Where Spirit and Bulldozer Roam: Environment and Anxiety in Highland Borneo

This paper explores changing perceptions of the natural environment among the Kelabit, an indigenous people of the Borneo interior. It considers both traditional and post-Christian conversion understandings about forest spaces. The former animistic ritual practices of the Kelabit centered on a spiritual dialogue with the natural world and this dialogue was often marked by active efforts to avoid or mitigate danger through ritual practice. One key example presented here is the former ceremony of \u27calling the eagle\u27 (nawar keniu), a ritual employed in times of crisis that exemplifies the dialogical and entwined relationship Kelabit had to the natural world. Such former animistic beliefs are contrasted with contemporary Christian practices, including a local mountain retreat on Mount Murud and present-day political and economic anxieties over logging in the Kelabit Highlands, as a means to consider relationships between religion and attitudes toward the environment among the Kelabit

Efficient Gene Targeting Mediated by Adeno-Associated Virus and DNA Double-Strand Breaks

Gene targeting is the in situ manipulation of the sequence of an endogenous gene by the introduction of homologous exogenous DNA. Presently, the rate of gene targeting is too low for it to be broadly used in mammalian somatic cell genetics or to cure genetic diseases. Recently, it has been demonstrated that infection with recombinant adeno-associated virus (rAAV) vectors can mediate gene targeting in somatic cells, but the mechanism is unclear. This paper explores the balance between random integration and gene targeting with rAAV. Both random integration and spontaneous gene targeting are dependent on the multiplicity of infection (MOI) of rAAV. It has previously been shown that the introduction of a DNA double-stranded break (DSB) in a target gene can stimulate gene targeting by several-thousand-fold in somatic cells. Creation of a DSB stimulates the frequency of rAAV-mediated gene targeting by over 100-fold, suggesting that the mechanism of rAAV-mediated gene targeting involves, at least in part, the repair of DSBs by homologous recombination. Absolute gene targeting frequencies reach 0.8% with a dual vector system in which one rAAV vector provides a gene targeting substrate and a second vector expresses the nuclease that creates a DSB in the target gene. The frequencies of gene targeting that we achieved with relatively low MOIs suggest that combining rAAV vectors with DSBs is a promising strategy to broaden the application of gene targeting

Terrorizing Advocacy and the First Amendment: Free Expression and the Fallacy of Mutual Exclusivity

Traditional free speech doctrine is inadequate to account for modern terrorist speech. Unprotected threats and substantially protected lawful advocacy are not mutually exclusive. This Article proposes recognizing a new hybrid category of speech called “terrorizing advocacy.” This is a type of traditionally protected public advocacy of unlawful conduct that simultaneously exhibits the unprotected pathologies of a true threat. This Article explains why this new category confounds existing First Amendment doctrine and details a proposed model for how the doctrine should be reshaped

A catalytic cycle for oxidation of tert-butyl methyl ether by a double C−H activation-group transfer process

A square-planar, iridium(I) carbene complex is shown to effect atom and group transfer from nitrous oxide and organic azides, releasing the corresponding formate or formimidate and an iridium(I)−dinitrogen adduct. The dinitrogen complex performs C−H activation upon photolysis or thermolysis, regenerating the carbene from tert-butyl methyl ether with loss of H_2. Taken together, these reactions represent a net catalytic cycle for C−H functionalization by double C−H activation to generate metal−carbon multiple bonds. Additionally, the unusual group transfer from diazo reagents underscores the unique nature of the reactivity observed for nucleophilic-at-metal carbene complexes
