What can managers learn online? Investigating possibilities for active understanding in the online MBA classroom
Online MBAs have become integral to business schools’ portfolios and the number of MBA students opting for an online version looks set to grow. In the wake of well documented critiques of traditional MBA formats, this expansion prompted us to examine the potential for critically reflexive learning ideals in asynchronous MBA learning environments. Building the Community of Inquiry (CoI) model we elaborate elements of Bakhtin and Shotter’s dialogism to develop the notion of ‘active understanding’ as a means to study an online MBA classroom. We present two illustrative episodes to show how aspects of active understanding may unfold and we point to the role of infrastructure, curriculum and instructor interventions in developing more genuine dialogical exchanges. Our findings suggest that online MBA course designers can learn from CoI approaches to which we add that critically reflexive learning is situationally sensitive; requiring the capacity to create and recognize nuance and difference in the written communication; making the other the focus of learning. We conclude with implications for pedagogy and technology infrastructure
Diphenyl diselenide, a simple glutathione peroxidase mimetic, inhibits human LDL oxidation in vitro
Oxidative modification of low-density lipoprotein (LDL) represents an important factor in atherogenesis. In the present study, we have investigated the antioxidant capability of diphenyl diselenide (PhSe)2, a simple organoseleno compound, against copper (Cu2+) and peroxyl radical-induced human LDL oxidation in vitro. In initial studies using human serum, (PhSe)2 caused a dose-dependent inhibition of Cu2+-induced lipid peroxidation, which was correlated to thiol consumption. (PhSe)2 increased lipid peroxidation lag phase and decreased lipid peroxidation rate in isolated human LDL, evaluated by measuring both conjugated diene (CD) and thiobarbituric acid reactive substances (TBARS) levels. Consistent with these observations, (PhSe)2 showed a marked inhibitory effect on 2,2-azobis(2-amidinopropane dihydrochloride) (AAPH)-induced oxidation of LDL or parinaric acid (PnA) incorporated into LDL. (PhSe)2 also displayed a dose-dependent protective effect against Cu2+-induced lipid peroxidation in rat aortic slices. Interestingly, besides the antioxidant effects of (PhSe)2 toward the lipid moieties of LDL, which was related to its thiol-peroxidase activity, protein moieties from human isolated LDL were also protected against Cu2+-induced oxidation. The results presented herein are the first to show that (i) (PhSe)2 inhibits lipid peroxidation in human isolated LDL in vitro, (ii) this phenomenon is related to its thiol-peroxidase activity, and (iii) this chalcogen also prevents the oxidation of protein moieties of human LDL. Probing formation of cargo/importin-α transport complexes in plant cells using a pathogen effector
Importin-αs are essential adapter proteins that recruit cytoplasmic proteins destined for active nuclear import to the nuclear transport machinery. Cargo proteins interact with the importin-α armadillo repeat domain via nuclear localization sequences (NLSs), short amino acids motifs enriched in Lys and Arg residues. Plant genomes typically encode several importin-α paralogs that can have both specific and partially redundant functions. Although some cargos are preferentially imported by a distinct importin-α it remains unknown how this specificity is generated and to what extent cargos compete for binding to nuclear transport receptors. Here we report that the effector protein HaRxL106 from the oomycete pathogen Hyaloperonospora arabidopsidis co-opts the host cell's nuclear import machinery. We use HaRxL106 as a probe to determine redundant and specific functions of importin-α paralogs from Arabidopsis thaliana. A crystal structure of the importin-α3/MOS6 armadillo repeat domain suggests that five of the six Arabidopsis importin-αs expressed in rosette leaves have an almost identical NLS-binding site. Comparison of the importin-α binding affinities of HaRxL106 and other cargos in vitro and in plant cells suggests that relatively small affinity differences in vitro affect the rate of transport complex formation in vivo. Our results suggest that cargo affinity for importin-α, sequence variation at the importin-α NLS-binding sites and tissue-specific expression levels of importin-αs determine formation of cargo/importin-α transport complexes in plant cells
A formal framework to prove the correctness of model driven engineering composition operators
International audienceCurrent trends in system engineering combine modeling, composition and verification technologies in order to harness their ever growing complexity. Each composition operator dedicated to a different modeling concern should be proven to be property preserving at assembly time. These proofs are usually burdensome with repetitive aspects. Our work targets the factorisation of these aspects relying on primitive generic composition operators used to express more sophisticated language specific ones. These operators are defined for languages expressed with OMG MOF metamodeling technologies. The proof are done with the Coq proof assistant relying on the Coq4MDE framework defined previously. These basic operators, Union and Substitution, are illustrated using the MOF Package Merge as composition operator and the preservation of model conformance as verified property
Phospholipase C2 Affects MAMP-Triggered Immunity by Modulating ROS Production
