    Will ocean acidification affect the digestive physiology and gut microbiota of whelk *Brunneifusus ternatanus*?

    To understand the physiological responses of the Brunneifusus ternatanus to future ocean acidification (OA), histology, enzyme activity and gut bacterial composition at different pH levels (Control : C group, pH 8.1; Exposure period : EP group, pH 7.3) for 28 days were studied under laboratory conditions. Microbiota composition was analyzed using 16S rRNA gene amplicon sequencing. Enzyme activities of trypsin (TRY), lipase (LPS), amylase (AMS), and lysozyme (LZM) were used as biochemical indicators, as well as weight gain rate (WGR), specific growth rate (SGR) as growth indicators. The stress caused by OA resulted in alterations to the intestine, including partially swollen and degranulated enterocytes and rough endoplasmic reticulum (RER). The relative abundance of the core phylum in the acidified group changed significantly, showing an increase in Tenericutes and a decrease in Proteobacteria. Firmicutes/Bacteroides ratio declined from 4.38 in the control group to 1.25 in the EP group. We found that the enzymes TRY, LPS, and AMS activities were inhibited at reduced pH, which was positively correlated with the dominant genera Mycoplasma and Bacteroides; while LZM activities showed a significant increment, but showing a strong negative correlation. Furthermore, both WG and SRG values showed a depression at low pH lever. These results suggest that if anthropogenic CO2 emissions continue to accelerate, OA could lead to a negative impact on the whelk health, also compromising their growth performance and even survival. These findings will benefit the future risk assessments of OA or other related emerging environmental issue

    Economic burden and its associated factors of hospitalized patients infected with A (H7N9) virus: a retrospective study in Eastern China, 2013–2014

    BACKGROUND: H7N9 continues to cause human infections and remains a pandemic concern. Understanding the economic impacts of this novel disease is important for making decisions on health resource allocation, including infectious disease prevention and control investment. However, there are limited data on such impacts. METHODS: Hospitalized laboratory-confirmed H7N9 patients or their families in Jiangsu Province of China were interviewed. Patients’ direct medical costs of hospitalization were derived from their hospital bills. A generalized linear model was employed to estimate the mean direct medical costs of patients with different characteristics. RESULTS: The mean direct cost of hospitalization for H7N9 was estimated to be ¥ 71 060 (95 % CI, 48 180–104 820), i.e., US10996(95 10 996 (95 % CI, 7 455–16 220), and was ¥12 060 (US 1 861), ¥136 120 (US21001)and¥218610(US 21 001) and ¥218 610 (US 33 728) for those who had mild or severe symptoms or who died, respectively. The principal components of the total fees differed among patients with different disease severity, although medication fees were always the largest contributors. Disease severity, proportion of reimbursement and family member monthly average income were identified as the key factors that contributed to a patient’s direct medical cost of hospitalization. CONCLUSIONS: The direct medical costs of hospitalized patients with H7N9 are significant, and far surpass the annual per capita income of Jiangsu Province, China. The influencing factors identified should be taken into account when developing related health insurance policies and making health resource allocation. TRIAL REGISTRATION: Not applicable.     Super-Resolution Imaging with Patchy Microspheres

    The diffraction limit is a fundamental barrier in optical microscopy, which restricts the smallest resolvable feature size of a microscopic system. Microsphere-based microscopy has proven to be a promising tool for challenging the diffraction limit. Nevertheless, the microspheres have a low imaging contrast in air, which hinders the application of this technique. In this work, we demonstrate that this challenge can be effectively overcome by using partially Ag-plated microspheres. The deposited Ag film acts as an aperture stop that blocks a portion of the incident beam, forming a photonic hook and an oblique near-field illumination. Such a photonic hook significantly enhanced the imaging contrast of the system, as experimentally verified by imaging the Blu-ray disc surface and colloidal particle arrays

    Does Serum Vitamin D Level Affect COVID-19 Infection and Its Severity?-A Case-Control Study

    Background: As effective medication to treat COVID-19 is currently unavailable, preventive remedies may be particularly important. Objective: To examine the relationship between serum 25-hydroxy vitamin D (25(OH)D) level and COVID-19 infection, its severity, and its clinical case characteristics. Methods: This case-control study compared serum 25(OH)D levels and rates of vitamin D deficiency (VDD) between 80 healthy controls and 62 patients diagnosed with COVID-19 and admitted to Guangxi People’s Hospital, China, 2/16/2020–3/16/2020. Cases were categorized into asymptomatic, mild/moderate, and severe/critical disease. Logistic regression analysis was conducted to examine the associations between 25(OH)D level, or VDD, and case status/severity of COVID-19 while controlling for demographics and comorbidities. A threshold level of vitamin D for conveying COVID-19 risk was estimated. Results: Severe/critical COVID-19 cases were significantly older and had higher percentages of comorbidity (renal failure) compared to mild cases. The serum 25(OH)D concentration in COVID-19 patient was much lower than that in healthy control. And 25(OH)D level was the lowest in severe/ critical cases, compared with mild cases. In further, significantly higher rates of VDD were found in COVID-19 cases (41.9%) compared to healthy controls (11.1%). And VDD was the greatest in severe/critical cases (80%), compared with mild cases (36%). These statistically significant associations remained even after controlling for demographics and comorbidities. A potential threshold of 25(OH)D (41.19nmol/L) to protect against COVID-19 was identified. Conclusion: Elderly and people with comorbidities were susceptible to severe COVID-19 infection. VDD was a risk factor for COVID-19, especially for severe/critical cases. While further confirmation is needed, vitamin D supplementation may have prevention or treatment potential for COVID- 19 disease

