    A Method for the Direct Identification of Differentiating Muscle Cells by a Fluorescent Mitochondrial Dye

    Identification of differentiating muscle cells generally requires fixation, antibodies directed against muscle specific proteins, and lengthy staining processes or, alternatively, transfection of muscle specific reporter genes driving GFP expression. In this study, we examined the possibility of using the robust mitochondrial network seen in maturing muscle cells as a marker of cellular differentiation. The mitochondrial fluorescent tracking dye, MitoTracker, which is a cell-permeable, low toxicity, fluorescent dye, allowed us to distinguish and track living differentiating muscle cells visually by epi-fluorescence microscopy. MitoTracker staining provides a robust and simple detection strategy for living differentiating cells in culture without the need for fixation or biochemical processing

    Seroprevalence of Helicobacter pylori infection in chagasic and nonchagasic patients from the same geographical region of Brazil

    INTRODUCTION: In this study, we evaluated the seroprevalence of Helicobacter pylori infection among chagasic and non-chagasic subjects as well as among the subgroups of chagasic patients with the indeterminate, cardiac, digestive, and cardiodigestive clinical forms. METHODS: The evaluated subjects were from the Triângulo Mineiro region, Minas Gerais, Brazil. Chagasic patients showed positive reactions to the conventional serological tests used and were classified according to the clinical form of their disease. Immunoglobulin G antibodies specific to H. pylori were measured using a commercial enzyme-linked immunosorbent assay kit. RESULTS: The overall H. pylori prevalence was 77.1% (239/310) in chagasic and 69.1% (168/243) in non-chagasic patients. This difference was statistically significant even after adjustment for age and sex (odds ratio = 1.57; 95% confidence interval, 1.02-2.42; p = 0.04) in multivariate analysis. The prevalence of infection increased with age in the non-chagasic group (p = 0.007, χ2 for trend), but not in the chagasic group (p = 0.15, χ2 for trend). H. pylori infection was not associated with digestive or other clinical forms of Chagas disease (p = 0.27). CONCLUSIONS: Our findings demonstrate that chagasic patients have a higher prevalence of H. pylori compared to non-chagasic subjects; a similar prevalence was found among the diverse clinical forms of the disease. The factors contributing to the frequent co-infection with H. pylori and Trypanosoma cruzi as well as its effects on the clinical outcome deserve further study

    Quantifying Microstegium vimineum Seed Movement by Non-Riparian Water Dispersal Using an Ultraviolet-Marking Based Recapture Method

    Microstegium vimineum is a shade tolerant annual C(4) invasive grass in the Eastern US, which has been shown to negatively impact species diversity and succession in hardwood forests. To date, empirical studies have shown that population expansion is limited to <1 m yr(−1), which is largely driven by gravity dispersal. However, this likely does not fully account for all mechanisms of population-scale dispersal as we observe greater rates of population expansion. Though water, both riparian and non-riparian water (i.e., ephemeral overland flow), have been speculated mechanisms for M. vimineum dispersal, few studies have empirically tested this hypothesis. We designed an experiment along the slopes of a Southwest Virginia hardwood forest to test the role of non-riparian water on local seed dispersal. We developed a seed marking technique by coating each seed with an ultraviolet (UV) powder that did not affect buoyancy to aid in situ seed recapture. Additionally, a new image analysis protocol was developed to automate seed identification from UV photos. Total seed mobility (summation of individual seed movement within each transect) was positively correlated with precipitation. Over a period of one month with 52.32 mm of precipitation, the maximum dispersal distance of any single recaptured seed was 2.4 m, and the average distance of dispersed seed was 0.21±0.04 m. This is the first quantitative evidence of non-riparian water dispersal in a forest understory, which accounts for an additional pathway of population expansion

    Exploring the Role of Diversity Management During Early Internationalizing Firms’ Internationalization Process

    Despite their rapid internationalization, early internationalizing firms (EIFs) struggle to sustain their growth over time. Among the factors influencing their internationalization process, the diversity of human resources, and particularly its management, has yet to be studied. Building on different perspectives of diversity management, through the lens of the resource-based view, this study explores the role of diversity management during EIFs’ internationalization process. The results of a multiple-case, qualitative study of French EIFs show that prevailing perspectives on diversity management within EIFs change with the nature of diversity as a resource (strategic, ordinary, or negative), with different influences on the firms’ progress along the phases of the internationalization process. The findings highlight the importance of adopting a learning perspective on managing diversity, including specific management practices, during the transition between the entry and post-entry phases of EIFs’ internationalization process. This study thus suggests several propositions and theoretical contributions, along with managerial recommendations
