    Existing motor state is favored at the expense of new movement during 13-35 Hz oscillatory synchrony in the human corticospinal system

    Oscillations in local field potentials in the β-frequency band (13-35 Hz) are a pervasive feature of human and nonhuman primate motor cortical areas. However, the function of such synchronous activity across populations of neurons remains unknown. Here, we test the hypothesis that β activity may promote existing motor set and posture while compromising processing related to new movements. Three experiments were performed. First, healthy subjects were instructed to make reaction time movements of the outstretched index finger in response to imperative cues triggered by transient increases in corticospinal synchrony, as evidenced by phasic elevations of β-frequency band microtremor and intermuscular synchrony. Second, healthy subjects were instructed to resist a stretch to the index finger triggered in the same way. Finger acceleration in the reaction time task and transcortical components of the stretch reflex were measured and compared with those elicited by random cue or stretch presentation. Finally, we sought a correlation between finger acceleration in the reaction time task and cortical synchrony directly measured from the electrocorticogram in two patients undergoing functional neurosurgery. We demonstrate that movements are slowed and transcortical responses to stretch are potentiated during periods of elevated β-band cortical synchrony. The results suggest that physiological periods of β synchrony are associated with a cortical state in which postural set is reinforced, but the speed of new movements impaired. The findings are of relevance to Parkinson's disease, in which subcortical and cortical β-band synchronization is exaggerated in the setting of increased tone and slowed movements

    A molecular biology and phase II trial of lapatinib in children with refractory CNS malignancies: a pediatric brain tumor consortium study.

    High expression of ERBB2 has been reported in medulloblastoma and ependymoma; EGFR is amplified and over-expressed in brainstem glioma suggesting these proteins as potential therapeutic targets. We conducted a molecular biology (MB) and phase II study to estimate inhibition of tumor ERBB signaling and sustained responses by lapatinib in children with recurrent CNS malignancies. In the MB study, patients with recurrent medulloblastoma, ependymoma, and high-grade glioma (HGG) undergoing resection were stratified and randomized to pre-resection treatment with lapatinib 900 mg/m(2) dose bid for 7-14 days or no treatment. Western blot analysis of ERBB expression and pathway activity in fresh tumor obtained at surgery estimated ERBB receptor signaling inhibition in vivo. Drug concentration was simultaneously assessed in tumor and plasma. In the phase II study, patients, stratified by histology, received lapatinib continuously, to assess sustained response. Eight patients, on the MB trial (four medulloblastomas, four ependymomas), received a median of two courses (range 1-6+). No intratumoral target inhibition by lapatinib was noted in any patient. Tumor-to-plasma ratios of lapatinib were 10-20 %. In the 34 patients (14 MB, 10 HGG, 10 ependymoma) in the phase II study, lapatinib was well-tolerated at 900 mg/m(2) dose bid. The median number of courses in the phase II trial was two (range 1-12). Seven patients (three medulloblastoma, four ependymoma) remained on therapy for at least four courses range (4-26). Lapatinib was well-tolerated in children with recurrent or CNS malignancies, but did not inhibit target in tumor and had little single agent activity.Fil: Fouladi, Maryam.     Effects of an Unusual Poison Identify a Lifespan Role for Topoisomerase 2 in Saccharomyces Cerevisiae

    A progressive loss of genome maintenance has been implicated as both a cause and consequence of aging. Here we present evidence supporting the hypothesis that an age-associated decay in genome maintenance promotes aging in Saccharomyces cerevisiae (yeast) due to an inability to sense or repair DNA damage by topoisomerase 2 (yTop2). We describe the characterization of LS1, identified in a high throughput screen for small molecules that shorten the replicative lifespan of yeast. LS1 accelerates aging without affecting proliferative growth or viability. Genetic and biochemical criteria reveal LS1 to be a weak Top2 poison. Top2 poisons induce the accumulation of covalent Top2-linked DNA double strand breaks that, if left unrepaired, lead to genome instability and death. LS1 is toxic to cells deficient in homologous recombination, suggesting that the damage it induces is normally mitigated by genome maintenance systems. The essential roles of yTop2 in proliferating cells may come with a fitness trade-off in older cells that are less able to sense or repair yTop2-mediated DNA damage. Consistent with this idea, cells live longer when yTop2 expression levels are reduced. These results identify intrinsic yTop2-mediated DNA damage as a potentially manageable cause of aging

    Cognitive function and mood at high altitude following acclimatization and use of supplemental oxygen and adaptive servoventilation sleep treatments.

    Impairments in cognitive function, mood, and sleep quality occur following ascent to high altitude. Low oxygen (hypoxia) and poor sleep quality are both linked to impaired cognitive performance, but their independent contributions at high altitude remain unknown. Adaptive servoventilation (ASV) improves sleep quality by stabilizing breathing and preventing central apneas without supplemental oxygen. We compared the efficacy of ASV and supplemental oxygen sleep treatments for improving daytime cognitive function and mood in high-altitude visitors (N = 18) during acclimatization to 3,800 m. Each night, subjects were randomly provided with ASV, supplemental oxygen (SpO2 > 95%), or no treatment. Each morning subjects completed a series of cognitive function tests and questionnaires to assess mood and multiple aspects of cognitive performance. We found that both ASV and supplemental oxygen (O2) improved daytime feelings of confusion (ASV: p < 0.01; O2: p < 0.05) and fatigue (ASV: p < 0.01; O2: p < 0.01) but did not improve other measures of cognitive performance at high altitude. However, performance improved on the trail making tests (TMT) A and B (p < 0.001), the balloon analog risk test (p < 0.0001), and the psychomotor vigilance test (p < 0.01) over the course of three days at altitude after controlling for effects of sleep treatments. Compared to sea level, subjects reported higher levels of confusion (p < 0.01) and performed worse on the TMT A (p < 0.05) and the emotion recognition test (p < 0.05) on nights when they received no treatment at high altitude. These results suggest that stabilizing breathing (ASV) or increasing oxygenation (supplemental oxygen) during sleep can reduce feelings of fatigue and confusion, but that daytime hypoxia may play a larger role in other cognitive impairments reported at high altitude. Furthermore, this study provides evidence that some aspects of cognition (executive control, risk inhibition, sustained attention) improve with acclimatization

