    An otoprotective role for the apoptosis inhibitor protein survivin

    Hearing impairment caused by ototoxic insults, such as noise or gentamicin is a worldwide health problem. As the molecular circuitries involved are not yet resolved, current otoprotective therapies are rather empirical than rational. Here, immunohistochemistry and western blotting showed that the cytoprotective protein survivin is expressed in the human and guinea pig cochlea. In the guinea pig model, moderate noise exposure causing only a temporary hearing impairment transiently evoked survivin expression in the spiral ligament, nerve fibers and the organ of Corti. Mechanistically, survivin upregulation may involve nitric oxide (NO)-induced Akt signaling, as enhanced expression of the endothelial NO synthase and phosphorylated Akt were detectable in some surviving-positive cell types. In contrast, intratympanic gentamicin injection inducing cell damage and permanent hearing loss correlated with attenuated survivin levels in the cochlea. Subsequently, the protective activity of the human and the guinea pig survivin orthologs against the ototoxin gentamicin was demonstrated by ectopic overexpression and RNAi-mediated depletion studies in auditory cells in vitro. These data suggest that survivin represents an innate cytoprotective resistor against stress conditions in the auditory system. The pharmacogenetic modulation of survivin may thus provide the conceptual basis for the rational design of novel therapeutic otoprotective strategies

    Measurement of the inclusive and differential Higgs boson production cross sections in the decay mode to a pair of τ Leptons in pp collisions at sqrt[s]=13  TeV

    Measurements of the inclusive and differential fiducial cross sections of the Higgs boson are presented, using the τ lepton decay channel. The differential cross sections are measured as functions of the Higgs boson transverse momentum, jet multiplicity, and transverse momentum of the leading jet in the event, if any. The analysis is performed using proton-proton collision data collected with the CMS detector at the LHC at a center-of-mass energy of 13  TeV and corresponding to an integrated luminosity of 138  fb^{-1}. These are the first differential measurements of the Higgs boson cross section in the final state of two τ leptons. In final states with a large jet multiplicity or with a Lorentz-boosted Higgs boson, these measurements constitute a significant improvement over measurements performed in other final states

    Search for a heavy resonance decaying into a top quark and a W boson in the lepton+jets final state at √ s = 13 TeV

A search for a heavy resonance decaying into a top quark and a W boson in proton-proton collisions at √s = 13 TeV is presented. The data analyzed were recorded with the CMS detector at the LHC and correspond to an integrated luminosity of 138 fb−1. The top quark is reconstructed as a single jet and the W boson, from its decay into an electron or muon and the corresponding neutrino. A top quark tagging technique based on jet clustering with a variable distance parameter and simultaneous jet grooming is used to identify jets from the collimated top quark decay. The results are interpreted in the context of two benchmark models, where the heavy resonance is either an excited bottom quark b∗ or a vector-like quark B. A statistical combination with an earlier search by the CMS Collaboration in the all-hadronic final state is performed to place upper cross section limits on these two models. The new analysis extends the lower range of resonance mass probed from 1.4 down to 0.7 TeV. For left-handed, right-handed, and vector-like couplings, b∗ masses up to 3.0, 3.0, and 3.2 TeV are excluded at 95% confidence level, respectively. The observed upper limits represent the most stringent constraints on the b∗ model to date.SCOAP3

    CMS pythia  8 colour reconnection tunes based on underlying-event data

New sets of parameter tunes for two of the colour reconnection models, quantum chromodynamics-inspired and gluon-move, implemented in the PYTHIA 8 event generator, are obtained based on the default CMS PYTHIA 8 underlying-event tune, CP5. Measurements sensitive to the underlying event performed by the CMS experiment at centre-of-mass energies s√=7 and 13TeV , and by the CDF experiment at 1.96TeV are used to constrain the parameters of colour reconnection models and multiple-parton interactions simultaneously. The new colour reconnection tunes are compared with various measurements at 1.96, 7, 8, and 13TeV including measurements of the underlying-event, strange-particle multiplicities, jet substructure observables, jet shapes, and colour flow in top quark pair (tt¯) events. The new tunes are also used to estimate the uncertainty related to colour reconnection modelling in the top quark mass measurement using the decay products of tt¯ events in the semileptonic channel at 13TeV.SCOAP3

The φη∗ observable, derived from angular measurements of the leptons and highly correlated with pT(ℓℓ) , is used to probe the low- pT(ℓℓ) region in a complementary way. Dilepton masses up to 1 TeV are investigated. Additionally, a measurement is performed requiring at least one jet in the final state. To benefit from partial cancellation of the systematic uncertainty, the ratios of the differential cross sections for various mℓℓ ranges to those in the Z mass peak interval are presented. The collected data correspond to an integrated luminosity of 36.3 fb-1 of proton–proton collisions recorded with the CMS detector at the LHC at a centre-of-mass energy of 13 TeV .     Evidence for WW/WZ vector boson scattering in the decay channel ℓνqq produced in association with two jets in proton-proton collisions at √s = 13 TeV

Evidence is reported for electroweak (EW) vector boson scattering in the decay channel ℓνqq of two weak vector bosons WV (V = W or Z), produced in association with two parton jets. The search uses a data set of proton-proton collisions at 13 TeV collected with the CMS detector during 2016–2018 with an integrated luminosity of 138 fb−1. Events are selected requiring one lepton (electron or muon), moderate missing transverse momentum, two jets with a large pseudorapidity separation and a large dijet invariant mass, and a signature consistent with the hadronic decay of a W/Z boson. The cross section is computed in a fiducial phase space defined at parton level requiring all parton transverse momenta pT > 10 GeV and at least one pair of outgoing partons with invariant mass mqq > 100 GeV. The measured and expected EW WV production cross sections are 1.90+0.53 −0.46 pb and 2.23+0.08 −0.11(scale) ± 0.05(PDF) pb, respectively, where PDF is the parton distribution function. The observed EW signal strength is μEW = 0.85 ± 0.12 (stat) +0.19 −0.17 (syst), corresponding to a signal significance of 4.4 standard deviations with 5.1 expected, and it is measured keeping the quantum chromodynamics (QCD) associated diboson production fixed to the standard model prediction. This is the first evidence of vector boson scattering in the ℓνqq decay channel at LHC. The simultaneous measurement of the EW and QCD associated diboson production agrees with the standard model prediction.SCOAP

    DPANN symbiont of Haloferax volcanii accelerates xylan degradation by the non-host haloarchaeon Halorhabdus sp.

    This study examines a natural consortium of halophilic archaea, comprising xylan-degrading Halorhabdus sp. SVX81, consortium cohabitant Haloferax volcanii SVX82 (formerly H. lucentense SVX82), and its DPANN ectosymbiont Ca. Nanohalococcus occultus SVXNc. Transcriptomics and targeted metabolomics demonstrated that the tripartite consortium outperformed individual and the Halorhabdus sp. SVX81 with H. volcanii SVX82 bipartite cultures in xylan degradation, exhibiting a division of labor: the DPANN symbiont processed glycolysis products, while other members performed xylan depolymerization and biosynthesis of essential compounds. Electron microscopy and cryo-electron tomography revealed the formation of heterocellular biofilms interlinked by DPANN cells. The findings demonstrated that DPANN symbionts can interact directly with other members of microbial communities, which are not their primary hosts, influencing their gene expression. However, DPANN proliferation requires their primary host presence. The study highlights the collective contribution of consortium members to xylan degradation and their potential for biotechnological applications in the management of hypersaline environments
