Finite-element analysis of a piled embankment with reinforcement and subsoil
In 2014, the authors of this paper considered predictions of reinforcement tension in a piled embankment from British standard BS 8006 published in 2010 and the 2012 amended version, and compared the results with finite-element model predictions. In keeping with BS 8006, any contribution from the subsoil beneath the embankment was ignored. The present paper extends that earlier work by also considering the potentially beneficial contribution of a lightly overconsolidated clay subsoil layer, both in the finite-element predictions and as a simple modification to the BS 8006 predictive method. It is assumed that there is no ‘working platform’ (granular) material below the pile cap level, and that the water table in the subsoil does not drop, since either of these factors would be likely to significantly reduce the ability of the subsoil to carry load from the embankment. As anticipated, the subsoil support reduces reinforcement tension. When compared to the finite-element results the proposed modified BS 8006 prediction is quite accurate. The BSI 2012 modified prediction is ‘best’ (but sometimes slightly unconservative), whereas the BSI 2010 modified prediction is conservative in all cases considered. </jats:p
Alemtuzumab pre-conditioning with tacrolimus monotherapy in pediatric renal transplantation
We employed antibody pre-conditioning with alemtuzumab and posttransplant immunosuppression with low-dose tacrolimus monotherapy in 26 consecutive pediatric kidney transplant recipients between January 2004 and December 2005. Mean recipient age was 10.7 ± 5.8 years, 7.7% were undergoing retransplantation, and 3.8% were sensitized, with a PRA >20%. Mean donor age was 32.8 ± 9.2 years. Living donors were utilized in 65% of the transplants. Mean cold ischemia time was 27.6 ± 6.4 h. The mean number of HLA mismatches was 3.3 ± 1.3. Mean follow-up was 25 ± 8 months. One and 2 year patient survival was 100% and 96%. One and 2 year graft survival was 96% and 88%. Mean serum creatinine was 1.1 ± 0.6 mg/dL, and calculated creatinine clearance was 82.3 ± 29.4 mL/min/1.73 m 2. The incidence of pre-weaning acute rejection was 11.5%; the incidence of delayed graft function was 7.7%. Eighteen (69%) of the children were tapered to spaced tacrolimus monotherapy, 10.5 ± 2.2 months after transplantation. The incidence of CMV, PTLD and BK virus was 0%; the incidence of posttransplant diabetes was 7.7%. Although more follow-up is clearly needed, antibody pre-conditioning with alemtuzumab and tacrolimus monotherapy may be a safe and effective regimen in pediatric renal transplantation. © 2007 The Authors
Breast cancer histologic grading using digital microscopy: concordance and outcome association
Aims: Virtual microscopy utilising digital whole slide imaging (WSI) is increasingly used in breast pathology. Histologic grade is one of the strongest prognostic factors in breast cancer (BC). This study aims at investigating the agreement between BC grading using traditional light microscopy (LM) and digital whole slide imaging (WSI) with consideration of reproducibility and impact on outcome prediction.
Methods: A large (n=1675) well-characterised cohort of BC originally graded by LM was re-graded using WSI. Two separate virtual-based grading sessions (V1 and V2) were performed with a three months washout period. Outcome was assessed using breast cancer specific and distant metastasis free survival.
Results: The concordance between LM grading and WSI was strong (LM/SWI Cramer’s V: V1=0.576, and V2=0.579). The agreement regarding grade components was as follows: Tubule formation=0.538, Pleomorphism=0.422 and Mitosis=0.514. Greatest discordance was observed between adjacent grades whereas high/low grade discordance was uncommon (1.5%). The intra-observer agreement for the two WSI sessions was substantial for grade (V1/V2 Cramer’s V=0.676; kappa=0.648) and grade components (Cramer’s V T=0.628, P=0.573 and M=0.580). Grading using both platforms showed strong association with outcome (All p-value <0.001). Although mitotic scores assessed using both platforms were strongly associated with outcome, WSI tends to underestimate mitotic counts.
