    Who Benefits Most from a University Degree?: A Cross-National Comparison of Selection and Wage Returns in the US, UK, and Germany

    Recent research on economic returns to higher education in the United States suggests that those with the highest wage returns to a college degree are least likely to obtain one. We extend the study of heterogeneous returns to tertiary education across multiple institutional contexts, investigating how the relationship between wage returns and the propensity to complete a degree varies by the level of expansion, differentiation, and cost of higher education. Drawing on panel data and matching techniques, we compare findings from the US with selection into degree completion in Germany and the UK. Contrary to previous studies, we find little evidence for population level heterogeneity in economic returns to higher education

    Hybrid Diffusion Imaging to Detect Acute White Matter Injury after Mild TBI

    poster abstractIntroduction: In the present study we used multi-shell Hybrid Diffusion Imaging (HYDI) to study white matter changes in the acute stage of mild traumatic brain injury (mTBI). Non-parametric diffusion analysis, q-space imaging as well as parametric analyses including conventional DTI and novel neurite orientation dispersion and density imaging (NODDI) were used to analyze the HYDI data. Method: Nineteen mTBI patients and 23 trauma-controlled subjects were recruited from the Emergency Department. Participants received T1W SPGR and HYDI in a Philips 3T Achieve TX scanner with 8-channel head coil and SENSE parallel imaging. The diffusion-weighting (DW) pulse sequence scan-time was about 24 min similar to (1). Results: Forty-eight WM ROIs were defined in the standard MNI space by intersecting subjects’ mean WM skeleton with WM atlas of Johns Hopkins University (JHU) ICBM-DTI-81(2). Linear model analysis was used to test the significance of diffusion metrics between mTBI and trauma-controlled groups with gender and age as covariates (model 3 in Table 1). Maps of DTI, q-space and NODDI diffusion metrics of an mTBI subject are shown in Figure 1. Among various diffusion metrics, only the NODDI derived parenchymal axonal density (Vic) was sensitive to mTBI with significant decreases in 60% of WM ROIs (Table 1). The mTBI subjects had an approximately 4% decrease in Vic. The affected WM tracts concentrated on pyramidal tracts and its cortical projections (bilateral corona radiatae). Most of the cerebella related tracts and hippocampal tracts are spared. Conclusion: HYDI and its diffusion metrics provide insights about microstructural changes of WM in the acute stage of mTBI and may prove useful as a marker of injury

    Has the 'Fast-Track' referral system affected the route of presentation and/or clinical outcomes in patients with colorectal cancer?

    Background: The aim of this study is to determine whether the 'Fast-Track' referral system has changed the route by which patients present with colorectal cancer (CRC) and whether the route of presentation has any effect on clinical outcome. Methods: A retrospective cohort study of patients diagnosed with CRC under the care of two consultant colorectal surgeons between April 2006 and December 2012. The route by which patients presented was categorised as Fast-Track (FT), non-Fast-Track (non-FT) or acute. Outcome variables were operative intent, disease stage and 2- and 5-year survival. Results: A total of 558 patients were identified. One hundred ninety-seven patients (35.3%) were referred as FT, 108 (19.4%) presented acutely and 253 patients (45.3%) presented via other routes (non-FT). Over the study period, the route of presentation did not change significantly (P=0.135). There was no significant difference between FT and non-FT groups in terms of the proportion of patients undergoing potentially curative surgery (70.6 vs 74.3%, P=0.092) or with node-negative disease (48.2 vs 52.2%, P=0.796) nor was there any difference in 2-year or 5-year survival (74.1 vs 73.9%, P=0.837 and 52.3 vs 53.8%, P=0.889, respectively). Patients who presented acutely were less likely to undergo curative resection, had more advanced disease and had worse 2- and 5-year survival. Conclusions: The Fast-Track referral system has not affected the route by which patients present with CRC nor has it had any effect on clinical outcomes. Alternative strategies are required if the desired improvement in outcomes is to be achieved

    Kinetic measurements to investigate the oxygen-sensing properties of plant cysteine oxidases

