunctional carriers for drug delivery. Combining small-angle X-ray scattering (SAXS) with a magnetic characterization, we find that, in bulk lipid cubic phases, the cubic-to-hexagonal transition determines the magnetic response of SPIONs. We then extend the investigation from bulk liquid-crystalline phases to colloidal dispersions, i.e., to lipid/SPIONs nanoparticles with cubic internal structure (“magnetocubosomes”). Through Synchrotron SAXS, we monitor the structural response of magnetocubosomes while exposed to an AMF: the magnetic energy, converted into heat by SPIONs, activates the cubic-to-hexagonal transition, and can thus be used as a remote stimulus to spike drug release “on-demand”. In addition, we show that the AMF-induced phase transition in magnetocubosomes steers the realignment of SPIONs into linear string assemblies and connect this effect with the change in their magnetic properties, observed at the bulk level. Finally, we assess the internalization ability and cytotoxicity of magnetocubosomes in vitro on HT29 adenocarcinoma cancer cells, in order to test the applicability of these smart carriers in drug delivery applications
Disembodied Mind: Cortical Changes Following Brainstem Injury in Patients with Locked-in Syndrome
Locked-in syndrome (LIS) following ventral brainstem damage is the most severe form of motor disability. Patients are
completely entrapped in an unresponsive body despite consciousness is preserved. Although the main feature of LIS is this extreme
motor impairment, minor non-motor dysfunctions such as motor imagery defects and impaired emotional recognition have been
reported suggesting an alteration of embodied cognition, defined as the effects that the body and its performances may have on
cognitive domains. We investigated the presence of structural cortical changes in LIS, which may account for the reported cognitive
dysfunctions. For this aim, magnetic resonance imaging scans were acquired in 11 patients with LIS (6 males and 5 females; mean
age: 52.3\ub15.2SD years; mean time interval from injury to evaluation: 9\ub11.2SD months) and 44 healthy control subjects matching
patients for age, sex and education. Freesurfer software was used to process data and to estimate cortical volumes in LIS patients as
compared to healthy subjects. Results showed a selective cortical volume loss in patients involving the superior frontal gyrus, the
pars opercularis and the insular cortex in the left hemisphere, and the superior and medium frontal gyrus, the pars opercularis, the
insular cortex, and the superior parietal lobule in the right hemisphere. As these structures are typically associated with the mirror
neuron system, which represents the neural substrate for embodied simulation processes, our results provide neuroanatomical support
for potential disembodiment in LIS
Profiling and assessing the risks of image- and performance-enhancing drugs use during the COVID-19 lockdown

This is an open access article distributed under the terms of the Creative Commons Attribution License (CC BY), https://creativecommons.org/licenses/by/4.0/Background: Image and Performance-Enhancing Drugs (IPEDs) can enhance mental and physical capabilities and impact one’s overall health. Initially confined in sport environments, IPEDs use has become increasingly widespread in a high-performing society. The present study was aimed at profiling IPEDs use during the COVID-19 lockdown among an international sample of young adults. Methods: A cross-sectional observational study was carried out in eight countries (United Kingdom, Italy, Lithuania, Hungary, Portugal, Spain, Brazil, and Japan) between April and May 2020. The survey questionnaire included validated measurements such as Exercise Addiction Inventory (EAI), Appearance Anxiety Inventory (AAI), and Self-Compassion Scale (SCS) as well as questions about the type of IPEDs, purchasing methods and socio-demographic information. Results: A total of 736 IPEDs users were included in the survey. Their mean age was 33.05 years (±SD = 10.06), and 64.2% were female participants. Overall, 6.8% were found at risk of exercise addiction (EAI >24), 27.6% presented high levels of appearance anxiety, and 24.9% revealed low levels of emotional regulation’s self-compassion. Most participants (55.6%) purchased IPEDs through pharmacies/specialized shops, while 41.3% purchased IPEDs on the Internet. Online IPEDs buyers were mainly men who had higher scores on the Exercise Addiction Inventory. One or more IPEDs classifiable as “potentially risky” were used by 66.3% of the sample. Users of “potentially risky IPEDs” were younger and primarily men. They showed higher scores both on the Exercise Addiction Inventory and Appearance Anxiety Inventory. Conclusion: This study profiled users of IPEDs when the most restrictive COVID-19 lockdown policies were implemented in all the participating countries. More targeted post-COVID 19 prevention strategies should be implemented according to the emerged socio-demographic and psychopathological traits and cross-cultural differences emerged. Longitudinal studies will also be needed to determine the long-term effect of the COVID-19 lockdown on IPEDs consumption.Peer reviewe
Patient Satisfaction with IBD Undergoing Colonoscopy: A Multicenter Cross-Sectional Study
Background: Colonoscopy is crucial for diagnosing and monitoring inflammatory bowel disease (IBD), assessing disease activity, and detecting dysplasia. However, patient adherence to surveillance remains suboptimal due to discomfort, anxiety, and concerns about bowel preparation. Methods: This multicenter cross-sectional study assessed patient satisfaction with colonoscopy in IBD patients across three Italian centers. Participants completed pre- and post-examination questionnaires, including the Endoscopy Customer Satisfaction Questionnaire (ECSQ) and Perceived Stress Scale (PSS-10). Clinical factors, bowel preparation methods, and healthcare provider expertise were analyzed. Results: Among 444 enrolled patients, overall satisfaction was high (98.8%) but varied across procedural phases. Higher satisfaction was predicted by expert endoscopists (β = 2.11, p = 0.012), disease remission (β = 1.70, p = 0.020), and frequent endoscopic procedures in the last 24 months (β = 0.46, p = 0.041). Conversely, severe disease activity (β = -3.87, p < 0.001) was associated with lower satisfaction. Deep sedation and high-volume bowel preparation negatively impacted satisfaction. Conclusions: Optimizing bowel preparation, enhancing healthcare provider expertise, and implementing stress-reducing strategies could improve patient adherence to surveillance guidelines in IBD care
Biologically driven cut-off definition of lymphocyte ratios in metastatic breast cancer and association with exosomal subpopulations and prognosis
High neutrophil to lymphocyte ratio (NLR) and monocyte to lymphocyte ratio (MLR) are respectively associated with systemic inflammation and immune suppression and have been associated with a poor outcome. Plasmatic exosomes are extracellular vesicles involved in the intercellular communication system that can exert an immunosuppressive function. Aim of this study was to investigate the interplay between the immune system and circulating exosomes in metastatic breast cancer (MBC). A threshold capable to classify patients according to MLR, NLR and PLR, was computed through a receiving operator curve analysis after propensity score matching with a series of female blood donors. Exosomes were isolated from plasma by ExoQuick solution and characterized by flow-cytometry. NLR, MLR, PLR and exosomal subpopulations potentially involved in the pre-metastatic niche were significantly different in MBC patients with respect to controls. MLR was significantly associated with number of sites at the onset of metastatic disease, while high levels of MLR and NLR were found to be associated with poor prognosis. Furthermore, exosomal subpopulations varied according to NLR, MLR, PLR and both were associated with different breast cancer subtypes and sites of distant involvement. This study highlights the nuanced role of immunity in MBC spread, progression and outcome. Moreover, they suggest potential interaction mechanisms between immunity, MBC and the metastatic niche
