    A versatile dual spot laser scanning confocal microscopy system for advanced fluorescence correlation spectroscopy analysis in living cell

    A fluorescence correlation spectroscopy (FCS) system based on two independent measurement volumes is presented. The optical setup and data acquisition hardware are detailed, as well as a complete protocol to control the location, size and shape of the measurement volumes. A method that allows to monitor independently the excitation and collection efficiency distribution is proposed. Finally, a few examples of measurements that exploit the two spots in static and/or scanning schemes, are reported.Comment: Accepted for publication in Review of Scientific Instrumen

    Microphotoluminescence study of disorder in ferromagnetic (Cd,Mn)Te quantum well

    Microphotoluminescence mapping experiments were performed on a modulation doped (Cd,Mn)Te quantum well exhibiting carrier induced ferromagnetism. The zero field splitting that reveals the presence of a spontaneous magnetization in the low-temperature phase, is measured locally; its fluctuations are compared to those of the spin content and of the carrier density, also measured spectroscopically in the same run. We show that the fluctuations of the carrier density are the main mechanism responsible for the fluctuations of the spontaneous magnetization in the ferromagnetic phase, while those of the Mn spin density have no detectable effect at this scale of observation.Comment: 4 pages, 3 figure

    Discrete complex analysis on planar quad-graphs

    We develop a linear theory of discrete complex analysis on general quad-graphs, continuing and extending previous work of Duffin, Mercat, Kenyon, Chelkak and Smirnov on discrete complex analysis on rhombic quad-graphs. Our approach based on the medial graph yields more instructive proofs of discrete analogs of several classical theorems and even new results. We provide discrete counterparts of fundamental concepts in complex analysis such as holomorphic functions, derivatives, the Laplacian, and exterior calculus. Also, we discuss discrete versions of important basic theorems such as Green's identities and Cauchy's integral formulae. For the first time, we discretize Green's first identity and Cauchy's integral formula for the derivative of a holomorphic function. In this paper, we focus on planar quad-graphs, but we would like to mention that many notions and theorems can be adapted to discrete Riemann surfaces in a straightforward way. In the case of planar parallelogram-graphs with bounded interior angles and bounded ratio of side lengths, we construct a discrete Green's function and discrete Cauchy's kernels with asymptotics comparable to the smooth case. Further restricting to the integer lattice of a two-dimensional skew coordinate system yields appropriate discrete Cauchy's integral formulae for higher order derivatives.Comment: 49 pages, 8 figure

    Matter Mixing in Aspherical Core-collapse Supernovae: Three-dimensional Simulations with Single Star and Binary Merger Progenitor Models for SN 1987A

    We perform three-dimensional hydrodynamic simulations of aspherical core-collapse supernovae focusing on the matter mixing in SN 1987A. The impacts of four progenitor (pre-supernova) models and parameterized aspherical explosions are investigated. The four pre-supernova models include a blue supergiant (BSG) model based on a slow merger scenario developed recently for the progenitor of SN 1987A (Urushibata et al. 2018). The others are a BSG model based on a single star evolution and two red supergiant (RSG) models. Among the investigated explosion (simulation) models, a model with the binary merger progenitor model and with an asymmetric bipolar-like explosion, which invokes a jetlike explosion, best reproduces constraints on the mass of high velocity 56^{56}Ni, as inferred from the observed [Fe II] line profiles. The advantage of the binary merger progenitor model for the matter mixing is the flat and less extended ρr3\rho \,r^3 profile of the C+O core and the helium layer, which may be characterized by the small helium core mass. From the best explosion model, the direction of the bipolar explosion axis (the strongest explosion direction), the neutron star (NS) kick velocity, and its direction are predicted.     Observation of strong-coupling effects in a diluted magnetic semiconductor (Ga,Fe)N

    A direct observation of the giant Zeeman splitting of the free excitons in (Ga,Fe)N is reported. The magnetooptical and magnetization data imply the ferromagnetic sign and a reduced magnitude of the effective p-d exchange energy governing the interaction between Fe^{3+} ions and holes in GaN, N_0 beta^(app) = +0.5 +/- 0.2 eV. This finding corroborates the recent suggestion that the strong p-d hybridization specific to nitrides and oxides leads to significant renormalization of the valence band exchange splitting.Comment: 4 pages, 2 figure

    Stool Xpert® MTB/RIF test for the diagnosis of childhood pulmonary tuberculosis at primary clinics in Zimbabwe.

