Transmission of integrin β7 transmembrane domain topology enables gut lymphoid tissue development.
Integrin activation regulates adhesion, extracellular matrix assembly, and cell migration, thereby playing an indispensable role in development and in many pathological processes. A proline mutation in the central integrin β3 transmembrane domain (TMD) creates a flexible kink that uncouples the topology of the inner half of the TMD from the outer half. In this study, using leukocyte integrin α4β7, which enables development of gut-associated lymphoid tissue (GALT), we examined the biological effect of such a proline mutation and report that it impairs agonist-induced talin-mediated activation of integrin α4β7, thereby inhibiting rolling lymphocyte arrest, a key step in transmigration. Furthermore, the α4β7(L721P) mutation blocks lymphocyte homing to and development of the GALT. These studies show that impairing the ability of an integrin β TMD to transmit talin-induced TMD topology inhibits agonist-induced physiological integrin activation and biological function in development
Probing the Dust Properties of Galaxies up to Submillimetre Wavelengths I. The Spectral Energy Distribution of dwarf galaxies using LABOCA
We present 870 micron images of four low metallicity galaxies (NGC1705,
Haro11, Mrk1089 and UM311) observed with the Large APEX BOlometer CAmera
(LABOCA). We model their spectral energy distributions combining the submm
observations of LABOCA, 2MASS, IRAS, Spitzer photometric data and the IRS data
for Haro11. We find that a significant mass of dust is revealed when using
submm constraints compared to that measured with only mid-IR to far-IR
observations extending only to 160 microns. For NGC1705 and Haro11, an excess
in submillimeter wavelengths is detected and we rerun our SED procedure adding
a cold dust component (10K) to better describe the high 870 micron flux derived
from LABOCA observations, which significantly improves the fit. We find that at
least 70% of the dust mass of these two galaxies can reside in a cold dust
component. We also show that the subsequent dust-to-gas mass ratios,
considering HI and CO observations, can be strikingly high for Haro11 in
comparison with what is usually expected for these low-metallicity
environments. Furthermore, we derive the SFR of our galaxies and compare them
to the Schmidt law. Haro11 falls anomalously far from the Schmidt relation.
These results may suggest that a reservoir of hidden gas could be present in
molecular form not traced by the current CO observations. We also derive the
total IR luminosities derived from our models and compare them with relations
that derive this luminosity from Spitzer bands. We find that the Draine & Li

The NAE Pathway : autobahn to the nucleus for cell surface receptors
Various growth factors and full-length cell surface receptors such as EGFR are translocated from the cell surface to the nucleoplasm, baffling cell biologists to the mechanisms and functions of this process. Elevated levels of nuclear EGFR correlate with poor prognosis in various cancers. In recent years, nuclear EGFR has been implicated in regulating gene transcription, cell proliferation and DNA damage repair. Different models have been proposed to explain how the receptors are transported into the nucleus. However, a clear consensus has yet to be reached. Recently, we described the nuclear envelope associated endosomes (NAE) pathway, which delivers EGFR from the cell surface to the nucleus. This pathway involves transport, docking and fusion of NAEs with the outer membrane of the nuclear envelope. EGFR is then presumed to be transported through the nuclear pore complex, extracted from membranes and solubilised. The SUN1/2 nuclear envelope proteins, Importin-beta, nuclear pore complex proteins and the Sec61 translocon have been implicated in the process. While this framework can explain the cell surface to nucleus traffic of EGFR and other cell surface receptors, it raises several questions that we consider in this review, together with implications for health and disease

Unconstrained Nonlinear Optimization of a Distributed SWE Model Using Modis and In Situ Measurements Over the French Alps
International audienceIn this paper we propose the optimization of the snow sub-model of MORDOR using MODIS and in situ measurements for the case study of the Serre-Ponçon reservoir (one of the largest artificial lakes in Western Europe) on the Durance River in the French Alps. The pea branching RMS2 gene encodes the PsAFB4/5 auxin receptor and is involved in an auxin-strigolactone regulation loop
Strigolactones (SLs) are well known for their role in repressing shoot branching. In pea,
increased transcript levels of SL biosynthesis genes are observed in stems of highly
branched SL deficient (ramosus1 (rms1) and rms5) and SL response (rms3 and rms4)
mutants indicative of negative feedback control. In contrast, the highly branched rms2
mutant has reduced transcript levels of SL biosynthesis genes. Grafting studies and hormone
quantification led to a model where RMS2 mediates a shoot-to-root feedback signal
that regulates both SL biosynthesis gene transcript levels and xylem sap levels of cytokinin
exported from roots. Here we cloned RMS2 using synteny with Medicago truncatula and
demonstrated that it encodes a putative auxin receptor of the AFB4/5 clade. Phenotypes
similar to rms2 were found in Arabidopsis afb4/5 mutants, including increased shoot branching,
low expression of SL biosynthesis genes and high auxin levels in stems. Moreover,
afb4/5 and rms2 display a specific resistance to the herbicide picloram. Yeast-two-hybrid
experiments supported the hypothesis that the RMS2 protein functions as an auxin receptor.
