    Map learning in a pressure situation: the role of individual differences

    openL'apprendimento di mappe spaziali è un'operazione fondamentale per la corretta navigazione spaziale, premessa necessaria per l'orientamento e il movimento nello spazio che ci circonda nella quotidianità. Nonostante ciò, non è ancora ben chiaro come questa operazione venga influenzata da diversi fattori di stato (come le emozioni suscitate dall'ambiente) e di tratto (come l'età). L'obbiettivo di questo studio è valutare dunque come lo stress, l'ansia spaziale, le emozioni e le differenze individuali possano incidere sull'apprendimento di una mappa spaziale, in particolare in una situazione pressante. Per fare ciò, 177 persone divise in tre fasce d'età (18-34 anni, 35-49 anni e 50-69 anni) hanno appreso due mappe, una in situazione pressante e una in situazione di controllo, e a seguito di ciò veniva richiesto di ricordare in ordine i punti di riferimento ed effettuarne una stima di distanza. Prima dell'apprendimento delle mappe, sono stati somministrati un test di valutazione dell'ansia spaziale (Spatial self-assessments; SAS) e un test per valutare le emozioni tipicamente esperite (Positive and Negative Affect Schedule; PANAS). Inoltre, prima e dopo ogni apprendimento spaziale, è stata valutata l'attivazione e lo stato emotivo di ciascun partecipante tramite un affective slider. I risultati hanno riportato come significativo il ruolo dell'arousal: i partecipanti che sono risultati attivati hanno avuto prestazioni migliori nel compito di ricordo dell'ordine dei punti di riferimento, con un declino della prestazione del gruppo 50-69 anni se paragonato al gruppo 18-34 anni. Per quanto riguarda il compito di stima della distanza, risulta significativo il ruolo dell'ansia spaziale: i partecipanti che hanno riferito di provarne maggiormente hanno poi avuto una prestazione peggiore in questa tipologia di compito.Learning spatial maps is a fundamental operation for proper spatial navigation, a prerequisite for orientation and movement in the space around us in everyday life. Despite this, it is still not well understood how this operation is affected by various state factors (such as emotions aroused by the environment) and trait factors (such as age). Thus, the aim of this study is to evaluate how stress, spatial anxiety, emotions, and individual differences may affect spatial map learning, particularly in a pressing situation. To do this, 177 people divided into three age groups (18-34 years, 35-49 years and 50-69 years) learned two maps, one in a pressing situation and one in a control situation, and following this they were asked to remember landmarks in order and make a distance estimate. Before learning the maps, a spatial anxiety assessment test (Spatial self-assessments; SAS) and a test to assess typically experienced emotions (Positive and Negative Affect Schedule; PANAS) were administered. In addition, before and after each spatial learning, each participant's emotional activation and state was assessed using an affective slider. The results reported the role of arousal as significant: participants who were activated performed better in the landmark order memory task, with a decline in performance of the 50-69 year old group when compared to the 18-34 year old group. Regarding the distance estimation task, the role of spatial anxiety is significant: participants who reported experiencing more of it then performed worse on this type of task

    A First Evaluation of a Multi-Modal Learning System to Control Surgical Assistant Robots via Action Segmentation

    The next stage for robotics development is to introduce autonomy and cooperation with human agents in tasks that require high levels of precision and/or that exert considerable physical strain. To guarantee the highest possible safety standards, the best approach is to devise a deterministic automaton that performs identically for each operation. Clearly, such approach inevitably fails to adapt itself to changing environments or different human companions. In a surgical scenario, the highest variability happens for the timing of different actions performed within the same phases. This paper presents a cognitive control architecture that uses a multi-modal neural network trained on a cooperative task performed by human surgeons and produces an action segmentation that provides the required timing for actions while maintaining full phase execution control via a deterministic Supervisory Controller and full execution safety by a velocity-constrained Model-Predictive Controller

    Inhomogeneous Diastereomeric Composition of Mongersen Antisense Phosphorothioate Oligonucleotide Preparations and Related Pharmacological Activity Impairment

    Mongersen is a 21-mer antisense oligonucleotide designed to downregulate Mothers against decapentaplegic homolog 7 (SMAD7) expression to treat Crohn's disease. Mongersen was manufactured in numerous batches at different scales during several years of clinical development, which all appeared identical, using common physicochemical analytical techniques, while only phosphorous-31 nuclear magnetic resonance (P-31-NMR) in solution showed marked differences. Close-up analysis of 27 mongersen batches revealed marked differences in SMAD7 downregulation in a cell-based assay. Principal component analysis of P-31-NMR profiles showed strong correlation with SMAD7 downregulation and, therefore, with pharmacological efficacy in vitro. Mongersen contains 20 phosphorothioate (PS) linkages, whose chirality (Rp/Sp) was not controlled during manufacturing. A different diastereomeric composition throughout batches would lead to superimposable analytical data, but to distinct P-31-NMR profiles, as indeed we found. We tentatively suggest that this may be the origin of different biological activity. As similar manifolds are expected for other PS-based oligonucleotides, the protocol described here provides a general method to identify PS chirality issues and a chemometric tool to score each preparation for this elusive feature

    Ceftriaxone induced hemolysis complicated by acute renal failure

    Over the last decade, second and third generation cephalosporins have been the most common drugs causing hemolytic anemia (HA). Of these cases, 20% have been attributed to ceftriaxone. The clinical presentation of ceftriaxone-induced HA is usually abrupt with sudden onset of pallor, tachypnea, cardio-respiratory arrest and shock. Acute renal failure (ARF) has been reported in 41% of such cases with a high fatality rate. We report a pediatric patient with ARF complicating ceftriaxone-induced HA who survived. Ceftriaxone is a commonly used drug, and early recognition of HA and institution of supportive care, including dialysis is likely to improve the outcome.
