Cation binding to 15-TBA quadruplex DNA is a multiple-pathway cation-dependent process
A combination of explicit solvent molecular dynamics simulation (30 simulations reaching 4 µs in total), hybrid quantum mechanics/molecular mechanics approach and isothermal titration calorimetry was used to investigate the atomistic picture of ion binding to 15-mer thrombin-binding quadruplex DNA (G-DNA) aptamer. Binding of ions to G-DNA is complex multiple pathway process, which is strongly affected by the type of the cation. The individual ion-binding events are substantially modulated by the connecting loops of the aptamer, which play several roles. They stabilize the molecule during time periods when the bound ions are not present, they modulate the route of the ion into the stem and they also stabilize the internal ions by closing the gates through which the ions enter the quadruplex. Using our extensive simulations, we for the first time observed full spontaneous exchange of internal cation between quadruplex molecule and bulk solvent at atomistic resolution. The simulation suggests that expulsion of the internally bound ion is correlated with initial binding of the incoming ion. The incoming ion then readily replaces the bound ion while minimizing any destabilization of the solute molecule during the exchange

Možnosti využití biologické ochrany lilku rajčete (Solanum lycopersicum) ve fóliovnících
In 2021, the occurrence of pests was observed in a foil plant for hydroponic tomato growing in the Ráječek Farm in Brno. The monitoring for Tuta absoluta (Meyrick, 1971) and visual inspection of the tomato growth took place from April to November by using yellow sticky traps and pheromone traps. The aim of the experiment was to determine the species spectrum and significance pests in the foil plant and their possible link to tomato varieties. The effect of used bioregulators was also evaluated. The application of bioagents took place once by the introduction of a polyphagous predator Macrolophus pygmaeus (Rambur, 1839). By using yellow sticky traps was found occurrence of Trialeurodes vaporariorum (Westwood, 1856) and species of the family Sciaridae. Visual inspection revealed the presence individuals of genus Liriomyza and the Aculops lycopersici (Tryon, 1917). No Tuta absoluta individuals were observed with pheromone traps. Adults of Trialeurodes vaporarium occurred predominantly in large truss varieties Tomicia and Plumola, individuals of genus Liriomyza were observed in mini varieties Belido and Maggino. The observed pests did not have a significant economic effect on the tomato growth
Polished Silver Solid Amalgam Electrode
The polished silver solid amalgam electrode (p-AgSAE) seems to be a suitable alternative to hanging mercury drop electrode (HMDE). The structure of electrode material and working electrode surface were analyzed by RTG diffraction, atomic forces microscopy (AFM) and scanning electron microscopy (SEM). It was found that silver amalgam has crystalline structure (without the presence of different phases), any liquid mercury is not presented on the electrode surface and also mercury is not evaporated from the amalgam. p-AgSAE was applied for the determination of inorganic cations (Cu2+, Pb2+, Cd2+, Zn2+) and anions (NO3-, SCN-), amino acids (cysteine, cystin), nitro compounds (2,4´-dinitritoluene, 1,3´-dinitrobenzene, 2-nitronaftalene), aldehydes (benzaldehyde), vitamin C and purine bases of DNA (adenine, guanine) up to now
