    Transcriptome assembly and profiling of Candida auris reveals novel insights into biofilm-mediated resistance

    Candida auris has emerged as a significant global nosocomial pathogen. This is primarily due to its antifungal resistance profile but also its capacity to form adherent biofilm communities on a range of clinically important substrates. While we have a comprehensive understanding of how other Candida species resist and respond to antifungal challenge within the sessile phenotype, our current understanding of C. auris biofilm-mediated resistance is lacking. In this study, we are the first to perform transcriptomic analysis of temporally developing C. auris biofilms, which were shown to exhibit phase- and antifungal class-dependent resistance profiles. A de novo transcriptome assembly was performed, where sequenced sample reads were assembled into an ~11.5-Mb transcriptome consisting of 5,848 genes. Differential expression (DE) analysis demonstrated that 791 and 464 genes were upregulated in biofilm formation and planktonic cells, respectively, with a minimum 2-fold change. Adhesin-related glycosylphosphatidylinositol (GPI)-anchored cell wall genes were upregulated at all time points of biofilm formation. As the biofilm developed into intermediate and mature stages, a number of genes encoding efflux pumps were upregulated, including ATP-binding cassette (ABC) and major facilitator superfamily (MFS) transporters. When we assessed efflux pump activity biochemically, biofilm efflux was greater than that of planktonic cells at 12 and 24 h. When these were inhibited, fluconazole sensitivity was enhanced 4- to 16-fold. This study demonstrates the importance of efflux-mediated resistance within complex C. auris communities and may explain the resistance of C. auris to a range of antimicrobial agents within the hospital environment

    Fish assemblages at a deep-water sewage disposal site (DWD-106) : a final contract report

    Deep-water dumpsite 106 is located over the continental slope and rise about 106 miles east of New Jersey (Fig. 1). From March 1986 to July 1992 approximately eight million wet metric tons of sewage sludge were disposed there per year (Robertson and Redford, 1992). Because of the potential impact of this dumping on deep-sea ecosystems, NOAA/NMFS contracted the Virginia Institute of Marine Science, College of William and Mary (VIMS) to make otter-trawl collections in and around DWD 106 in 1990 and 1991. Specific objectives of these collections were: 1. To sample the demersal megafauna, especially fishes; 2. To provide frozen series of dominant species of fishes and invertebrates to NMFS for subsequent heavy metal and organic analyses; 3. To provide a description of the demersal fish community structure in the area; 4. To compare these data with similar data collected from 1973 to 1978 from DWD 106 and an adjacent area to the south near Norfolk submarine canyon. This final contract report summarizes the results of these studies

    Paul F. Lazarsfeld\u27s theory of uses and gratifications looks at why people chose to use or not use the media that is available to them. It was first used with the technology of radio and eventually applied to various other forms of media technologies such as comic books, television, and the Internet. In this study, the researcher expands uses and gratifications research to include one of the newest communication technologies -- e-books. Participants of the study completed a survey in regards to their electronic and print reading habits, and the results of the survey along with quantitative data found in press releases from various e-book companies answers four research question in regards to who uses e-books, why people chose to use or not use e-books, and how the sales of e-books differ in comparison to traditional print books

    Einfluss von Spontanatmung auf die endexspiratorische Belüftung der Lunge bei tierexperimentellem Lungenschaden

    Ziel dieser Arbeit war es, den Einfluss von erhaltener Spontanatmung unter APRV auf die Belüftung des Lungengewebes und das Lungenvolumen bei schwerem Lungenversagen zu untersuchen. Zudem sollte der Effekt einer veränderten Lungenbelüftung auf den Gasaustausch ermittelt werden. Hierzu wurden Schweine mit experimentell induziertem Lungenversagen maschinell im APRV Modus mit und ohne Erhalt der Spontanatmung beatmet. Die Belüftung des Lungengewebes, das Ausmaß an Atelektasen und das end-exspiratorische Lungenvolumen wurden dabei mittels densitometrischer Analysen computertomographischer Lungenaufnahmen und mithilfe der Stickstoffauswaschmethode ermittelt. Wir konnten zeigen, dass die erhaltene Spontanatmung unter APRV-Beatmung zu einer Erhöhung des endexspiratorischen Lungenvolumens (EELV) führt. Erhaltene Spontanatmung verhindert eine Kranialwertsverschiebung des Zwerchfells und erhöht so die kranio-kaudalen Lungendimensionen. Besonders in zwerchfellnahen Lungenregionen konnten wir zusätzlich zeigen, dass die Zunahme des EELV nicht durch Überblähung von bereits offenem Lungengewebe sondern durch Wiedereröffnung von Atelektasen, also durch Rekrutierung von nicht-belüftetem Lungengewebe zu erklären war. Unsere Untersuchungen lassen den Schluss zu, dass erhaltene Spontanatmung unter APRV durch die Steigerung des EELV das Ventilations-Perfusionsverhältnis verbessert und dadurch die arterielle Oxygenierung steigert. In Verbindung mit einer spontanatmungsbedingten globalen Blutflusssteigerung wird so das Sauerstoffangebot erhöht. Der aktiven Zwerchfellkontraktion während Spontanatmung kommt dabei offenbar eine entscheidende Rolle zu

    Diversity and Evolution of Sensor Histidine Kinases in Eukaryotes

    Histidine kinases (HKs) are primary sensor proteins that act in cell signaling pathways generically referred to as "two component systems" (TCSs). TCSs are among the most widely distributed transduction systems used by both prokaryotic and eukaryotic organisms to detect and respond to a broad range of environmental cues. The structure and distribution of HK proteins are now well documented in prokaryotes but information is still fragmentary for eukaryotes. Here, we have taken advantage of recent genomic resources to explore the structural diversity and the phylogenetic distribution of HKs in the prominent eukaryotic supergroups. Searches of the genomes of 67 eukaryotic species spread evenly throughout the phylogenetic tree of life identified 748 predicted HK proteins. Independent phylogenetic analyses of predicted HK proteins were carried out for each of the major eukaryotic supergroups. This allowed most of the compiled sequences to be categorised into previously described HK groups. Beyond the phylogenetic analysis of eukaryotic HKs, this study revealed some interesting findings: (i) characterisation of some previously undescribed eukaryotic HK groups with predicted functions putatively related to physiological traits; (ii) discovery of HK groups that were previously believed to be restricted to a single kingdom in additional supergroups and (iii) indications that some evolutionary paths have led to the appearance, transfer, duplication, and loss of HK genes in some phylogenetic lineages. This study provides an unprecedented overview of the structure and distribution of HKs in the Eukaryota and represents a first step towards deciphering the evolution of TCS signaling in living organisms
