    Interaction between Yeast Cdc6 Protein and B-Type Cyclin/Cdc28 Kinases

    During purification of recombinant Cdc6 expressed in yeast, we found that Cdc6 interacts with the critical cell cycle, cyclin-dependent protein kinase Cdc28. Cdc6 and Cdc28 can be coimmunoprecipitated from extracts, Cdc6 is retained on the Cdc28-binding matrix p13-agarose, and Cdc28 is retained on an affinity column charged with bacterially produced Cdc6. Cdc6, which is a phosphoprotein in vivo, contains five Cdc28 consensus sites and is a substrate of the Cdc28 kinase in vitro. Cdc6 also inhibits Cdc28 histone H1 kinase activity. Strikingly, Cdc6 interacts preferentially with B-type cyclin/Cdc28 complexes and not Cln/Cdc28 in log-phase cells. However, Cdc6 does not associate with Cdc28 when cells are blocked at the restrictive temperature in a cdc34 mutant, a point in the cell cycle when the B-type cyclin/Cdc28 inhibitor p40Sic1 accumulates and purified p40Sic1 inhibits the Cdc6/Cdc28 interaction. Deletion of the Cdc28 interaction domain from Cdc6 yields a protein that cannot support growth. However, when overproduced, the mutant protein can support growth. Furthermore, whereas overproduction of wild-type Cdc6 leads to growth inhibition and bud hyperpolarization, overproduction of the mutant protein supports growth at normal rates with normal morphology. Thus, the interaction may have a role in the essential function of Cdc6 in initiation and in restraining mitosis until replication is complete

    The morphology of peroxide-cured, styrene crosslinked, bisphenol A-based vinyl ester (VE) resin was investigated by atomic force microscopy (AFM) after ‘physical’ etching with different methods. Etching was achieved by laser ablation, atmospheric plasma treatment and argon ion bombardment. Parameters of the etching were varied to get AFM scans of high topography resolution. VE exhibited a nanoscaled nodular structure the formation of which was ascribed to complex intra- and intermolecular reactions during crosslinking. The microstructure resolved after all the above physical etching techniques was similar provided that optimized etching and suitable AFM scanning conditions were selected. Nevertheless, with respect to the ‘morphology visualization’ these methods follow the power ranking: argon bombardment > plasma treatment > laser ablation

    Dissipative and nonaxisymmetric standard-MRI in Kepler disks

    Deviations from axial symmetry are necessary to maintain self-sustained MRI-turbulence. We define the parameters region where nonaxisymmetric MRI is excited and study dependence of the unstable modes structure and growth rates on the relevant parameters. We solve numerically the linear eigenvalue problem for global axisymmetric and nonaxisymmetric modes of standard-MRI in Keplerian disks with finite diffusion. For small magnetic Prandtl number the microscopic viscosity completely drops out from the analysis so that the stability maps and the growth rates expressed in terms of the magnetic Reynolds number Rm and the Lundquist number S do not depend on the magnetic Prandtl number Pm. The minimum magnetic field for onset of nonaxisymmetric MRI grows with Rm. For given S all nonaxisymmetric modes disappear for sufficiently high Rm. This behavior is a consequence of the radial fine-structure of the nonaxisymmetric modes resulting from the winding effect of differential rotation. It is this fine-structure which presents severe resolution problems for the numerical simulation of MRI at large Rm. For weak supercritical magnetic fields only axisymmetric modes are unstable. Nonaxisymmetric modes need stronger fields and not too fast rotation. If Pm is small its real value does not play any role in MRI.Comment: 4 pages, 6 figures, A&A Lette

    Landscape benefits of a forest conversion programme in North East Germany: results of a choice experiment

    The article describes the results of a choice experiment aimedat valuing landscape benefits of different kinds of forests in NE Germany by using computer generated images. Preferences for broadleaved/mixed forests over conifers amount to 40-85 € per year and household, additional visual diversity has a monetary value of about 20 €/a. This is true for the summer aspect of forests only. The same experiment conducted with winter images reveals no general preference for broadleaves, whereas visual diversity is valued even higher under winter conditions.The results are part of a study which aimed at valuing the impacts of a regional forest conversionprogramme. Beyond landscape value, the valuation has covered recreational value, the value for climate protection as well as timber production value. The development of landscape values over time can compensate for diminished timber returns until about 2080; afterwards the balance becomes negative. Carbon values are relatively minor in comparison to landscape and timber values