    Screening performance of COPD-PS scale, COPD-SQ scale, peak expiratory flow, and their combinations for chronic obstructive pulmonary disease in the primary healthcare in Haicang District, Xiamen City

    ObjectivesThis study aimed to evaluate the screening performance of COPD-PS questionnaire, COPD-SQ questionnaire, peak expiratory flow (PEF), COPD-PS questionnaire combined with PEF, and COPD-SQ questionnaire combined with PEF for chronic obstructive pulmonary disease (COPD).MethodsThis was a cross-sectional study. We distributed self-designed surveys and COPD screening scales (COPD-PS questionnaire and COPD-SQ questionnaire) to residents who underwent physical examination in five community health centers in Haicang District, Xiamen City, from February 2023 to May 2023, and measured their lung function and PEF with a portable device. We used logistic regression to obtain the coefficients of COPD-PS questionnaire, COPD-SQ questionnaire, and PEF, and plotted the receiver operating characteristic curves of each tool for diagnosing COPD and moderate-to-severe COPD. We evaluated and compared the optimal cut-off points and scores of sensitivity, specificity, Youden index, and area under the curve (AUC) values, and assessed the screening efficiency of different methods.ResultsOf the 3,537 residents who completed the COPD-SQ questionnaire, COPD-PS questionnaire, and spirometry, 840 were diagnosed with COPD. We obtained the coefficients of COPD-PS questionnaire combined with peak expiratory flow (PEF), and COPD-SQ questionnaire combined with PEF, by logistic regression as −0.479-0.358 × PEF +0.321 × COPD-PS score and − 1.286-0.315 × PEF +0.125 × COPD-SQ score, respectively. The sensitivity of diagnosing COPD by COPD-SQ questionnaire, COPD-PS questionnaire, PEF, COPD-PS questionnaire combined with PEF, and COPD-SQ questionnaire combined with PEF were 0.439, 0.586, 0.519, 0.586, 0.612 respectively, and the specificity were 0.725, 0.621, 0.688, 0.689, 0.663 respectively, with ROC values of 0.606 (95%CI: 0.586–0.626), 0.640 (0.619–0.661), 0.641 (0.619–0.663), 0.678 (0.657–0.699), 0.685 (0.664–0.706) respectively. The sensitivity of diagnosing GOLD II and above by COPD-SQ questionnaire, COPD-PS questionnaire, PEF, COPD-PS questionnaire combined with PEF, and COPD-SQ questionnaire combined with PEF were 0.489, 0.620, 0.665, 0.630, 0.781 respectively, and the specificity were 0.714, 0.603, 0.700, 0.811, 0.629 respectively, with ROC values of 0.631 (95%CI: 0.606–0.655), 0.653 (0.626–0.679), 0.753 (0.730–0.777), 0.784 (0.762–0.806), 0.766 (0.744–0.789) respectively.ConclusionOur study found that the accuracy of COPD screening by COPD-SQ questionnaire and COPD-PS questionnaire can be improved by combining the results of PEF. The screening performance of COPD-SQ questionnaire combined with PEF is relatively better. In future research, further studies are needed to optimize the performance of screening tools and understand whether their use will affect clinical outcomes

    Risk Assessment of Etanercept in Mice Chronically Infected With Toxoplasma gondii

    Toxoplasma gondii (T. gondii) is a zoonotic parasite that severely harms the health of the host. The cysts of T. gondii can reactivate from bradyzoites to tachyzoites, if the individual develops low or defective immunity, causing lethal toxoplasmosis. The host resists T. gondii infection by mediating Th1-type cellular immunity to generate pro-inflammatory cytokines. Tumor necrosis factor (TNF) is an important pro-inflammatory cytokine, which can induce lysosomal fusion of parasitophorous vacuole (PV) to kill parasites. Etanercept is a soluble TNF receptor fusion protein, which is widely used clinically to cure autoimmune diseases. The effects and specific molecular mechanisms of etanercept treatment on patients co-infected with autoimmune diseases and chronic toxoplasmosis are rarely reported. In our study, a mouse model of chronic infection with T. gondii and murine macrophages RAW264.7 cells infected with T. gondii were employed to investigate the impact of etanercept on the status of chronic infection. The cytokines levels and a series of phenotypic experiments in vivo and in vitro were measured. In the present study, the expression levels of TNF, IL-1β, and IL-6 were decreased and the brain cysts number was increased in mice chronically infected with T. gondii after being treated with etanercept. In vivo experiments confirmed that etanercept caused a decrease in the immune levels of the mice and activated the brain cysts, which would lead to conversion from chronic infection to acute infection, causing severe clinical and pathological symptoms. Murine macrophages RAW264.7 cells were pretreated with etanercept, and then infected with T. gondii. In vitro experiments, the expression levels of cytokines were decreased, indicating that etanercept could also reduce the cells’ immunity and promote the transformation of bradyzoites to tachyzoites, but did not affect the intracellular replication of tachyzoites. In summary, etanercept treatment could activate the conversion of bradyzoites to tachyzoites through reducing host immunity in vivo and in vitro. The results obtained from this study suggest that the use of etanercept in patients co-infected with autoimmune diseases and chronic toxoplasmosis may lead to the risk of activation of chronic infection, resulting in severe acute toxoplasmosis

