Kinases play a critical
role in cellular signaling and are dysregulated
in a number of diseases, such as cancer, diabetes, and neurodegeneration.
Therapeutics targeting kinases currently account for roughly 50% of
cancer drug discovery efforts. The ability to explore human kinase
biochemistry and biophysics in the laboratory is essential to designing
selective inhibitors and studying drug resistance. Bacterial expression
systems are superior to insect or mammalian cells in terms of simplicity
and cost effectiveness but have historically struggled with human
kinase expression. Following the discovery that phosphatase coexpression
produced high yields of Src and Abl kinase domains in bacteria, we
have generated a library of 52 His-tagged human kinase domain constructs
that express above 2 μg/mL of culture in an automated bacterial
expression system utilizing phosphatase coexpression (YopH for Tyr
kinases and lambda for Ser/Thr kinases). Here, we report a structural
bioinformatics approach to identifying kinase domain constructs previously
expressed in bacteria and likely to express well in our protocol,
experiments demonstrating our simple construct selection strategy
selects constructs with good expression yields in a test of 84 potential
kinase domain boundaries for Abl, and yields from a high-throughput
expression screen of 96 human kinase constructs. Using a fluorescence-based
thermostability assay and a fluorescent ATP-competitive inhibitor,
we show that the highest-expressing kinases are folded and have well-formed
ATP binding sites. We also demonstrate that these constructs can enable
characterization of clinical mutations by expressing a panel of 48
Src and 46 Abl mutations. The wild-type kinase construct library is
available publicly via Addgene

The location was mainly in the head and or neck (94.3%). SK was most frequently treated with a CO2 laser (56.2%), followed by electrocautery (36.2%).
Conclusion: Most SK patients in this study were female, middle-aged (40-59 years), with lesions on the head and or neck, and were treated with CO2 laser
ROLE OF STUDENTS IN THE COMMUNITY SERVICE PROGRAM IN THE NATIONAL CHILD IMMUNIZATION MONTH TO REALIZE THE HEALTHY HUMAN RESOURCES
The community service program refers to the lecturer and student activities that support real facilities to implement various academic activities that can have an impact on the community. The COVID-19 pandemic caused many Indonesian children did not receive complete routine immunization. Based on that condition the Ministry of Health launched National Child Immunization Month 2022/BIAN to boost the routine immunization coverage of the children. The BIAN socialization, the first step the team members met the Village Head and his staff to invoke for permission and support from local leaders to socialize BIAN to the community. Then, the doctors and midwives in Primary Health Care, community unit officers, and village cadres were met to be asked for guidance. The BIAN socialization was decided to be performed by two ways: By including BIAN socialization events at routine Primary Health Care events and implementation by several meetings in each Community Unit. Vaccination data in Mekarwangi Village were 60% of children that have been vaccinated and 40% have not been vaccinated. This situation is not only due to the effects of the COVID-19 pandemic, but also because the understanding and awareness of the parents about vaccination is still low. The BIAN socialization explained the benefits of vaccination and invited the parents to participate in preventing and maintaining the health of their families, especially their children by participating the immunization of their children in BIAN activities. The events involved 286 mothers. The community service program students have contributed in realizing healthy human resources

lncRNAs in Hallmarks of Cancer and Clinical Applications