    Components of the ubiquitin-proteasome pathway compete for surfaces on Rad23 family proteins

    Background: The delivery of ubiquitinated proteins to the proteasome for degradation is a key step in the regulation of the ubiquitin-proteasome pathway, yet the mechanisms underlying this step are not understood in detail. The Rad23 family of proteins is known to bind ubiquitinated proteins through its two ubiquitin-associated (UBA) domains, and may participate in the delivery of ubiquitinated proteins to the proteasome through docking via the Rad23 ubiquitin-like (UBL) domain. Results: In this study, we investigate how the interaction between the UBL and UBA domains may modulate ubiquitin recognition and the delivery of ubiquitinated proteins to the proteasome by autoinhibition. We have explored a competitive binding model using specific mutations in the UBL domain. Disrupting the intramolecular UBL-UBA domain interactions in HHR23A indeed potentiates ubiquitin-binding. Additionally, the analogous surface on the Rad23 UBL domain overlaps with that required for interaction with both proteasomes and the ubiquitin ligase Ufd2. We have found that mutation of residues on this surface affects the ability of Rad23 to deliver ubiquitinated proteins to the proteasome. Conclusions: We conclude that the competition of ubiquitin-proteasome pathway components for surfaces on Rad23 is important for the role of the Rad23 family proteins in proteasomal targeting

    Helicity and alpha-effect by current-driven instabilities of helical magnetic fields

    Helical magnetic background fields with adjustable pitch angle are imposed on a conducting fluid in a differentially rotating cylindrical container. The small-scale kinetic and current helicities are calculated for various field geometries, and shown to have the opposite sign as the helicity of the large-scale field. These helicities and also the corresponding α\alpha-effect scale with the current helicity of the background field. The α\alpha-tensor is highly anisotropic as the components αϕϕ\alpha_{\phi\phi} and αzz\alpha_{zz} have opposite signs. The amplitudes of the azimuthal α\alpha-effect computed with the cylindrical 3D MHD code are so small that the operation of an αΩ\alpha\Omega dynamo on the basis of the current-driven, kink-type instabilities of toroidal fields is highly questionable. In any case the low value of the α\alpha-effect would lead to very long growth times of a dynamo in the radiation zone of the Sun and early-type stars of the order of mega-years.Comment: 6 pages, 7 figures, submitted to MNRA

    The History of Nuclidic Masses and of their Evaluation

    This paper is centered on some historical aspects of nuclear masses, and their relations to major discoveries. Besides nuclear reactions and decays, the heart of mass measurements lies in mass spectrometry, the early history of which will be reviewed first. I shall then give a short history of the mass unit which has not always been defined as one twelfth of the carbon-12 mass. When combining inertial masses from mass spectrometry with energy differences obtained in reactions and decays, the conversion factor between the two is essential. The history of the evaluation of the nuclear masses (actually atomic masses) is only slightly younger than that of the mass measurements themselves. In their modern form, mass evaluations can be traced back to 1955.     An updated bibliography and database on forest ecosystem service valuation studies in Austria, Germany and Switzerland

    This collection presents an updated bibliography of those empirical forest ecosystem service valuation studies in the German speaking countries which relate to demand oriented measures of the utility of public goods. The associated database (which is provided as a separately downloadable Excel-file) contains 77 data sets by October 2016. These data sets are characterised by about 40 descriptors (the exact number depending on the specific valuation method used in the respective study) which present details of the valuation object, the statistical and economic methods applied, the results of the valuation exercise, some other descriptors for study quality, and the associated publications.     Self-Pulsating Semiconductor Lasers: Theory and Experiment

    We report detailed measurements of the pump-current dependency of the self-pulsating frequency of semiconductor CD lasers. A distinct kink in this dependence is found and explained using rate-equation model. The kink denotes a transition between a region where the self-pulsations are weakly sustained relaxation oscillations and a region where Q-switching takes place. Simulations show that spontaneous emission noise plays a crucial role for the cross-over.Comment: Revtex, 16 pages, 7 figure
