Nefrotoxicidade da doxorrubicina em gatas (Felis catus) com neoplasias mamárias malignas
O objetivo do estudo foi investigar a nefrotoxicidade da doxorrubicina em gatas com neoplasias
malignas da glândula mamária. Todas as gatas selecionadas não apresentaram comorbidades como
nefropatias e/ou cardiomiopatias, comprovadas por exames físicos e laboratoriais, foram submetidas à
mastectomia radical associada à exérese de linfonodos regionais e tratadas com protocolo baseado em
doxorrubicina. Os marcadores renais ureia, creatinina, dimetilarginina simétrica foram avaliados
durante todo o tratamento e dois meses após o término. Ultrassonografia abdominal, urinálise e análise
bioquímica de proteínas, creatinina urinária e GGT urinária foram realizadas no início e dois meses
após o término do tratamento, para avaliar possíveis alterações renais. Seis gatas não apresentaram
alterações renais em nenhum dos exames realizados durante o estudo. Duas gatas apresentaram
azotemia durante este estudo, e uma delas interrompeu o tratamento precocemente, devido à intensa
azotemia. A individualidade de cada paciente deve ser sempre considerada, pois são muitas as variáveis.
O monitoramento clínico, com exames complementares, tais como bioquímicos séricos e de imagem dos
pacientes durante todo o tratamento, é essencial, especialmente a mensuração de analitos que detectam
alterações renais precocemente. Diante disso, ressalta-se que a doxorrubicina é um medicamento seguro
para utilização em gatas não nefropatas.The aim of the study was to investigate the nephrotoxicity of doxorubicin in cats with malignant
neoplasms of the mammary gland. All selected cats did not present comorbidities such as nephropathies
and/or cardiomyopathies, confirmed by physical and laboratory tests, underwent radical mastectomy
associated with regional lymph node excision and were treated with a protocol based on doxorubicin.
Renal markers of urea, creatinine, symmetrical dimethylarginine were evaluated during all the treatment,
and two months after the end. Abdominal ultrasound, urinalysis and biochemical analysis of protein,
urinary creatinine and urinary GGT were performed in the beginning and two months after the end of the
treatment, to evaluate possible renal alterations. Six cats did not present renal alterations in any exams
performed during the study. Two cats presented azotemia during this study, and one of them stopped the
treatment early, because of the intense azotemia. Clinical imaging and laboratory monitoring of patients
throughout the treatment is essential, including the measurement of analytes that detect kidney changes
early. Thus, it is emphasized that doxorubicin is a safe drug for use in non-nephropathic cats
Columnar cell lesions of the canine mammary gland: pathological features and immunophenotypic analysis
<p>Abstract</p> <p>Background</p> <p>It has been suggested that columnar cell lesions indicate an alteration of the human mammary gland involved in the development of breast cancer. They have not previously been described in canine mammary gland. The aim of this paper is describe the morphologic spectrum of columnar cell lesions in canine mammary gland specimens and their association with other breast lesions.</p> <p>Methods</p> <p>A total of 126 lesions were subjected to a comprehensive morphological review based upon the human breast classification system for columnar cell lesions. The presence of preinvasive (epithelial hyperplasia and in situ carcinoma) and invasive lesions was determined and immunophenotypic analysis (estrogen receptor (ER), progesterone receptor (PgR), high molecular weight cytokeratin (34βE-12), E-cadherin, Ki-67, HER-2 and P53) was perfomed.</p> <p>Results</p> <p>Columnar cell lesions were identified in 67 (53.1%) of the 126 canine mammary glands with intraepithelial alterations. They were observed in the terminal duct lobular units and characterized at dilated acini may be lined by several layers of columnar epithelial cells with elongated nuclei. Of the columnar cell lesions identified, 41 (61.2%) were without and 26 (38.8%) with atypia. Association with ductal hyperplasia was observed in 45/67 (67.1%). Sixty (89.5%) of the columnar cell lesions coexisted with neoplastic lesions (20 in situ carcinomas, 19 invasive carcinomas and 21 benign tumors). The columnar cells were ER, PgR and E-cadherin positive but negative for cytokeratin 34βE-12, HER-2 and P53. The proliferation rate as measured by Ki-67 appeared higher in the lesions analyzed than in normal TDLUs.</p> <p>Conclusions</p> <p>Columnar cell lesions in canine mammary gland are pathologically and immunophenotypically similar to those in human breast. This may suggest that dogs are a suitable model for the comparative study of noninvasive breast lesions.</p
