    La clusterine: un nouveau régulateur de la voie NF- B et de la mort cellulaire Plusieurs fonctions physiologiques ont été attribuées à la clusterine (CLU), dont le rôle important au cours de l'inflammation et de la tumorogenèse. Nous avons montré que CLU interagit avec I b-a phosphorylé et diminue la translocation de p50/p65. Nous avons aussi généré un ensemble de constructions " flaggées " pour les différentes isoformes de CLU. Les résultats de cette étude montrent que la régulation de la voie NF- B pourrait être restreinte à des séquences particulières de CLU. Nous avons développe une nouvelle approche de ciblage de CLU par la technique d' exon skipping qui permet d'induire préférentiellement la forme nucléaire épissée du gène très mal caractérisée. Nous avons montré que la surexpression de cette forme par cette stratégie induit la mort cellulaire.Clusterin: a new regulator of NF-kappaB pathway and cell death Clusterin is a multifunctional protein that plays numerous roles in mammalian cells. By mean of transcriptomic analysis, we previously demonstrated that lower expression of clu both in tissues and cultured fibroblast-like synoviocytes of rheumatoid arthritis patients compared to osteoarthritic patients. We showed that CLU interacts with phospho-IkB-a and decreases the translocation of p50/p65 to the nucleus. To specify the interaction sites of CLU with its partners and to study the CLU isoforms roles, we generated several molecular constructs coding for various CLU regions of interest and test their role on NF- B pathway and CLU subcellular localization. We have also developed a new approach of "exon skipping" in order to induce preferential expression of the nuclear spliced form of the gene.     Breeding Habitat Requirements of Bonelli’s Eagle and Trends in Eastern Morocco: Implications for Conservation Planning

    The habitat requirements and biology of the nesting Bonelli’s Eagle (Aquila fasciata) in the Moulouya river gorges of southwestern Middle Atlas were studied from 2005–2015. Fifteen to fifty-eight Bonelli’s Eagle territories were occupied in one decade and produce in averaged 1.34 fledglings per occupied site. Population of Bonelli’s Eagles in the Moulouya river gorges seems healthy, though much of the apparent growth in population must be attributed to the increase in Houbara Buastard (Chlamydotis undata), Black-bellied Sandgrouse (Pterocles orientalis) and Barbary Partridge (Alectoris barbara) in an area of 80.000 km² near the study area were a conservation and hunting game reserve was established

    BACKGROUND: In prostate cancer the secreted form of clusterin (sCLU) has been described as an anti-apoptotic protein whose expression is increased after therapeutic intervention, whereas, the nuclear protein form nCLU was reported to have pro-apoptotic properties. METHODOLOGY: In order to provide new therapeutic approaches targeting CLU, we developed a strategy based on exon skipping by using a lentiviral construct to preferentially induce the nuclear spliced form of the protein. The molecular construct was transduced in LNCaP cells for testing the modulation of sensitivity of the transduced cells to pro-apoptotic stress. RESULTS AND CONCLUSIONS: We showed an increase of nCLU/sCLU expression ratio in the prostate cancer cell line "LNCaP" after lentiviral vector-U7 nCLU transduction. Moreover, we showed a significant inhibition of cell proliferation in nCLU-U7 LNCaP cells after treatment with cisplatin and after exposure to ionizing radiation compared to control cells. Finally, we showed that nCLU-U7 LNCaP cells exposure to UV-C significantly reduced an increase of cell death compared to control. Finally, we showed that modulating nCLU expression had profound impact on Ku70/Bax interaction as well as Rad17 expression which could be a key mechanism in sensitizing cells to cell death. In conclusion, this is the first report showing that increasing of nCLU/sCLU expression ratio by using an "on demand alternative splicing" strategy successfully increased sensitivity to radiotherapy and chemotherapy of prostate cancer cells
