    Effects of Low Carbohydrate High Protein (LCHP) diet on atherosclerotic plaque phenotype in ApoE/LDLR/ApoE/LDLR^{-/-} mice : FT-IR and Raman imaging

    Low Carbohydrate High Protein (LCHP) diet displays pro-atherogenic effects, however, the exact mechanisms involved are still unclear. Here, with the use of vibrational imaging, such as Fourier transform infrared (FT-IR) and Raman (RS) spectroscopies, we characterize biochemical content of plaques in Brachiocephalic Arteries (BCA) from ApoE/LDLR−/− mice fed LCHP diet as compared to control, recomended by American Institute of Nutrition, AIN diet. FT-IR images were taken from 6–10 sections of BCA from each mice and were complemented with RS measurements with higher spatial resolution of chosen areas of plaque sections. In aortic plaques from LCHP fed ApoE/LDLR−/− mice, the content of cholesterol and cholesterol esters was increased, while that of proteins was decreased as evidenced by global FT-IR analysis. High resolution imaging by RS identified necrotic core/foam cells, lipids (including cholesterol crystals), calcium mineralization and fibrous cap. The decreased relative thickness of the outer fibrous cap and the presence of buried caps were prominent features of the plaques in ApoE/LDLR−/− mice fed LCHP diet. In conclusion, FT-IR and Raman-based imaging provided a complementary insight into the biochemical composition of the plaque suggesting that LCHP diet increased plaque cholesterol and cholesterol esters contents of atherosclerotic plaque, supporting the cholesterol-driven pathogenesis of LCHP–induced atherogenesis

The selection of all submissions was subject to a double blind procedure with three reviews for each paper and an overall acceptance rate of 25 percent.     By adopting principles of cloud computing to the \ logistics domain the paradigm of Cloud Logistics is derived. It \ appears to be a promising paradigm in order to evolve logistics \ into being more flexible and collaborative. Yet, appropriate \ concepts that enable the cloud logistics paradigm are missing. \ In the paper, existing body of literature is reviewed and a \ definition and a framework of cloud logistics is given. Further, \ service blueprinting is combined with domain engineering and \ general morphological analysis in order to create a suitable \ method for designing cloud oriented service blueprints. Those \ are focusing on domain-specific flows and transformations \ enabling cloud oriented business collaboration. The method \ is applied to the logistics domain and a cloud logistics service \ blueprint is designed. Finally, the concept is evaluated with \ real use cases from logistics service providers

    Structure of rise in monthly temperature in Europe as estimated by machine learning

    The rise in air temperature is a leading research topic. This is not only from the cognitive point of view, but also for practical reasons because it involves many effects that are dangerous to humans and their activities. Although this is not a new issue, it requires continuous monitoring as well as the application of multiple methods, including the latest, apparently most objective methods offered by, inter alia, artificial intelligence. In the present paper, the authors have undertaken to investigate the structure of the rise in mean monthly air temperatures in Europe using unsupervised machine learning methods. The last 70 years can be divided into two periods, one of which is relatively stable and the second of which shows an evident rise in temperature. The correct determination of the year in which that change occurred is crucial. Mean monthly temperatures in Europe and its direct surroundings were used for this purpose. The data originated from 210 meteorological stations and covered the period 1951-2020. The analysis was performed using the hierarchical clustering and k-means clustering methods. The research was conducted in two phases. The first phase involved the analysis of area-average values, followed by the analysis of each station separately. Clear results were obtained, which confirms the usefulness of machine learning as a tool for monitoring temperature change. The quantitative change in the behavior of monthly temperature recorded from 1950 all over Europe is positioned at 1999, when the linear rise started

    Customers as service innovators within business networks

    Today there is still a gap between the theory of customer-driven business models and their implementation in practice. Furthermore the question emerges how companies could profit from a more sophisticated interaction with their customers. We want to introduce a concept of a technological platform which empowers the customer to co-create value with one or more companies by combining services. The main issue in this scenario is the changing of the business model from company-centric to customer-centric, i.e. the customer is the main driver for composing services and also the main driver for value creation. The company still has to deliver the value proposition to the customer who matches it with his own needs and then chooses a specific service from a portfolio of services. For this we analyze similar concepts and platforms found in literature and compare them to our approach. We finally conclude with an outlook on how can this concept be realized in practice

    Enhancing Roadway Safety: LiDAR-based Tree Clearance Analysis

    In the efforts for safer roads, ensuring adequate vertical clearance above roadways is of great importance. Frequently, trees or other vegetation is growing above the roads, blocking the sight of traffic signs and lights and posing danger to traffic participants. Accurately estimating this space from simple images proves challenging due to a lack of depth information. This is where LiDAR technology comes into play, a laser scanning sensor that reveals a three-dimensional perspective. Thus far, LiDAR point clouds at the street level have mainly been used for applications in the field of autonomous driving. These scans, however, also open up possibilities in urban management. In this paper, we present a new point cloud algorithm that can automatically detect those parts of the trees that grow over the street and need to be trimmed. Our system uses semantic segmentation to filter relevant points and downstream processing steps to create the required volume to be kept clear above the road. Challenges include obscured stretches of road, the noisy unstructured nature of LiDAR point clouds, and the assessment of the road shape. The identified points of non-compliant trees can be projected from the point cloud onto images, providing municipalities with a visual aid for dealing with such occurrences. By automating this process, municipalities can address potential road space constraints, enhancing safety for all. They may also save valuable time by carrying out the inspections more systematically. Our open-source code gives communities inspiration on how to automate the process themselves

    Removal of epididymal visceral adipose tissue prevents obesity-induced multi-organ insulin resistance in male mice

    Obesity is associated with insulin resistance, an important risk factor of type 2 diabetes, atherogenic dyslipidemia, and nonalcoholic fatty liver disease. The major purpose of this study was to test hypothesize that prophylactic removal of epididymal visceral adipose tissue (VAT) prevents obesity-induced multi-organ (liver, skeletal muscle, adipose tissue) insulin resistance. Accordingly, we surgically removed epididymal VAT pads from adult C57BL/6J mice and evaluated in vivo and cellular metabolic pathways involved in glucose and lipid metabolism following chronic high-fat diet (HFD) feeding. We found that VAT removal decreases HFD-induced body weight gain while increasing subcutaneous adipose tissue (SAT) mass. Strikingly, VAT removal prevents obesity-induced insulin resistance and hyperinsulinemia and markedly enhances insulin-stimulated AKT-phosphorylation at serine-473 (Ser473) and threonine-308 (Thr308) sites in SAT, liver, and skeletal muscle. VAT removal leads to decreases in plasma lipid concentrations and hepatic triglyceride (TG) content. In addition, VAT removal increases circulating adiponectin, a key insulin-sensitizing adipokine, whereas it decreases circulating interleukin 6, a pro-inflammatory adipokine. Consistent with these findings, VAT removal increases adenosine monophosphate-activated protein kinase C phosphorylation, a major downstream target of adiponectin signaling. Data obtained from RNA sequencing suggest that VAT removal prevents obesity-induced oxidative stress and inflammation in liver and SAT, respectively. Taken together, these findings highlight the metabolic benefits and possible action mechanisms of prophylactic VAT removal on obesity-induced insulin resistance and hepatosteatosis. Our results also provide important insight into understanding the extraordinary capability of adipose tissue to influence whole-body glucose and lipid metabolism as an active endocrine organ