Immunophenotypic features of tumor infiltrating lymphocytes from mammary carcinomas in female dogs associated with prognostic factors and survival rates
<p>Abstract</p> <p>Background</p> <p>The immune system plays an important role in the multifactorial biologic system during the development of neoplasias. However, the involvement of the inflammatory response in the promotion/control of malignant cells is still controversial, and the cell subsets and the mechanisms involved are poorly investigated. The goal of this study was to characterize the clinical-pathological status and the immunophenotyping profile of tumor infiltrating lymphocytes and their association with the animal survival rates in canine mammary carcinomas.</p> <p>Methods</p> <p>Fifty-one animals with mammary carcinomas, classified as carcinomas in mixed tumors-MC-BMT = 31 and carcinomas-MC = 20 were submitted to systematic clinical-pathological analysis (tumor size; presence of lymph node and pulmonary metastasis; clinical stage; histological grade; inflammatory distribution and intensity as well as the lymphocytic infiltrate intensity) and survival rates. Twenty-four animals (MC-BMT = 16 and MC = 8) were elected to the immunophenotypic study performed by flow cytometry.</p> <p>Results</p> <p>Data analysis demonstrated that clinical stage II-IV and histological grade was I more frequent in MC-BMT as compared to MC. Univariate analysis demonstrated that the intensity of inflammation (moderate/intense) and the proportion of CD4<sup>+ </sup>(≥ 66.7%) or CD8<sup>+ </sup>T-cells (<33.3%) were not associated with worse survival rate. Multivariate analysis demonstrated that only lymphocytic infiltrate intensity ≥ 600 (<it>P </it>= 0.02) remained as independent prognostic factor. Despite the clinical manifestation, the lymphocytes represented the predominant cell type in the tumor infiltrate. The percentage of T-cells was higher in animals with MC-BMT without metastasis, while the percentage of B-lymphocytes was greater in animals with metastasized MC-BMT (<it>P </it>< 0.05). The relative percentage of CD4<sup>+ </sup>T-cells was significantly greater in metastasized tumors (both MC-BMT and MC), (<it>P </it>< 0.05) while the proportion of CD8<sup>+ </sup>T-cells was higher in MC-BMT without metastasis. Consequently, the CD4<sup>+</sup>/CD8<sup>+ </sup>ratio was significantly increased in both groups with metastasis. Regardless of the tumor type, the animals with high proportions of CD4<sup>+ </sup>and low CD8<sup>+ </sup>T-cells had decreased survival rates.</p> <p>Conclusion</p> <p>The intensity of lymphocytic infiltrate and probably the relative abundance of the CD4<sup>+ </sup>and CD8<sup>+ </sup>T-lymphocytes may represent important survival prognostic biomarkers for canine mammary carcinomas.</p
Differential expression of follistatin and FLRG in human breast proliferative disorders
<p>Abstract</p> <p>Background</p> <p>Activins are growth factors acting on cell growth and differentiation. Activins are expressed in high grade breast tumors and they display an antiproliferative effect inducing G0/G1 cell cycle arrest in breast cancer cell lines. Follistatin and follistatin- related gene (FLRG) bind and neutralize activins. In order to establish if these activin binding proteins are involved in breast tumor progression, the present study evaluated follistatin and FLRG pattern of mRNA and protein expression in normal human breast tissue and in different breast proliferative diseases.</p> <p>Methods</p> <p>Paraffin embedded specimens of normal breast (NB - n = 8); florid hyperplasia without atypia (FH - n = 17); fibroadenoma (FIB - n = 17); ductal carcinoma <it>in situ </it>(DCIS - n = 10) and infiltrating ductal carcinoma (IDC - n = 15) were processed for follistatin and FLRG immunohistochemistry and <it>in situ </it>hybridization. The area and intensity of chromogen epithelial and stromal staining were analyzed semi-quantitatively.</p> <p>Results</p> <p>Follistatin and FLRG were expressed both in normal tissue and in all the breast diseases investigated. Follistatin staining was detected in the epithelial cytoplasm and nucleus in normal, benign and malignant breast tissue, with a stronger staining intensity in the peri-alveolar stromal cells of FIB at both mRNA and protein levels. Conversely, FLRG area and intensity of mRNA and protein staining were higher both in the cytoplasm and in the nucleus of IDC epithelial cells when compared to NB, while no significant changes in the stromal intensity were observed in all the proliferative diseases analyzed.