The German Socio-Economic Panel as Reference Data Set
This paper discusses how household panels in general?and the German Socio- Economic Panel (SOEP) in particular?can serve as reference data for researchers collecting datasets that do not represent the full universe of the population of interest (e.g., through clinical trials, intervention studies, laboratory and behavioural experiments, and cohort studies). We first discuss potential benefits of using questions similar to those in the SOEP for studies where researchers are interested in using the SOEP as reference data. We present a comprehensive list of SOEP core questions that we recommend researchers to consider when collecting their own data. We focus on seven topics: (1) demographic and parental characteristics; (2) labour market; (3) health; (4) personality, preferences, and subjective orientations; (5) subjective wellbeing; (6) political involvement and participation, and finally, a set of core questions for young children before they enter school. Of course the selection of a minimum set of questions depends on the research question. A Forgotten Issue: Distributional Effects of Day Care Subsidies in Germany
In general, day care subsidies are accepted as a means of creating equal chances for both children and mothers in the labour market. Although there is a broad consensus that the use of children's day care should be publicly supported, there is no consensus on how this should be done. Moreover, there is little knowledge on the distributional effects of day care subsidies. In order to assess whether public expenditures are targeted efficiently, however, it is vital to know which social groups profit most from public expenditures on children's day care and whether taxpayers' money is spent effectively. In Germany, as in other European countries, day care subsidies are mainly provided ‘in-kind’. Municipalities and NPOs provide day care for children, which is - apart from a small fee - free of charge. In this study we estimate the distributional effects of state-funded day care in Germany using microdata on households and data on the expenditure of public-funded day care. Major results are that day care subsidies have only modest redistributional effects. Primarily it is the middle-income range that profits from the public provision of children's day care. Eine solche Unkenntnis über die Verteilungseffekte der öffentlichen Förderung von Kinderbetreuung kann jedoch dazu führen, dass Einkommensgruppen öffentliche Mittel in Anspruch nehmen, die dieser Gelder nicht bedürfen. Wie in einigen anderen europäischen Staaten werden auch in Deutschland bestimmte Anbieter von Kindertageseinrichtungen öffentlich gefördert bzw. die Kommunen betreiben selbst Kindertageseinrichtungen. Damit kommen den Eltern, deren Kinder Kindertageseinrichtungen besuchen, indirekt öffentliche Gelder zu, gleichwohl sie über Elternbeiträge einen geringen Teil der Kosten selbst abdecken. In dieser Studie untersuchen wir die distributiven Effekte der Förderung von Kindertageseinrichtungen in Deutschland, indem wir Mikrodaten auf Haushaltsebene mit Informationen über die öffentlichen Ausgaben im Bereich von Kindertageseinrichtungen kombinieren. Ein wichtiges Ergebnis der Analyse ist, dass über die gegenwärtige „Anbieter-Förderung“ nur sehr geringe Umverteilungswirkungen erzielt werden. Vorrangig kommen die Subventionen im Bereich der Kinderbetreuung mittleren Einkommensgruppen zugute, was dem politischen und gesellschaftlichen Ziel, im Kindertagesstättenbereich insbesondere Kinder aus unteren Einkommensgruppen zu fördern, widerspricht
Novel Chemokine-Based Immunotoxins for Potent and Selective Targeting of Cytomegalovirus Infected Cells
Immunotoxins as antiviral therapeutics are largely unexplored but have promising prospective due to their high selectivity potential and their unparalleled efficiency. One recent example targeted the virus-encoded G protein-coupled receptor US28 as a strategy for specific and efficient treatment of human cytomegalovirus (HCMV) infections. US28 is expressed on virus-infected cells and scavenge chemokines by rapid internalization. The chemokine-based fusion-toxin protein (FTP) consisted of a variant (F49A) of CX3CL1 specifically targeting US28 linked to the catalytic domain of Pseudomonas exotoxin A (PE). Here, we systematically seek to improve F49A-FTP by modifications in its three structural domains; we generated variants with (1) altered chemokine sequence (K14A, F49L, and F49E), (2) shortened and elongated linker region, and (3) modified toxin domain. Only F49L-FTP displayed higher selectivity in its binding to US28 versus CX3CR1, the endogenous receptor for CX3CL1, but this was not matched by a more selective killing of US28-expressing cells. A longer linker and different toxin variants decreased US28 affinity and selective killing. Thereby, F49A-FTP represents the best candidate for HCMV treatment. Many viruses encode internalizing receptors suggesting that not only HCMV but also, for instance, Epstein-Barr virus and Kaposi’s sarcoma-associated herpesvirus may be targeted by FTPs
Gene transfer into hepatocytes using asialoglycoprotein receptor mediated endocytosis of DNA complexed with an artificial tetra-antennary galactose ligand
We have constructed an artificial ligand for the hepatocyte-specific asialoglycoprotein receptor for the purpose of generating a synthetic delivery system for DNA. This ligand has a tetra-antennary structure, containing four terminal galactose residues on a branched carrier peptide. The carbohydrate residues of this glycopeptide were introduced by reductive coupling of lactose to the alpha- and epsilon-amino groups of the two N-terminal lysines on the carrier peptide. The C-terminus of the peptide, containing a cysteine separated from the branched N-terminus by a 10 amino acid spacer sequence, was used for conjugation to 3-(2-pyridyldithio)propionate-modified polylysine via disulfide bond formation. Complexes containing plasmid DNA bound to these galactose-polylysine conjugates have been used for asialoglycoprotein receptor-mediated transfer of a luciferase gene into human (HepG2) and murine (BNL CL.2) hepatocyte cell lines. Gene transfer was strongly promoted when amphipathic peptides with pH-controlled membrane-disruption activity, derived from the N-terminal sequence of influenza virus hemagglutinin HA-2, were also present in these DNA complexes. Thus, we have essentially borrowed the small functional domains of two large proteins, asialoglycoprotein and hemagglutinin, and assembled them into a supramolecular complex to generate an efficient gene-transfer system
Increasing 3D Supramolecular Order by Decreasing Molecular Order. A Comparative Study of Helical Assemblies of Dendronized Nonchlorinated and Tetrachlorinated Perylene Bisimides
A nonplanar, twisted, and flexible tetrachlorinated perylene bisimide (Cl4PBI) was functionalized with two AB3 minidendrons containing hydrogenated or semifluorinated dodecyl groups. The hydrogenated dendron was attached to the imide groups of Cl4PBI via m = 0, 1, and 2 methylenic units, whereas the dendron containing semifluorinated groups was attached via m = 3 or a di(ethylene oxide) linker (m = 2EO). The supramolecular structures of these compounds, determined by a combination of differential scanning calorimetry, X-ray diffraction, and solid-state NMR, were compared with those of nonchlorinated planar and rigid PBI reported previously, which demonstrated the thermodynamically controlled formation of 2D periodic arrays at high temperatures and 3D arrays at low temperatures. The molecularly less ordered Cl4PBI containing hydrogenated dendrons self-organize into exclusively 3D crystalline periodic arrays under thermodynamic control for m = 0 and 2, while the more highly molecularly ordered PBI produced less stable and ordered 3D crystals and also 2D assemblies. This induction of a higher degree of 3D order in supramolecular assemblies of the less well-ordered molecular building blocks was unanticipated. The semifluorinated dendronized Cl4PBI with m = 3 formed a 2D columnar hexagonal array under kinetic control, whereas the compound with m = 2EO formed an unusual 2D honeycomb-like hexagonal phase under thermodynamic control. These Cl4PBI compounds provide a new route to stable crystalline assemblies via thermodynamic control at lower temperatures than previously obtained with PBI, thus generating 3D order in an accessible range of temperature of interest for structural analysis and for technological applications

