    Second-harmonic phonon spectroscopy of α\alpha-quartz

    We demonstrate midinfrared second-harmonic generation as a highly sensitive phonon spectroscopy technique that we exemplify using α\alpha-quartz (SiO2_2) as a model system. A midinfrared free-electron laser provides direct access to optical phonon resonances ranging from 350 cm1350\ \mathrm{cm}^{-1} to $1400\ \mathrm{cm}^{-1}.Whiletheextremelywidetunabilityandhighpeakfieldsofanfreeelectronlaserpromotenonlinearspectroscopicstudiescomplementedbysimultaneouslinearreflectivitymeasurementsazimuthalscansrevealcrystallographicsymmetryinformationofthesample.Additionally,temperaturedependentmeasurementsshowhowdampingratesincrease,phononmodesshiftspectrallyandincertaincasesdisappearcompletelywhenapproaching. While the extremely wide tunability and high peak fields of an free-electron laser promote nonlinear spectroscopic studies---complemented by simultaneous linear reflectivity measurements---azimuthal scans reveal crystallographic symmetry information of the sample. Additionally, temperature-dependent measurements show how damping rates increase, phonon modes shift spectrally and in certain cases disappear completely when approaching T_c=846\ \mathrm{K}wherequartzundergoesastructuralphasetransitionfromtrigonal where quartz undergoes a structural phase transition from trigonal \alphaquartztohexagonal-quartz to hexagonal \beta$-quartz, demonstrating the technique's potential for studies of phase transitions

    Autophagy gene expression profiling identifies a defective microtubule-associated protein light chain 3A mutant in cancer.

    The cellular stress response autophagy has been implicated in various diseases including neuro-degeneration and cancer. The role of autophagy in cancer is not clearly understood and both tumour promoting and tumour suppressive effects of autophagy have been reported, which complicates the design of therapeutic strategies based on targeting the autophagy pathway. Here, we have systematically analyzed gene expression data for 47 autophagy genes for deletions, amplifications and mutations in various cancers. We found that several cancer types have frequent autophagy gene amplifications, whereas deletions are more frequent in prostate adenocarcinomas. Other cancer types such as glioblastoma and thyroid carcinoma show very few alterations in any of the 47 autophagy genes. Overall, individual autophagy core genes are altered at low frequency in cancer, suggesting that cancer cells require functional autophagy. Some autophagy genes show frequent single base mutations, such as members of the ULK family of protein kinases. Furthermore, we found hotspot mutations in the arginine-rich stretch in MAP1LC3A resulting in reduced cleavage of MAP1LC3A by ATG4B both in vitro and in vivo, suggesting a functional implication of this gene mutation in cancer development

    Resonant enhancement of second harmonic generation in the mid-infrared using localized surface phonon polaritons in sub-diffractional nanostructures

    We report on strong enhancement of mid-infrared second harmonic generation (SHG) from SiC nanopillars due to the resonant excitation of localized surface phonon-polaritons within the Reststrahlen band. The magnitude of the SHG peak at the monopole mode experiences a strong dependence on the resonant frequency beyond that described by the field localization degree and the dispersion of linear and nonlinear-optical SiC properties. Comparing the results for the identical nanostructures made of 4H and 6H SiC polytypes, we demonstrate the interplay of localized surface phonon polaritons with zone-folded weak phonon modes of the anisotropic crystal. Tuning the monopole mode in and out of the region where the zone-folded phonon is excited in 6H-SiC, we observe a prominent increase of the already monopole-enhanced SHG output when the two modes are coupled. Envisioning this interplay as one of the showcase features of mid-infrared nonlinear nanophononics, we discuss its prospects for the effective engineering of nonlinear-optical materials with desired properties in the infrared spectral range.Comment: 16 pages, 3 figure

    Phosphatidylinositol 4-kinase IIβ negatively regulates invadopodia formation and suppresses an invasive cellular phenotype

    The type II PI 4-kinases enzymes synthesise the lipid phosphatidylinositol 4-phosphate (PI(4)P) which has been detected at the Golgi complex and endosomal compartments, and which recruits clathrin adaptors. Despite common mechanistic similarities between the isoforms, the extent of their redundancy is unclear.We found that depletion of PI4KIIα and PI4KIIβ using siRNA led to actin remodelling. Depletion of PI4KIIβ also induced the formation of invadopodia containing membrane type I matrix metalloproteinase (MT1-MMP).Depletion of PI4KII isoforms also differentially affected TGN pools of PI(4)P and post-TGN traffic. PI4KIIβ depletion caused increased MT1-MMP trafficking to invasive structures at the plasma membrane and was accompanied by reduced colocalisation of MT1-MMP with membranes containing the endosomal markers Rab5 and Rab7, but increased localisation with the exocytic Rab8. Depletion of PI4KIIβ was sufficient to confer an aggressive invasive phenotype on minimally invasive HeLa and MCF-7 cell lines. Mining oncogenomic databases revealed that loss of the PI4K2B allele and underexpression of PI4KIIβ mRNA is associated with human cancers. This finding supports the cell data and suggests that PI4KIIβ may be a clinically significant suppressor of invasion. We propose that PI4KIIβ synthesises a pool of PI(4)P that maintains MT1-MMP traffic in the degradative pathway and suppresses the formation of invadopodia

    Elucidating Surface Structure with Action Spectroscopy

    Surface Action Spectroscopy, a vibrational spectroscopy method developed in recent years at the Fritz Haber Institute is employed for structure determination of clean and H2O-dosed (111) magnetite surfaces. Surface structural information is revealed by using the microscopic surface vibrations as a fingerprint of the surface structure. Such vibrations involve just the topmost atomic layers, and therefore the structural information is truly surface related. Our results strongly support the view that regular Fe3O4(111)/Pt(111) is terminated by the so-called Fetet1 termination, that the biphase termination of Fe3O4(111)/Pt(111) consists of FeO and Fe3O4(111) terminated areas, and we show that the method can differentiate between different water structures in H2O-derived adsorbate layers on Fe3O4(111)/Pt(111). With this, we conclude that the method is a capable new member in the set of techniques providing crucial information to elucidate surface structures. The method does not rely on translational symmetry and can therefore also be applied to systems which are not well ordered. Even an application to rough surfaces is possible

    German early career scholars in Gender Studies: Do networks matter?

    Purpose of the study: The field of Gender Studies in Germany demonstrates a rather heterogeneous culture due to its unclear status in relation to other disciplines, while its scope varies from diversity management to critical feminism. Career origins, paths and options for new generations of researchers in this field have to date been only minimally analysed. The contribution of this essay aims at reducing the existing research gap by focusing on the career start and qualification stage in Gender Studies and highlighting the significance of social networks in this process. Methodology: Building upon a qualitative analysis, this study is based on an explorative investigation into German early career researchers. It utilises primary data collected during semi-structured problem-centred telephone interviews with 30 PhD students and Postdocs. The findings evidence three forms of recruitment of PhD students into Gender Studies and question the same sex co-optation principle reported within other fields of scientific inquiry. At the same time, results show that network composition and modes of support are based on the supervisor’s and the early career scholar’s mutual interest in contributing to theory, rather than maximising political and administrative power. The main contribution, thus, addresses social networks and institutional nepotism in general and as a recruiting strategy in particular, as well as the role of graduate schools as a ‘second best’ option for junior researchers in Gender Studies