SL root feeding using hydroponics repressed auxin levels in stems and down-regulated transcript
levels of auxin biosynthesis genes within one hour. This auxin down-regulation was
also observed in plants treated with the polar auxin transport inhibitor NPA. Together these
data suggest a homeostatic feedback loop in which auxin up-regulates SL synthesis in an
RMS2-dependent manner and SL down-regulates auxin synthesis in an RMS3 and RMS4-
dependent manner
The small heat shock protein B8 (HSPB8) confers resistance to bortezomib by promoting autophagic removal of misfolded proteins in multiple myeloma cells
Velcade is one of the inescapable drug to treat patient suffering from multiple myeloma (MM) and resistance to this drug represents a major drawback for patients. However, the mechanisms underlying velcade resistance remain incompletely understood. We derived several U266 MM cell clones that resist to velcade. U266-resistant cells were resistant to velcade-induced cell death but exhibited a similar sensitivity to various proapoptotic stimuli. Careful analysis of proteosomal subunits and proteasome enzymatic activities showed that neither the composition nor the activity of the proteasome was affected in velcade-resistant cells. Elimination of velcade-induced poly-ubiquitinated proteins and protein aggregates was drastically stimulated in the resistant cells and correlated with increased cell survival. Inhibition of the lysosomal activity in velcade-resistant cells resulted in an increase of cell aggregates and decrease survival, indicating that aggregates are eliminated through lysosomal degradation. In addition, pangenomic profiling of velcade-sensitive and resistant cells showed that the small heat shock protein HSPB8 was overexpressed in resistant cells. Finally, gain and loss of function experiment demonstrated that HSPB8 is a key factor for velcade resistance. In conclusion, HSPB8 plays an important role for the elimination of aggregates in velcade-resistant cells that contributes to their enhanced survival
Evolution of substrate recognition sites (SRSs) in cytochromes P450 from Apiaceae exemplified by the CYP71AJ subfamily
Background
Large proliferations of cytochrome P450 encoding genes resulting from gene duplications can be termed as ‘blooms’, providing genetic material for the genesis and evolution of biosynthetic pathways. Furanocoumarins are allelochemicals produced by many of the species in Apiaceaous plants belonging to the Apioideae subfamily of Apiaceae and have been described as being involved in the defence reaction against phytophageous insects.[br/]
Results
A bloom in the cytochromes P450 CYP71AJ subfamily has been identified, showing at least 2 clades and 6 subclades within the CYP71AJ subfamily. Two of the subclades were functionally assigned to the biosynthesis of furanocoumarins. Six substrate recognition sites (SRS1-6) important for the enzymatic conversion were investigated in the described cytochromes P450 and display significant variability within the CYP71AJ subfamily. Homology models underline a significant modification of the accession to the iron atom, which might explain the difference of the substrate specificity between the cytochromes P450 restricted to furanocoumarins as substrates and the orphan CYP71AJ.[br/]
Conclusion
Two subclades functionally assigned to the biosynthesis of furanocoumarins and four other subclades were identified and shown to be part of two distinct clades within the CYP71AJ subfamily. The subclades show significant variability within their substrate recognition sites between the clades, suggesting different biochemical functions and providing insights into the evolution of cytochrome P450 ‘blooms’ in response to environmental pressures