Conclusions: Virtual microscopy is a reliable and reproducible method for assessing BC histologic grade. Regardless of the observer or assessment platform, histologic grade is a significant predictor of outcome. Continuing advances in imaging technology could potentially provide improved performance of WSI BC grading and in particular mitotic count assessment
Targeting BRCA1-BER deficient breast cancer by ATM or DNA-PKcs blockade either alone or in combination with cisplatin for personalized therapy
BRCA1, a key factor in homologous recombination repair may also regulate base excision repair (BER). Targeting BRCA1-BER deficient cells by blockade of ATM and DNA-PKcs could be a promising strategy in breast cancer. We investigated BRCA1, XRCC1 and pol β protein expression in two cohorts (n=1602 sporadic and n=50 germ-line BRCA1 mutated) and mRNA expression in two cohorts (n=1952 and n=249). Artificial neural network analysis for BRCA1-DNA repair interacting genes was conducted in 249 tumours. Pre-clinically, BRCA1 proficient and deficient cells were DNA repair expression profiled and evaluated for synthetic lethality using ATM and DNA-PKcs inhibitors either alone or in combination with cisplatin. In human tumours, BRCA1 negativity was strongly associated with low XRCC1, and low pol β at mRNA and protein levels (p<0.0001). In patients with BRCA1 negative tumours, low XRCC1 or low pol β expression was significantly associated with poor survival in univariate and multivariate analysis compared to high XRCC1 or high pol β expressing BRCA1 negative tumours (ps<0.05). Pre-clinically, BRCA1 negative cancer cells exhibit low mRNA and low protein expression of XRCC1 and pol β. BRCA1-BER deficient cells were sensitive to ATM and DNA-PKcs inhibitor treatment either alone or in combination with cisplatin and synthetic lethality was evidenced by DNA double strand breaks accumulation, cell cycle arrest and apoptosis. We conclude that XRCC1 and pol β expression status in BRCA1 negative tumours may have prognostic significance. BRCA1-BER deficient cells could be targeted by ATM or DNA-PKcs inhibitors for personalized therapy
Challenges of Profile Likelihood Evaluation in Multi-Dimensional SUSY Scans
Statistical inference of the fundamental parameters of supersymmetric
theories is a challenging and active endeavor. Several sophisticated algorithms
have been employed to this end. While Markov-Chain Monte Carlo (MCMC) and
nested sampling techniques are geared towards Bayesian inference, they have
also been used to estimate frequentist confidence intervals based on the
profile likelihood ratio. We investigate the performance and appropriate
configuration of MultiNest, a nested sampling based algorithm, when used for
profile likelihood-based analyses both on toy models and on the parameter space
of the Constrained MSSM. We find that while the standard configuration is
appropriate for an accurate reconstruction of the Bayesian posterior, the
profile likelihood is poorly approximated. We identify a more appropriate
MultiNest configuration for profile likelihood analyses, which gives an
excellent exploration of the profile likelihood (albeit at a larger
computational cost), including the identification of the global maximum
likelihood value. We conclude that with the appropriate configuration MultiNest
is a suitable tool for profile likelihood studies, indicating previous claims

Mass extinctions and supernova explosions
A nearby supernova (SN) explosion could have negatively influenced life on
Earth, maybe even been responsible for mass extinctions. Mass extinction poses
a significant extinction of numerous species on Earth, as recorded in the
paleontologic, paleoclimatic, and geological record of our planet. Depending on
the distance between the Sun and the SN, different types of threats have to be
considered, such as ozone depletion on Earth, causing increased exposure to the
Sun's ultraviolet radiation, or the direct exposure of lethal x-rays. Another
indirect effect is cloud formation, induced by cosmic rays in the atmosphere
which result in a drop in the Earth's temperature, causing major glaciations of
the Earth. The discovery of highly intensive gamma ray bursts (GRBs), which
could be connected to SNe, initiated further discussions on possible
life-threatening events in Earth's history. The probability that GRBs hit the
Earth is very low. Nevertheless, a past interaction of Earth with GRBs and/or
SNe cannot be excluded and might even have been responsible for past extinction

Live to cheat another day: bacterial dormancy facilitates the social exploitation of beta-lactamases
The breakdown of antibiotics by β-lactamases may be cooperative, since resistant cells can detoxify their environment and facilitate the growth of susceptible neighbours. However, previous studies of this phenomenon have used artificial bacterial vectors or engineered bacteria to increase the secretion of β-lactamases from cells. Here, we investigated whether a broad-spectrum β-lactamase gene carried by a naturally occurring plasmid (pCT) is cooperative under a range of conditions. In ordinary batch culture on solid media, there was little or no evidence that resistant bacteria could protect susceptible cells from ampicillin, although resistant colonies could locally detoxify this growth medium. However, when susceptible cells were inoculated at high densities, late-appearing phenotypically susceptible bacteria grew in the vicinity of resistant colonies. We infer that persisters, cells that have survived antibiotics by undergoing a period of dormancy, founded these satellite colonies. The number of persister colonies was positively correlated with the density of resistant colonies and increased as antibiotic concentrations decreased. We argue that detoxification can be cooperative under a limited range of conditions: if the toxins are bacteriostatic rather than bacteridical; or if susceptible cells invade communities after resistant bacteria; or if dormancy allows susceptible cells to avoid bactericides. Resistance and tolerance were previously thought to be independent solutions for surviving antibiotics. Here, we show that these are interacting strategies: the presence of bacteria adopting one solution can have substantial effects on the fitness of their neighbours
Central Exclusive Production in QCD
We investigate the theoretical description of the central exclusive
production process, h1+h2 -> h1+X+h2. Taking Higgs production as an example, we
sum logarithmically enhanced corrections appearing in the perturbation series
to all orders in the strong coupling. Our results agree with those originally
presented by Khoze, Martin and Ryskin except that the scale appearing in the
Sudakov factor, mu=0.62 \sqrt{\hat{s}}, should be replaced with
mu=\sqrt{\hat{s}}, where \sqrt{\hat{s}} is the invariant mass of the centrally
produced system. We confirm this result using a fixed-order calculation and
show that the replacement leads to approximately a factor 2 suppression in the