Long noncoding RNAs (lncRNAs) are transcripts longer than 200 nucleotides in length that, in general, do not appear to have protein-coding potential. lncRNAs act in gene regulation involved with several biological processes. Furthermore, lncRNAs have been associated with a significant number of cancers, suggesting a potential role in tumorigenesis and progression. For example, HOTAIR regulates proliferation processes and other lncRNAs like highly upregulated in liver cancer (HULC), H19, PTENP1, HEIH, and antisense noncoding RNA in the INK4 locus (ANRIL). Other lncRNAs as AFAP1-AS1 and lincRNA-p21 can interact with BCL-2 and TP53, acting in apoptosis. Moreover, NORAD plays a vital role in genomic stability. Additionally, due to deregulated expression and high tissue specificity level, lncRNAs exhibit great potential as prognostic markers. In this chapter, we review the most highlighted lncRNAs acting in hallmarks of cancer and clinical application
Curcuma longa L. Prevents Hepatotoxicity Induced Isoniazid and Rifampicin: An Experiment in Wistar Rats Model
Background: Hepatotoxicity induced by the combination of isoniazid (INH) and rifampicin (RIF) in the treatment of tuberculosis (TB) remains a major concern. Oxidative stress has a role in mechanism of hepatotoxicity. Curcuma longa L. has been widely used as a traditional medicine and has shown antioxidant activity. This study aimed to provide evidence of Curcuma longa L as protection against oxidative stress induced by isoniazid and rifampicin therapy. Methods: This was an experimental study on male Wistar rats weighing 150-200 grams, aged 8-10 weeks which were divided into a negative control group (K0), a positive control group with INH + RIF (K1), a treatment group with a dose of 2.2 gr/kg/day Curcuma longa L powder (K2), and treatment group with INH+RIF and additional 2.2 gr/kg/day turmeric rhizome powder (K2+). SGOT and SGPT were measured from blood plasma on the 28th day; then hepatic tissue was obtained to measure MDA levels and observed histologically. Statistical analysis was done using ANOVA and continued with Duncan procedure using SPSS ver. 27.Results: SGOT, SGPT, the highest average MDA level in the liver, and the highest mean necrotic cell count in the positive control group showed a significant difference (p<0.05). The treatment group had a smaller average number of necrotic cells than the positive control group with a significant difference (p <0.05).Conclusion: Curcuma longa L powder has been shown to prevent elevation in SGOT, SGPT, MDA of liver tissue and hepatocyte necrosis, indicating its potential in protecting the liver from oxidative stress
Predicting the Proteins of Angomonas deanei, Strigomonas culicis and Their Respective Endosymbionts Reveals New Aspects of the Trypanosomatidae Family
Endosymbiont-bearing trypanosomatids have been considered excellent models for the study of cell evolution because the host protozoan co-evolves with an intracellular bacterium in a mutualistic relationship. Such protozoa inhabit a single invertebrate host during their entire life cycle and exhibit special characteristics that group them in a particular phylogenetic cluster of the Trypanosomatidae family, thus classified as monoxenics. in an effort to better understand such symbiotic association, we used DNA pyrosequencing and a reference-guided assembly to generate reads that predicted 16,960 and 12,162 open reading frames (ORFs) in two symbiont-bearing trypanosomatids, Angomonas deanei (previously named as Crithidia deanei) and Strigomonas culicis (first known as Blastocrithidia culicis), respectively. Identification of each ORF was based primarily on TriTrypDB using tblastn, and each ORF was confirmed by employing getorf from EMBOSS and Newbler 2.6 when necessary. the monoxenic organisms revealed conserved housekeeping functions when compared to other trypanosomatids, especially compared with Leishmania major. However, major differences were found in ORFs corresponding to the cytoskeleton, the kinetoplast, and the paraflagellar structure. the monoxenic organisms also contain a large number of genes for cytosolic calpain-like and surface gp63 metalloproteases and a reduced number of compartmentalized cysteine proteases in comparison to other TriTryp organisms, reflecting adaptations to the presence of the symbiont. the assembled bacterial endosymbiont sequences exhibit a high A+T content with a total of 787 