P. brasiliensis virulence is affected by SconC, the negative regulator of inorganic sulfur assimilation
Conidia/mycelium-to-yeast transition of Paracoccidioidesbrasiliensis is a critical step for the establishment of paracoccidioidomycosis, a systemic mycosis endemic in Latin America. Thus, knowledge of the factors that mediate this transition is of major importance for the design of intervention strategies. So far, the only known pre-requisites for the accomplishment of the morphological transition are the temperature shift to 37°C and the availability of organic sulfur compounds. In this study, we investigated the auxotrophic nature to organic sulfur of the yeast phase of Paracoccidioides, with special attention to P. brasiliensis species. For this, we addressed the role of SconCp, the negative regulator of the inorganic sulfur assimilation pathway, in the dimorphism and virulence of this pathogen. We show that down-regulation of SCONC allows initial steps of mycelium-to-yeast transition in the absence of organic sulfur compounds, contrarily to the wild-type fungus that cannot undergo mycelium-to-yeast transition under such conditions. However, SCONC down-regulated transformants were unable to sustain yeast growth using inorganic sulfur compounds only. Moreover, pulses with inorganic sulfur in SCONC down-regulated transformants triggered an increase of the inorganic sulfur metabolism, which culminated in a drastic reduction of the ATP and NADPH cellular levels and in higher oxidative stress. Importantly, the down-regulation of SCONC resulted in a decreased virulence of P. brasiliensis, as validated in an in vivo model of infection. Overall, our findings shed light on the inability of P. brasiliensis yeast to rely on inorganic sulfur compounds, correlating its metabolism with cellular energy and redox imbalances. A phase I randomized therapeutic MVA-B vaccination improves the magnitude and quality of the T cell immune responses in HIV-1-infected subjects on HAART
Trial Design
Previous studies suggested that poxvirus-based vaccines might be instrumental in the therapeutic
HIV field. A phase I clinical trial was conducted in HIV-1-infected patients on highly
active antiretroviral therapy (HAART), with CD4 T cell counts above 450 cells/mm3 and
undetectable viremia. Thirty participants were randomized (2:1) to receive either 3 intramuscular
injections of MVA-B vaccine (coding for clade B HIV-1 Env, Gag, Pol and Nef antigens)
or placebo, followed by interruption of HAART.
Methods
The magnitude, breadth, quality and phenotype of the HIV-1-specific T cell response were
assayed by intracellular cytokine staining (ICS) in 22 volunteers pre- and post-vaccination.
Results
MVA-B vaccine induced newly detected HIV-1-specific CD4 T cell responses and expanded
pre-existing responses (mostly against Gag, Pol and Nef antigens) that were high in magnitude,
broadly directed and showed an enhanced polyfunctionality with a T effector memory
(TEM) phenotype, while maintaining the magnitude and quality of the pre-existing HIV-1-
specific CD8 T cell responses. In addition, vaccination also triggered preferential CD8+ T
cell polyfunctional responses to the MVA vector antigens that increase in magnitude after
two and three booster doses
Increased cortical surface area and gyrification following long-term survival from early monocular enucleation
AbstractPurposeRetinoblastoma is typically diagnosed before 5 years of age and is often treated by enucleation (surgical removal) of the cancerous eye. Here, we sought to characterize morphological changes of the cortex following long-term survival from early monocular enucleation.MethodsNine adults with early right-eye enucleation (≤48 months of age) due to retinoblastoma were compared to 18 binocularly intact controls. Surface area, cortical thickness, and gyrification estimates were obtained from T1 weighted images and group differences were examined.ResultsEarly monocular enucleation was associated with increased surface area and/or gyrification in visual (i.e., V1, inferior temporal), auditory (i.e., supramarginal), and multisensory (i.e., superior temporal, inferior parietal, superior parietal) cortices compared with controls. Visual cortex increases were restricted to the right hemisphere contralateral to the remaining eye, consistent with previous subcortical data showing asymmetrical lateral geniculate nucleus volume following early monocular enucleation.ConclusionsAltered morphological development of visual, auditory, and multisensory regions occurs subsequent to long-time survival from early eye loss
Mechanisms of Hearing Loss after Blast Injury to the Ear
Given the frequent use of improvised explosive devices (IEDs) around the world, the study of traumatic blast injuries is of
