The proposed set of tools will be integrated into an intelligent framework for collaborative book authoring and experiencing called SmartBook.     Easy Creation of Semantics-Enhanced Digital Artwork Collections

    In this paper we propose an approach for cost-effective employing of semantic technologies to improve the efficiency of searching and browsing of digital artwork collections. It is based on a semi-automatic creation of a Topic Map-based virtual art gallery portal by using existing Topic Maps tools. Such a ‘cheap’ solution could enable small art museums or art-related educational programs that lack sufficient funding for software development and publication infrastructure to take advantage of the emerging semantic technologies.     Pharmacokinetics, Tissue Distribution and Therapeutic Effect of Cationic Thermosensitive Liposomal Doxorubicin Upon Mild Hyperthermia

    Purpose: To evaluate pharmacokinetic profile, biodistribution and therapeutic effect of cationic thermosensitive liposomes (CTSL) encapsulating doxorubicin (Dox) upon mild hyperthermia (HT). Methods: Non-targeted thermosensitive liposomes (TSL) and CTSL were developed, loaded with Dox and characterized. Blood kinetics and biodistribution of Dox-TSL and Dox-CTSL were followed in B16BL6 tumor bearing mice upon normothermia (NT) or initial hyperthermia conditions. Efficacy study in B16BL6 tumor bearing mice was followed with Dox-TSL or Dox-CTSL upon NT or HT. Efficacy study in LLC tumor bearing mice was performed upon two HT conditions. Intravital microscopy was performed on B16BL6 tumors implanted in dorsal-skin fold window-bearing mice. Results: Targeting did not cause faster blood clearance of CTSL compared to TSL. Highest uptake of liposomes was observed in spleen, kidneys and liver. Applying HT prior to CTSL administration increased drug delivery to the tumor and CTSL delivered ∼1.7 fold higher Dox concentration compared to TSL. Efficacy in B16BL6 murine melanoma showed that HT had a significant effect on CTSL in tumor suppression and prolonged survival. Efficacy in LLC Lewis lung carcinoma tumor model demonstrates that two HT treatments hold promises for a successful treatment option. Conclusion: CTSL have potency to increase drug efficacy in tumors due to their targeted and drug release functions

    The aim of this PhD thesis is to analyze the medical care of adult women in Germany suf-fering from uncomplicated urinary tract infections, as these are the most common bacterial infections encountered in ambulatory care settings. Acute uncomplicated cystitis is one of the most common indications for prescribing of an-timicrobials to otherwise healthy community-dwelling women. Antimicrobial use, whether appropriate or inappropriate, leads to a continuing increase in antimicrobial resistance. The use of administrative data from statutory health insurance funds offers great opportu-nities to assess information about the antibiotic prescribing and it may allow concordance with prescribing guidelines to be assessed. First, computerized literature searches and a review of international MEDLINE-published literature concerning uncomplicated urinary tract infections and antimicrobial resistance in uropathogens have been conducted. Second, epidemiological data were collected as a basis for the following analyses. Based on diagnosis claims data 7,3 % of the BARMER GEK women population, aged 12 years and older, had at least once an urinary tract infection in the year 2013, 1,7 % had at least once cystitis and 0,16 % had at least once pyelonephritis. Despite published national and international guidelines for the optimal selection of an an-timicrobial agent and the duration of therapy, this study demonstrated a wide variation in prescribing practices. Most patients with urinary tract infection were not treated according to the current guidelines. A broad regional variability in antimicrobial prescribing among the German federal states has been manifested, particularly in the prescribing rates of fluoroquinolones. The prescribing rates of fluoroquinolones for cystitis varied 2013 among the different age groups between 27 and 40 %. An optimal approach to therapy must include consideration of antimicrobial resistance and collateral damage such as the selection of drug-resistant organisms and colonization or infection with multidrug-resistant organisms. This could be achieved by evidence based guidelines and a constant monitoring of the antimicrobial prescribing. Informational pro-grams should be initiated to raise the physiciansa awareness of the problem of antimicrobial resistance and improve their guideline adherence

    Tumour basement membrane laminin expression predicts outcome following curative resection of pancreatic head cancer

    Background: Although widely fragmented BMs have been associated with adverse outcome in several cancer types, comparatively little is known with respect to its effect on the prognosis of pancreatic cancer. The aim of the current study was therefore to determine the prognostic value of tumour basement membrane (BM) continuity in two anatomically closely related, however, prognostically different tumours, pancreatic head-and periampullary cancer. Methods: Tumour BM continuity was determined by immunohistochemical staining of its two major components, laminin and collagen type IV. Associations were made with recurrence free survival (RFS), cancer-specific survival (CSS), overall survival (OS) and conventional prognostic factors. Results: Fifty-nine and 61% of pancreatic head and periampullary tumours, respectively, showed limited BM laminin expression. Whereas 43% and 41% of pancreatic head and periampullary cancers, respectively, showed limited BM collagen type IV expression. Limited BM laminin was associated with poor outcome following curative resection of pancreatic head cancer (P=0.034, 0.013 and 0.017 for RFS, CSS and OS, respectively). Two and a half times as many patients with 25% BM laminin were recurrence free and alive 5 years following resection compared with those with limited BM laminin. Although staining patterns of both BM components were weakly correlated with each other, BM collagen type IV expression was not significantly associated with outcome in either tumour type.Conclusion: Discontinuous BMs, determined by laminin expression, are associated with poor outcome following curative resection of pancreatic head cancer