</p> <p>Conclusion</p> <p>The present findings suggest a role for follistatin in breast benign disease, particularly in FIB, where its expression was increased in stromal cells. The up regulation of FLRG in IDC suggests a role for this protein in the progression of breast malignancy. As activin displays an anti-proliferative effect in human mammary cells, the present findings indicate that an increased FST and FLRG expression in breast proliferative diseases might counteract the anti-proliferative effects of activin in human breast cancer.</p
A comparative study between mixed-type tumours from human salivary and canine mammary glands
<p>Abstract</p> <p>Background</p> <p>In comparative pathology, canine mammary tumours have special interest because of their similarities with human breast cancer. Mixed tumours are uncommon lesions in the human breast, but they are found most frequently in the mammary gland of the female dogs and in the human salivary glands. The aim of the study was to compare clinical, morphological and immunohistochemical features of human salivary and canine mammary gland mixed tumours, in order to evaluate the latter as an experimental model for salivary gland tumours.</p> <p>Methods</p> <p>Ten examples of each mixed tumour type (human pleomorphic adenoma and carcinomas ex-pleomorphic adenomas and canine mixed tumour and metaplastic carcinoma) were evaluated. First, clinical and morphologic aspects of benign and malignant variants were compared between the species. Then, streptavidin-biotin-peroxidase immunohistochemistry was performed to detect the expression of cytokeratins, vimentin, p63 protein, estrogen receptor, β-catenin, and E-cadherin.</p> <p>Results</p> <p>After standardization, similar age and site distributions were observed in human and canine tumours. Histological similarities were identified in the comparison of the benign lesions as well. Metaplastic carcinomas also resembled general aspects of carcinomas ex-pleomorphic adenomas in morphological evaluation. Additionally, immunohistochemical staining further presented similar antigenic expression between lesions.</p> <p>Conclusion</p> <p>There are many similar features between human salivary and canine mammary gland mixed tumours. This observation is of great relevance for those interested in the study and management of salivary gland tumours, since canine lesions may constitute useful comparative models for their investigations.</p
Histomorphological and immunohistochemical characterization of 172 cutaneous round cell tumours in dogs
Platelet-Activating Factor Receptor Plays a Role in Lung Injury and Death Caused by Influenza A in Mice
Influenza A virus causes annual epidemics which affect millions of people
worldwide. A recent Influenza pandemic brought new awareness over the health
impact of the disease. It is thought that a severe inflammatory response against
the virus contributes to disease severity and death. Therefore, modulating the
effects of inflammatory mediators may represent a new therapy against Influenza
infection. Platelet activating factor (PAF) receptor (PAFR) deficient mice were
used to evaluate the role of the gene in a model of experimental infection with
Influenza A/WSN/33 H1N1 or a reassortant Influenza A H3N1 subtype. The following
parameters were evaluated: lethality, cell recruitment to the airways, lung
pathology, viral titers and cytokine levels in lungs. The PAFR antagonist
PCA4248 was also used after the onset of flu symptoms. Absence or antagonism of
PAFR caused significant protection against flu-associated lethality and lung
injury. Protection was correlated with decreased neutrophil recruitment, lung
edema, vascular permeability and injury. There was no increase of viral load and
greater recruitment of NK1.1+ cells. Antibody responses were
similar in WT and PAFR-deficient mice and animals were protected from
re-infection. Influenza infection induces the enzyme that synthesizes PAF,
lyso-PAF acetyltransferase, an effect linked to activation of TLR7/8. Therefore,
it is suggested that PAFR is a disease-associated gene and plays an important
role in driving neutrophil influx and lung damage after infection of mice with
two subtypes of Influenza A. Further studies should investigate whether
targeting PAFR may be useful to reduce lung pathology associated with Influenza
A virus infection in humans