We present a calculation of a fourth-order, time-dependent density
correlation function that measures higher-order spatiotemporall correlations of
the density of a liquid. From molecular dynamics simulations of a glass-forming
Lennard-Jones liquid, we find that the characteristic length scale of this
function has a maximum as a function of time which increases steadily beyond
the characteristic length of the static pair correlation function in the
temperature range approaching the mode coupling temperature from above

Molecular mode-coupling theory applied to a liquid of diatomic molecules
We study the molecular mode coupling theory for a liquid of diatomic
molecules. The equations for the critical tensorial nonergodicity parameters
and the critical amplitudes of the - relaxation
are solved up to a cut off = 2 without any
further approximations.
Here are indices of spherical harmonics. Contrary to previous studies,
where additional approximations were applied, we find in agreement with
simulations, that all molecular degrees of freedom vitrify at a single
temperature . The theoretical results for the non ergodicity parameters
and the critical amplitudes are compared with those from simulations. The
qualitative agreement is good for all molecular degrees of freedom. To study
the influence of the cut off on the non ergodicity parameter, we also calculate
the non ergodicity parameters for an upper cut off . In addition we
also propose a new method for the calculation of the critical nonergodicity

Evaluation of a candidate breast cancer associated SNP in ERCC4 as a risk modifier in BRCA1 and BRCA2 mutation carriers. Results from the Consortium of Investigators of Modifiers of BRCA1/BRCA2 (CIMBA)
Background: In this study we aimed to evaluate the role of a SNP in intron 1 of the ERCC4 gene (rs744154), previously reported to be associated with a reduced risk of breast cancer in the general population, as a breast cancer risk modifier in BRCA1 and BRCA2 mutation carriers. Methods: We have genotyped rs744154 in 9408 BRCA1 and 5632 BRCA2 mutation carriers from the Consortium of Investigators of Modifiers of BRCA1/2 (CIMBA) and assessed its association with breast cancer risk using a retrospective weighted cohort approach. Results: We found no evidence of association with breast cancer risk for BRCA1 (per-allele HR: 0.98, 95% CI: 0.93–1.04, P=0.5) or BRCA2 (per-allele HR: 0.97, 95% CI: 0.89–1.06, P=0.5) mutation carriers. Conclusion: This SNP is not a significant modifier of breast cancer risk for mutation carriers, though weak associations cannot be ruled out. Retinal glycoprotein enrichment by concanavalin a enabled identification of novel membrane autoantigen synaptotagmin-1 in equine recurrent uveitis.
Complete knowledge of autoantigen spectra is crucial for understanding pathomechanisms of autoimmune diseases like equine recurrent uveitis (ERU), a spontaneous model for human autoimmune uveitis. While several ERU autoantigens were identified previously, no membrane protein was found so far. As there is a great overlap between glycoproteins and membrane proteins, the aim of this study was to test whether pre-enrichment of retinal glycoproteins by ConA affinity is an effective tool to detect autoantigen candidates among membrane proteins. In 1D Western blots, the glycoprotein preparation allowed detection of IgG reactions to low abundant proteins in sera of ERU patients. Synaptotagmin-1, a Ca2+-sensing protein in synaptic vesicles, was identified as autoantigen candidate from the pre-enriched glycoprotein fraction by mass spectrometry and was validated as a highly prevalent autoantigen by enzyme-linked immunosorbent assay. Analysis of Syt1 expression in retinas of ERU cases showed a downregulation in the majority of ERU affected retinas to 24%. Results pointed to a dysregulation of retinal neurotransmitter release in ERU. Identification of synaptotagmin-1, the first cell membrane associated autoantigen in this spontaneous autoimmune disease, demonstrated that examination of tissue fractions can lead to the discovery of previously undetected novel autoantigens. Further experiments will address its role in ERU pathology