    Enzymatic O2 sensors transduce the availability of O2 within the cell into a physiological, typically adaptive response. One such O2-sensing enzymatic family is the N-terminal cysteine dioxygenases in plants (plant cysteine oxidases [PCOs]). In vitro kinetic studies have determined the O2-sensing capacity of PCOs. Here we describe the rationale and experimental protocol for an assay with which the O2 sensitivity of Arabidopsis thaliana PCOs (AtPCOs) can be measured. We explain each step from the recombinant protein synthesis of AtPCOs to the steady-state kinetic assays of AtPCOs for primary substrate and O2 from which kinetic parameters can be derived. The same techniques can be applied to other N-terminal cysteine thiol dioxygenases, e.g. 2-aminoethanethiol dioxygenase (ADO), and similar principles can be applied to determine kinetic characteristics of other oxygenase enzymes towards O2

    Measuring plant cysteine oxidase interactions with substrates using intrinsic tryptophan fluorescence

    Plant Cysteine Oxidases (PCOs) are oxygen-sensing enyzmes that catalyse oxidation of cysteinyl residues at the N-termini of target proteins, triggering their degradation via the N-degron pathway. PCO oxygen sensitivity means that in low oxygen conditions (hypoxia), their activity reduces and target proteins are stabilised. PCO substrates include Group VII Ethylene Response Factors (ERFVIIs) involved in adaptive responses to the acute hypoxia experienced upon plant submergence, as well as Little Zipper 2 (ZPR2) and Vernalisation 2 (VRN2) which are involved in developmental processes in hypoxic niches. The PCOs are potential targets for improving submergence tolerance through enzyme engineering or chemical treatment. To achieve this, a detailed understanding of their biological function is required. Here, we report development of an assay that exploits the intrinsic fluorescence of Arabidopsis thaliana PCO tryptophan residues. By using Ni(II)-substitued enzymes and preparing the assay under anaerobic conditions, tryptophan fluorescence quenching is observed on enzyme:substrate complex formation, allowing quantification of binding affinities. Our assay revealed that, broadly, AtPCO4 and AtPCO5 have stronger interactions with ERFVII substrates than ZPR2 and VRN2, suggesting ERFVIIs are primary targets of these enzymes. It also revealed a positive cooperative binding effect for interactions between AtPCOs4/5 and ERFVIIs and ZPR2. The assay is experimentally straightforward and can be used to further interogate PCO interactions with substrates

    Methylphenidate and Memory and Attention Adaptation Training for persistent cognitive symptoms after traumatic brain injury: a randomized, placebo-controlled trial

    The purpose of this multicenter, prospective, randomized, placebo-controlled study was to evaluate and compare the efficacy of two cognitive rehabilitation interventions (Memory and Attention Adaptation Training (MAAT) and Attention Builders Training (ABT)), with and without pharmacological enhancement (ie, with methylphenidate (MPH) or placebo), for treating persistent cognitive problems after traumatic brain injury (TBI). Adults with a history of TBI at least 4 months before study enrollment with either objective cognitive deficits or subjective cognitive complaints were randomized to receive MPH or placebo and MAAT or ABT, yielding four treatment combinations: MAAT/MPH (N=17), ABT/MPH (N=19), MAAT/placebo (N=17), and ABT/placebo (N=18). Assessments were conducted pre-treatment (baseline) and after 6 weeks of treatment (post treatment). Outcome measures included scores on neuropsychological measures and subjective rating scales. Statistical analyses used linear regression models to predict post-treatment scores for each outcome variable by treatment type, adjusting for relevant covariates. Statistically significant (PABT/placebo), nonverbal learning (MAAT/MPH>MAAT/placebo and MAAT/MPH>ABT/MPH), and auditory working memory and divided attention (MAAT/MPH>ABT/MPH). These results suggest that combined treatment with metacognitive rehabilitation (MAAT) and pharmacotherapy (MPH) can improve aspects of attention, episodic and working memory, and executive functioning after TBI
