Electrochemical Boron-Doped Diamond Film Microcells Micromachined with Femtosecond Laser: Application to the Determination of Water Framework Directive Metals
Planar electrochemical microcells were micromachined in a microcrystalline boron-doped diamond (BDD) thin layer using a femtosecond laser (Photo 1). The electrochemical performances of the new laser-machined BDD microcell were assessed by differential pulse anodic stripping voltammetry (DPASV) determinations, at nM level, of the four heavy metal ions of the European Water Framework Directive (WFD): Cd(II), Ni(II), Pb(II), Hg(II). The results are compared with those of previously published BDD electrodes [1]. The calculated detection limits are 0.4 nM, 6.8 nM and 5.5 nm 2.3 nM, and the linearities go up to 35nM, 97nM, 48nM and 5nM for respectively Cd(II), Ni(II) Pb(II) and Hg(II). The detection limits meet with the environmental quality standard of the WFD for three of the four metals. It was shown that the four heavy metals could be detected simultaneously, in the concentration ratio usually measured in sewage or runoff waters

Assessment of aerobic biodegradation of lower-chlorinated benzenes in contaminated groundwater using field-derived microcosms and compound-specific carbon isotope fractionation
Biodegradation of lower chlorinated benzenes (tri-, di- and monochlorobenzene) was assessed at a coastal aquifer contaminated with multiple chlorinated aromatic hydro- carbons. Field-derived microcosms, established with groundwater from the source zone and amended with a mixture of lower chlorinated benzenes, evidenced biodegradation of monochlorobenzene (MCB) and 1,4-dichlorobenzene (1,4-DCB) in aerobic microcosms, whereas the addition of lactate in anaerobic microcosms did not enhance anaerobic reduc- tive dechlorination. Aerobic microcosms established with groundwater from the plume consumed several doses of MCB and concomitantly degraded the three isomers of dichloroben- zene with no observable inhibitory effect. In the light of these results, we assessed the applicability of compound stable isotope analysis to monitor a potential aerobic remediation treatment of MCB and 1,4-DCB in this site. The carbon isotopic fractionation factors ( ε) obtained from field-derived microcosms were -0.7 ¿ ± 0.1 ¿ and -1.0 ¿ ± 0.2 ¿ for MCB and 1,4-DCB, respectively. For 1,4-DCB, the carbon isotope fractionation during aerobic biodegra- dation was reported for the first time. The weak carbon isotope fractionation values for the aerobic pathway would only allow tracing of in situ degradation in aquifer parts with high extent of biodegradation. However, based on the carbon isotope effects measured in this and previous studies, relatively high carbon isotope shifts (i.e., δ13 C > 4.0 ¿ ) of MCB or 1,4- DCB in contaminated groundwater would suggest that their biodegradation is controlled by anaerobic reductive dechlorination
Assessing forage research and education needs of organic dairy farms in the United States
The viability of organic dairy operations in the United States (US) relies on forage production. The objectives of this study were to (1) assess producer and farm information regarding current forage production practices and producer knowledge gaps and (2) identify forage research and educational needs of organic dairy producers across the US. A survey was distributed to 643 organic dairy producers across the US, with 165 respondents (26% response rate). A focus group consisting of extension professionals, university researchers and staff, consultants, dairy industry representatives and organic dairy producers was also consulted for forage research needs. Results showed that approximately half (51%) of surveyed producers were somewhat satisfied with their forage production systems and sometimes experienced negative weather-related impacts on forage yield and quality. A majority (64%) of producers felt their knowledge to meet farm goals was adequate but they reported a lack of resources to implement this knowledge especially for balancing high-forage diets and selecting soil amendments. This study revealed that 54% of producers rely on peer experiences as information resources to make decisions on forage programs. Producer knowledge gaps included pasture renovation with reduced or no-tillage, forage mixtures that match their needs, and forage management practices aiming for high-quality forage. Based on the survey and focus group findings, forage research and educational activities should foster climate change resilience regarding forage diversity adapted to local and regional climatic conditions, improve forage quality, enhance economic returns from soil fertility amendments and pasture renovation, and introduce new forages and forage mixtures that suit economical, agronomical, and environmental needs

The market has price sensitive consumers. The developed model has been demonstrated on two power systems: only diesel genset and a wind-diesel hybrid system showing the impact of the generation costs and capacity of generation from wind on the tariff levels, expected profit and reliability

Telomerase activity in human leukemic cells with or without monosomy 7 or 7q-
BACKGROUND: In bone marrow material from patients with various leukemias we noted that samples with either a deletion on the long arm of one chromosome 7 (7q-) or a monosomy 7 had a higher telomerase activity. Considering that introduction of a chromosome 7 into a cancer cell line had been reported to eliminate telomerase activity, that 7q- is a common negative prognostic finding in cancers, and that the deleted segment (band 7q31) contains an unidentified tumor suppressor gene, we wondered if this gene might be a telomerase inhibitor. RESULTS: We found no significant difference in telomerase activity between the three groups of patient samples. In contrast to reports on tumor cell lines we observed no amplification of the telomerase genes. METHODS: We analyzed telomerase activity and copy number of the telomerase genes hTERT and hTR in frozen archival bone marrow samples from leukemia patients with a referral diagnosis of AML, and either a monosomy for chromosome 7, a deletion on the long arm of chromosome 7 (7q-), or none of these aberrations. Telomerase activity was measured with a commercially available kit, and the copy number of the telomerase genes was tested by FISH. CONCLUSIONS: We found no evidence of a telomerase inhibitor in band 7q31. The lack of telomerase gene amplification found in cell lines from solid tumors could reflect that this amplification is a property of solid tumors, not of hematological cancers