The activation of phosphoinositide-specific phospholipase C (PI-PLC) is one of the earliest responses triggered by the recognition of several microbe-associated molecular patterns (MAMPs) in plants. The Arabidopsis (Arabidopsis thaliana) PI-PLC gene family is composed of nine members. Previous studies suggested a role for PLC2 in MAMP-triggered immunity, as it is rapidly phosphorylated in vivo upon treatment with the bacterial MAMP flg22. Here, we analyzed the role of PLC2 in plant immunity using an artificial microRNA to silence PLC2 expression in Arabidopsis. We found that PLC2-silenced plants are more susceptible to the type III secretion system-deficient bacterial strain Pseudomonas syringae pv tomato (Pst) DC3000 hrcC− and to the nonadapted pea (Pisum sativum) powdery mildew Erysiphe pisi. However, PLC2-silenced plants display normal susceptibility to virulent (Pst DC3000) and avirulent (Pst DC3000 AvrRPM1) P. syringae strains, conserving typical hypersensitive response features. In response to flg22, PLC2-silenced plants maintain wild-type mitogen-activated protein kinase activation and PHI1, WRKY33, and FRK1 immune marker gene expression but have reduced reactive oxygen species (ROS)-dependent responses such as callose deposition and stomatal closure. Accordingly, the generation of ROS upon flg22 treatment is compromised in the PLC2-defficient plants, suggesting an effect of PLC2 in a branch of MAMP-triggered immunity and nonhost resistance that involves early ROS-regulated processes. Consistently, PLC2 associates with the NADPH oxidase RBOHD, suggesting its potential regulation by PLC2
Impacts of rumen fluid, refrigerated or reconstituted from a refrigerated pellet, on gas production measured at 24h of fermentation
3noRumen fluid is used as fresh inoculum for gas production fermentations to predict the nutritional value of feeds and rations for ruminants. However, collection of rumen fluid from animal donors is invasive, expensive, time consuming and results in fluids of variable quality. The general aim was to identify a procedure to manipulate rumen inoculum in order to facilitate its storage and transfer between laboratories. This strategy would also limit fluid collections from animals. Two experiments were completed based on gas production from graduated 100 mL glass syringe with five feeds as substrates. In experiment 1, the gas production and some fermentation parameters of fresh rumen fluids were compared with those preserved at 4 °C for 24, 48, 72 and 96 h. Refrigeration did not modify concentration of volatile fatty acids and pH, but ammonia in liquids refrigerated for 48–96 h was higher (P < 0.05) compared to fresh. In contrast, rumen fluid refrigeration for 24, 48 or 72 h did not depress gas production at 24 h, but it was lower at 96 h. In experiment 2, the rumen fluid was centrifugated at 13,000 x g and sedimented material (i.e., pellet) was refrigerated for 48 h at 4 °C. The asymptote of gas production kinetics from rumen fluid regenerated from the pellet was 8 % lower (P < 0.05) than that from fresh. However for 24 h gas production, the correlation between fresh liquid and pellet inoculum, calculated for five ingredients, was high (R2 = 0.94). Results support the use of rumen fluid preserved by refrigeration for up to 72 h, and rumen fluid reconstituted from refrigerated pellet, as an alternative to fresh. An RxLR effector from phytophthora infestans prevents re-localisation of two plant NAC transcription factors from the endoplasmic reticulum to the nucleus
The plant immune system is activated following the perception of exposed, essential and invariant microbial molecules that are recognised as non-self. A major component of plant immunity is the transcriptional induction of genes involved in a wide array of defence responses. In turn, adapted pathogens deliver effector proteins that act either inside or outside plant cells to manipulate host processes, often through their direct action on plant protein targets. To date, few effectors have been shown to directly manipulate transcriptional regulators of plant defence. Moreover, little is known generally about the modes of action of effectors from filamentous (fungal and oomycete) plant pathogens. We describe an effector, called Pi03192, from the late blight pathogen Phytophthora infestans, which interacts with a pair of host transcription factors at the endoplasmic reticulum (ER) inside plant cells. We show that these transcription factors are released from the ER to enter the nucleus, following pathogen perception, and are important in restricting disease. Pi03192 prevents the plant transcription factors from accumulating in the host nucleus, revealing a novel means of enhancing host susceptibility
First Measurement of the Transverse Spin Asymmetries of the Deuteron in Semi-Inclusive Deep Inelastic Scattering
First measurements of the Collins and Sivers asymmetries of charged hadrons
produced in deep-inelastic scattering of muons on a transversely polarized
6-LiD target are presented. The data were taken in 2002 with the COMPASS
spectrometer using the muon beam of the CERN SPS at 160 GeV/c. The Collins
asymmetry turns out to be compatible with zero, as does the measured Sivers

Keyframe Extraction in Endoscopic Video
In medical endoscopy more and more surgeons archive the recorded video streams in a long-term storage. One reason for this development, which is enforced by law in some countries, is to have evidence in case of lawsuits from patients. Another more practical reason is to allow later inspection of previous procedures and also to use parts of such videos for research and for training. However, due to the dramatic amount of video data recorded in a hospital on a daily basis, it is very important to have good preview images for these videos in order to allow for quick filtering of undesired content and for easier browsing through such a video archive. Unfortunately, common shot detection and keyframe extraction methods cannot be used for that video data, because these videos contain unedited and highly similar content, especially in terms of color and texture, and no shot boundaries at all. We propose a new keyframe extraction approach for this special video domain and show that our method is signi�cantly better than a previously proposed approach
Measurement of the Spin Structure of the Deuteron in the DIS Region
We present a new measurement of the longitudinal spin asymmetry A_1^d and the
spin-dependent structure function g_1^d of the deuteron in the range 1 GeV^2 <
Q^2 < 100 GeV^2 and 0.004< x <0.7. The data were obtained by the COMPASS
experiment at CERN using a 160 GeV polarised muon beam and a large polarised
6-LiD target. The results are in agreement with those from previous experiments
and improve considerably the statistical accuracy in the region 0.004 < x <