    Mapping aerial metal deposition in metropolitan areas from tree bark : a case study in Sheffield, England

    We investigated the use of metals accumulated on tree bark for mapping their deposition across metropolitan Sheffield by sampling 642 trees of three common species. Mean concentrations of metals were generally an order of magnitude greater than in samples from a remote uncontaminated site. We found trivially small differences among tree species with respect to metal concentrations on bark, and in subsequent statistical analyses did not discriminate between them. We mapped the concentrations of As, Cd and Ni by lognormal universal kriging using parameters estimated by residual maximum likelihood ({\sc reml}). The concentrations of Ni and Cd were greatest close to a large steel works, their probable source, and declined markedly within 500~metres of it and from there more gradually over several kilometres. Arsenic was much more evenly distributed, probably as a result of locally mined coal burned in domestic fires for many years. Tree bark seems to integrate airborne pollution over time, and our findings show that sampling and analysing it are cost-effective means of mapping and identifying sources

    A Survey of the Helminth Parasites of Pheasants from Brown County, South Dakota

    This paper presents a study of the helminth endoparasite of the Chinese or ring-necked pheasant (Phasianus colchicus). The digestive and respiratory tracts of 262 pheasants, all of which were collected in Brown County, South Dakota, were examined. Collections were ,made from January, 1961, through November of the same year. The problem was divided as follows: (1) locating and Identifying the helminth parasites, (2) relating parasitism to the age of the birds, (3) ascertaining the seasonal distribution of the parasites, and (4) determining what gross pathological changes were due to parasitism. Previous to this study, little or no literature was available regarding the pheasant parasites of the state, and few systemic investigations of this nature had been undertaken in any region. Most of the reports from areas in the United State where the pheasant has become established limit the discussion of parasitism to isolated notations of occurrence or to host lists. Thus, this study was made in order to obtain a record of the occurrence, incidence and effect on the host by pheasant parasites in South Dakota

    Greening Capitalism? A Marxist Critique of Carbon Markets

    Climate change is increasingly being recognized as a serious threat to dominant modes of social organization, inspiring suggestions that capitalism itself needs to be transformed if we are to ‘decarbonize’ the global economy. Since the Kyoto Protocol in 1997, carbon markets have emerged as the main politico-economic tools in global efforts to address climate change. Newell and Paterson (2010) have recently claimed that the embrace of carbon markets by financial and political elites constitutes a possible first step towards the transformation of current modes of capitalist organization into a new form of greener, more sustainable ‘climate capitalism.’ In this paper, we argue that the institutionalization of carbon markets does not, in fact, represent a move towards the radical transformation of capitalism, but is better understood as the most recent expression of ongoing trends of ecological commodification and expropriation, driving familiar processes of uneven and crisis-prone development. In this paper, we review four critical Marxist concepts: metabolic rift (Foster, 1999), capitalism as world ecology (Moore, 2011a), uneven development and accumulation through dispossession (Harvey, 2003, 2006), and sub-imperialism (Marini, 1972, 1977), developing a framework for a Marxist analysis of carbon markets. Our analysis shows that carbon markets form part of a longer historical development of global capitalism and its relation to nature. Carbon markets, we argue, serve as creative new modes of accumulation, but are unlikely to transform capitalist dynamics in ways that might foster a more sustainable global economy. Our analysis also elucidates, in particular, the role that carbon markets play in exacerbating uneven development within the Global South, as elites in emerging economies leverage carbon market financing to pursue new strategies of sub-imperial expansion. </jats:p

    Characterizations of how species mediate ecosystem properties require more comprehensive functional effect descriptors

    The importance of individual species in mediating ecosystem process and functioning is generally accepted, but categorical descriptors that summarize species-specific contributions to ecosystems tend to reference a limited number of biological traits and underestimate the importance of how organisms interact with their environment. Here, we show how three functionally contrasting sediment-dwelling marine invertebrates affect fluid and particle transport - important processes in mediating nutrient cycling - and use high-resolution reconstructions of burrow geometry to determine the extent and nature of biogenic modification. We find that individual functional effect descriptors fall short of being able to adequately characterize how species mediate the stocks and flows of important ecosystem properties and that, in contrary to common practice and understanding, they are not substitutable with one another because they emphasize different aspects of species activity and behavior. When information derived from these metrics is combined with knowledge of how species behave and modify their environment, however, detailed mechanistic information emerges that increases the likelihood that a species functional standing will be appropriately summarized. Our study provides evidence that more comprehensive functional effect descriptors are required if they are to be of value to those tasked with projecting how altered biodiversity will influence future ecosystems