    Hydrogen sulphide alleviates depression-like behaviors by suppressing the hippocampal necroptosis-neuroinflammation-KP imbalance axis

    BackgroundWe have previously demonstrated that hydrogen sulfide (H2S) is sufficient to attenuate depressive-like behavior induced by chronic unpredictable mild stress (CUMS), but its underlying mechanisms remain largely unknown. It is well known that necroptosis is a trigger of inflammation and neuroinflammation-induced kynurenine pathway (KP) imbalance is the main pathogenesis of depression. Meanwhile, H2S plays a pivotal role in the inhibition of inflammatory pathways. Hence, the present study sought to investigate whether the antidepressant effect of H2S is attributable to the inhibition of the axis of hippocampal necroptosis-neuroinflammation-KP imbalance.MethodsThe depression model of Sprague-Dawley (SD) rats was established by CUMS for four consecutive weeks. The expressions of necroptosis-related protein and KP-related protein were evaluated by Western blotting (WB). The levels of inflammatory factors were measured by enzyme-linked immunosorbent assay (ELISA). The hippocampal metabolites of tryptophan were determined by LC-MS/MS.ResultsIn this study, we found that H2S was capable of inhibiting necroptosis and neuroinflammation and correcting KP imbalance in the hippocampus of CUMS-exposed rats. The enhancement of necroptosis via overexpressing RIPK3 reversed the antagonistic role of H2S in the depressive-like behaviors of CUMS-exposed rats and the inhibitory effects of H2S on necroptosis, neuroinflammation, and KP imbalance in the hippocampus of CUMS-exposed rats. Furthermore, overexpressing indoleamine 2,3-dioxygenase 1 (IDO1) abolished the correcting effect of H2S on hippocampal KP imbalance and the attenuating effect of H2S on depressive-like behaviors in the CUMS-exposed rats.ConclusionThe antidepressant-like role of H2S is achieved by negatively regulating the axis of necroptosis-neuroinflammation-KP imbalance in the hippocampus

    Rapid profiling of carcinogenic types of Helicobacter pylori infection via deep learning analysis of label-free SERS spectra of human serum

    WHO classified Helicobacter pylori as a Group I carcinogen for gastric cancer as early as 1994. However, despite the high prevalence of H. pylori infection, only about 3 % of infected individuals eventually develop gastric cancer, with the highly virulent H. pylori strains expressing cytotoxin-associated protein (CagA) and vacuolating cytotoxin (VacA) being critical factors in gastric carcinogenesis. It is well known that H. pylori infection is divided into two types in terms of the presence and absence of CagA and VacA toxins in serum, that is, carcinogenic Type I infection (CagA+/VacA+, CagA+/VacA-, CagA-/VacA+) and non-carcinogenic Type II infection (CagA-/VacA-). Currently, detecting the two carcinogenic toxins in active modes is mainly done by diagnosing their serological antibodies. However, the method is restricted by expensive reagents and intricate procedures. Therefore, establishing a rapid, accurate, and cost-effective way for serological profiling of carcinogenic H. pylori infection holds significant implications for effectively guiding H. pylori eradication and gastric cancer prevention. In this study, we developed a novel method by combining surface-enhanced Raman spectroscopy with the deep learning algorithm convolutional neural network to create a model for distinguishing between serum samples with Type I and Type II H. pylori infections. This method holds the potential to facilitate rapid screening of H. pylori infections with high risks of carcinogenesis at the population level, which can have long-term benefits in reducing gastric cancer incidence when used for guiding the eradication of H. pylori infections

    Efficacy and safety of the compound Chinese medicine SaiLuoTong in vascular dementia: A randomized clinical trial

    Introduction: No licensed medications are available to treat vascular dementia (VaD). Methods: Patients were randomly assigned to experimental groups (SaiLuoTong [SLT] 360 or 240 mg for groups A and B for 52 weeks, respectively) or placebo group (SLT 360 mg and 240 mg for group C only from weeks 27 to 52, respectively). Results: Three hundred twenty-five patients were included in final analysis.