and 769 ORFs for the Angomonas deanei and Strigomonas culicis endosymbionts, respectively, and indicate that these organisms hold a common ancestor related to the Alcaligenaceae family. Importantly, both symbionts contain enzymes that complement essential host cell biosynthetic pathways, such as those for amino acid, lipid and purine/pyrimidine metabolism. These findings increase our understanding of the intricate symbiotic relationship between the bacterium and the trypanosomatid host and provide clues to better understand eukaryotic cell evolution.Fundação de Amparo à Pesquisa do Estado do Rio de Janeiro (FAPERJ)Fundação de Amparo à Pesquisa do Estado de São Paulo (FAPESP)Conselho Nacional de Desenvolvimento Científico e Tecnológico (CNPq)ERC AdG SISYPHEUniv Fed Rio de Janeiro, Inst Biofis Carlos Chagas Filho, Lab Ultraestrutura Celular Hertha Meyer, BR-21941 Rio de Janeiro, BrazilUniv Fed Rio de Janeiro, Inst Biofis Carlos Chagas Filho, Lab Metab Macromol Firmino Torres de Castro, BR-21941 Rio de Janeiro, BrazilLab Bioinformat, Lab Nacl Computacao Cient, Rio de Janeiro, BrazilINRIA Grenoble Rhone Alpes, BAMBOO Team, Villeurbanne, FranceUniv Lyon 1, CNRS, UMR5558, Lab Biometrie & Biol Evolut, F-69622 Villeurbanne, FranceUniv Estadual Campinas, Inst Biol, Dept Genet Evolucao & Bioagentes, São Paulo, BrazilUniv São Paulo, Fac Ciencias Farmaceut Ribeirao Preto, Dept Ciencias Farmaceut, São Paulo, BrazilLab Nacl Ciencia & Tecnol Bioetano, São Paulo, BrazilUniv Fed Minas Gerais, Inst Ciencias Biol, Dept Bioquim & Imunol, Belo Horizonte, MG, BrazilUniv Fed Goias, Inst Ciencias Biol, Mol Biol Lab, Goiania, Go, BrazilFundacao Oswaldo Cruz, Inst Carlos Chagas, Lab Biol Mol Tripanossomatideos, Curitiba, Parana, BrazilFundacao Oswaldo Cruz, Inst Carlos Chagas, Lab Genom Func, Curitiba, Parana, BrazilUniv Estadual Campinas, Ctr Pluridisciplinar Pesquisas Quim Biol & Agr, São Paulo, BrazilUniv Fed Minas Gerais, Inst Ciencias Biol, Dept Parasitol, Belo Horizonte, MG, BrazilUniv Fed Santa Catarina, Dept Microbiol Imunol & Parasitol, Ctr Ciencias Biol, Lab Protozool & Bioinformat, Florianopolis, SC, BrazilUniv Fed Vicosa, Dept Bioquim & Biol Mol, Ctr Ciencias Biol & Saude, Vicosa, MG, BrazilInst Butantan, Lab Especial Ciclo Celular, São Paulo, BrazilUniv São Paulo, Dept Biol, Fac Filosofia Ciencias & Letras Ribeirao Preto, São Paulo, BrazilUniversidade Federal de São Paulo, Escola Paulista Med, Dept Microbiol Imunol & Parasitol, São Paulo, BrazilUniversidade Federal de São Paulo, Escola Paulista Med, Dept Microbiol Imunol & Parasitol, São Paulo, BrazilWeb of Scienc
The Design of Parking Building 5 Stories with Steel Construction in Surakarta Based on SNI 1729-2015
Surakarta city is located on the southern island of Java, Central Java Province, Indonesia. Surakarta has many public facilities such as universities, malls, terminals, airport and etc. Need a lot of parking area to accommodate all the drivers to take their vehicle. This final project designed the parking building 5 stories with steel construction as the main solution to overcome the limitation of land area. The method used to analysis this parking building was Special Moment Frame (SMF). The SMF is the best system in the building’s concept because the beam structure has plastic hinge that can develop the seismic force. The best connection in the SMF is Reduction Beam Section (RBS), because the plastic hinge of the beam can occur in the planned point. The standard used for this parking building was Indonesia National Standard (SNI). Structural analysis calculation used ETABS software version 2015. The final result of the design has dimension of Rafter I300x150, Secondary beam I350x175, Non-SMF Beam I500x200, Non-SMF column H400x400, SMF beam I600x300, and SMF column H500x500. The base plate with dimension of 80 cm x 80 cm x 5 cm is satisfied. To support the load from column and transfer the load to soil used the pile foundation with 11 m in depth. The pile foundation diameter of 30 cm x 30 cm is satisfied to resist the load. Sloof dimension used 30 cm width and 50 cm height. The reinforcement used 7D16 for longitudinal and Ф10-200 mm for shear reinforcement