increasing interest. The ear is the most common organ affected by blast injury because it is the bodyメs most sensitive
pressure transducer. We fabricated a blast chamber to re-create blast profiles similar to that of IEDs and used it to develop a
reproducible mouse model to study blast-induced hearing loss. The tympanic membrane was perforated in all mice after
blast exposure and found to heal spontaneously. Micro-computed tomography demonstrated no evidence for middle ear or
otic capsule injuries; however, the healed tympanic membrane was thickened. Auditory brainstem response and distortion
product otoacoustic emission threshold shifts were found to be correlated with blast intensity. As well, these threshold
shifts were larger than those found in control mice that underwent surgical perforation of their tympanic membranes,
indicating cochlear trauma. Histological studies one week and three months after the blast demonstrated no disruption or
damage to the intra-cochlear membranes. However, there was loss of outer hair cells (OHCs) within the basal turn of the
cochlea and decreased spiral ganglion neurons (SGNs) and afferent nerve synapses. Using our mouse model that
recapitulates human IED exposure, our results identify that the mechanisms underlying blast-induced hearing loss does not
include gross membranous rupture as is commonly believed. Instead, there is both OHC and SGN loss that produce auditory
dysfunction
Comparative Genomic Analysis of Human Fungal Pathogens Causing Paracoccidioidomycosis
Paracoccidioides is a fungal pathogen and the cause of paracoccidioidomycosis, a health-threatening human systemic mycosis endemic to Latin America. Infection by Paracoccidioides, a dimorphic fungus in the order Onygenales, is coupled with a thermally regulated transition from a soil-dwelling filamentous form to a yeast-like pathogenic form. To better understand the genetic basis of growth and pathogenicity in Paracoccidioides, we sequenced the genomes of two strains of Paracoccidioides brasiliensis (Pb03 and Pb18) and one strain of Paracoccidioides lutzii (Pb01). These genomes range in size from 29.1 Mb to 32.9 Mb and encode 7,610 to 8,130 genes. To enable genetic studies, we mapped 94% of the P. brasiliensis Pb18 assembly onto five chromosomes. We characterized gene family content across Onygenales and related fungi, and within Paracoccidioides we found expansions of the fungal-specific kinase family FunK1. Additionally, the Onygenales have lost many genes involved in carbohydrate metabolism and fewer genes involved in protein metabolism, resulting in a higher ratio of proteases to carbohydrate active enzymes in the Onygenales than their relatives. To determine if gene content correlated with growth on different substrates, we screened the non-pathogenic onygenale Uncinocarpus reesii, which has orthologs for 91% of Paracoccidioides metabolic genes, for growth on 190 carbon sources. U. reesii showed growth on a limited range of carbohydrates, primarily basic plant sugars and cell wall components; this suggests that Onygenales, including dimorphic fungi, can degrade cellulosic plant material in the soil. In addition, U. reesii grew on gelatin and a wide range of dipeptides and amino acids, indicating a preference for proteinaceous growth substrates over carbohydrates, which may enable these fungi to also degrade animal biomass. These capabilities for degrading plant and animal substrates suggest a duality in lifestyle that could enable pathogenic species of Onygenales to transfer from soil to animal hosts.National Institute of Allergy and Infectious Diseases (U.S.)National Institutes of Health. Gut Microbiota, Probiotics and Diabetes
Diabetes is a condition of multifactorial origin, involving several molecular mechanisms related to the intestinal
microbiota for its development. In type 2 diabetes, receptor activation and recognition by microorganisms from
the intestinal lumen may trigger inflammatory responses, inducing the phosphorylation of serine residues in insulin
receptor substrate-1, reducing insulin sensitivity. In type 1 diabetes, the lowered expression of adhesion proteins
within the intestinal epithelium favours a greater immune response that may result in destruction of pancreatic
β cells by CD8+ T-lymphocytes, and increased expression of interleukin-17, related to autoimmunity. Research in
animal models and humans has hypothesized whether the administration of probiotics may improve the prognosis
of diabetes through modulation of gut microbiota. We have shown in this review that a large body of evidence
suggests probiotics reduce the inflammatory response and oxidative stress, as well as increase the expression of