    OBJECTIVE: To evaluate the diagnostic performance of Xpert® MTB/RIF on stool samples from children with clinical suspicion of pulmonary tuberculosis (PTB) at primary care clinics. DESIGN: A cross-sectional diagnostic evaluation enrolling 5-16 year olds from whom one induced sputum (IS) sample was tested for microbiological TB confirmation. Results of a single stool sample tested using Xpert were compared against microbiologically confirmed TB, defined as a positive result on sputum microscopy and/or culture and/or IS Xpert. RESULTS: Of 222 children enrolled, 218 had complete microbiological results. The median age was 10.6 years (interquartile range 8-13). TB was microbiologically confirmed in 19/218 (8.7%) children. Of these, respectively 5 (26%), 9 (47%) and 15 (79%) were smear-, culture- and IS Xpert-positive. Stool Xpert was positive in 13/19 (68%) microbiologically confirmed cases and 4/199 (2%) microbiologically negative cases. Stool Xpert detected 76.9% (10/13) of human immunodeficiency virus (HIV) infected and 50% (3/6) of non-HIV-infected children with microbiologically confirmed TB (P = 0.241). CONCLUSION: Stool Xpert is a potential alternative screening test for children with suspected TB if sputum is unavailable. Strategies to optimise the diagnostic yield of stool Xpert assay need further study

    Approximation of conformal mappings using conformally equivalent triangular lattices

    Consider discrete conformal maps defined on the basis of two conformally equivalent triangle meshes, that is edge lengths are related by scale factors associated to the vertices. Given a smooth conformal map ff, we show that it can be approximated by such discrete conformal maps fϵf^\epsilon. In particular, let TT be an infinite regular triangulation of the plane with congruent triangles and only acute angles (i.e.\ <π/2<\pi/2). We scale this tiling by ϵ>0\epsilon>0 and approximate a compact subset of the domain of ff with a portion of it. For ϵ\epsilon small enough we prove that there exists a conformally equivalent triangle mesh whose scale factors are given by logf\log|f'| on the boundary.     Acquired epidermodysplasia verruciformis due to multiple and unusual HPV infection among vertically-infected, HIV-positive adolescents in Zimbabwe.

    BACKGROUND: We have previously described the presentation of epidermodysplasia verruciformis (EV)-like eruptions in almost a quarter of hospitalized adolescents with vertically-acquired human immunodeficiency virus (HIV) infection in Harare, Zimbabwe, a region with a high prevalence of HIV infection. METHODS: We performed a clinical case note review and skin biopsy from affected sites in 4 HIV-infected adolescents with EV-like lesions in Harare. Biopsies were processed for histology and for human papillomavirus (HPV) typing. RESULTS: All patients had long-standing skin lesions that pre-dated the diagnosis of HIV by several years. The histology of skin biopsies from all patients was consistent with EV. In each biopsy, EV-associated β-HPV type 5 was identified (additionally, type 19 was found in 1 biopsy). Cutaneous wart-associated HPV types 1 and 2 were detected in all biopsies, together with genital lesion-associated HPV types 6, 16, and 52, (as well as ≥3 other genital lesion-associated HPV types). Despite immune reconstitution with combination antiretroviral therapy (cART), there was no improvement in EV-like lesions in any patient. CONCLUSIONS: EV is a disfiguring and potentially stigmatizing condition among this patient group and is difficult to treat; cART appears to have no impact on the progression of skin disease. Among adolescents with longstanding HIV-induced immunosuppression and with high levels of sun exposure, close dermatological surveillance for potential skin malignancy is required

    p-Type doping of II-VI heterostructures from surface states: application to ferromagnetic Cd1x_{1-x}Mnx_xTe quantum wells

    We present a study of p-type doping of CdTe and Cd1x_{1-x}Mnx_xTe quantum wells from surface states. We show that this method is as efficient as usual modulation doping with nitrogen acceptors, and leads to hole densities exceeding 2×10112 \times 10^{11} cm2^{-2}. Surface doping was successfully applied to obtain carrier-induced ferromagnetism in a Cd1x_{1-x}Mnx_xTe quantum well. The observed temperature dependence of photoluminescence spectra, and the critical temperature, correspond well to those previously reported for ferromagnetic quantum wells doped with nitrogen.Comment: 4 figure