adhesion proteins within the intestinal epithelium, reducing intestinal permeability. Such effects increase insulin sensitivity and reduce autoimmune response. However, further investigations are required to clarify whether the administration of probiotics can be efficiently used for the prevention and management of diabetes
Inter-Effort Recovery Intermittent Hypoxia and Force Parameters in Sprint Interval Exercise
International Journal of Exercise Science 18(3): 672-685, 2025. Performing training sessions in hypoxia leads to a decrease in effort quality. The inter-effort recovery intermittent hypoxia model seems to ensure training session performance. The aim of this study is to investigate the effects of sprint interval exercise under normoxic (NOR), continuous hypoxic (HYP), and inter-effort hypoxic (IEH) conditions on force parameters. Seven swimmers (age: 26.04 ± 4.64 years) volunteered and performed one session of the 10 × 30-s all-out tethered swimming efforts with 4-min passive recovery intervals for each condition. Considering the simulated altitude at FiO2 = 0.13 and NOR at FiO2 = 0.209. The sessions were separated at least 48 hours. The peak force (PF), mean force (MF), impulse, fatigue index (FI) and percentage mean force relative to peak force (PFPERC) were determined for each effort. The force parameters were higher on IEH (PF: Δ = 21.32 N; MF: Δ = 9.65 N; impulse: Δ = 626.78 N∙s) and HYP (PF: Δ = 15.80 N; MF: Δ = 6.92 N; impulse: Δ = 621.77 N∙s) in relation of the NOR (p \u3c 0.001). PFPERC evidenced lower values in IEH in relation to HYP (Δ= 3.1 %, p = 0.017). The HYP and IEH condition can be considered a model that enhances performance in force parameters compared to the NOR condition
Gestión por procesos y los indicadores de desempeño en la entidad pública distrital de Bambas, 2022
The study presented as a general objective of this study is to determine the relationship between process management and indicators in the performance of its district municipality of Bambas, 2022, for which it was based on the basic type, with a quantitative approach, to present numerical results. , with non-experimental design - Correlational and cross-sectional scope, working with a population of 20 workers and the sample was made up of the same amount, selected in a non-probabilistic way at the discretion of the researcher, applying the survey as a technique and the questionnaire as an instrument, carrying out validation by 3 experts and reliability by Cronbach's Alpha method, obtaining a value of 0.896 for the process management questionnaire and 0.880 for the performance indicators questionnaire, reaching the conclusion: Significant relationship was determined ( Rho = 0.940) between process management and indicators in the performance of its district municipality l de Bambas, 2022, showing that the actions carried out in process management are beneficial for it to be achieved in compliance with the performance indicators. Laparoscopy in management of appendicitis in high-, middle-, and low-income countries: a multicenter, prospective, cohort study.
BACKGROUND: Appendicitis is the most common abdominal surgical emergency worldwide. Differences between high- and low-income settings in the availability of laparoscopic appendectomy, alternative management choices, and outcomes are poorly described. The aim was to identify variation in surgical management and outcomes of appendicitis within low-, middle-, and high-Human Development Index (HDI) countries worldwide. METHODS: This is a multicenter, international prospective cohort study. Consecutive sampling of patients undergoing emergency appendectomy over 6 months was conducted. Follow-up lasted 30 days. RESULTS: 4546 patients from 52 countries underwent appendectomy (2499 high-, 1540 middle-, and 507 low-HDI groups). Surgical site infection (SSI) rates were higher in low-HDI (OR 2.57, 95% CI 1.33-4.99, p = 0.005) but not middle-HDI countries (OR 1.38, 95% CI 0.76-2.52, p = 0.291), compared with high-HDI countries after adjustment. A laparoscopic approach was common in high-HDI countries (1693/2499, 67.7%), but infrequent in low-HDI (41/507, 8.1%) and middle-HDI (132/1540, 8.6%) groups. After accounting for case-mix, laparoscopy was still associated with fewer overall complications (OR 0.55, 95% CI 0.42-0.71, p < 0.001) and SSIs (OR 0.22, 95% CI 0.14-0.33, p < 0.001). In propensity-score matched groups within low-/middle-HDI countries, laparoscopy was still associated with fewer overall complications (OR 0.23 95% CI 0.11-0.44) and SSI (OR 0.21 95% CI 0.09-0.45). CONCLUSION: A laparoscopic approach is associated with better outcomes and availability appears to differ by country HDI. Despite the profound clinical, operational, and financial barriers to its widespread introduction, laparoscopy could significantly improve outcomes for patients in low-resource environments.
